Demonstration of monoclonal IgE by isoelectric focusing: first reported case of IgE myeloma in Australia.

A 56-year-old man who presented with back pain was diagnosed with multiple myeloma. Serum protein analysis by isoelectric focusing identified a monoclonal IgE lambda of unique spectrotype compared with other monoclonal immunoglobulins. The similarity of the spectrotype of this monoclonal IgE to that of two purified monoclonal IgEs suggests that these proteins have a characteristic spectrotype, sufficiently different from those of other immunoglobulin classes to allow its recognition.

Liquid-chromatographic determination of aminoglutethimide in plasma.

A simple, rapid "high-performance" liquid-chromatographic procedure is presented for the determination of aminoglutethimide in plasma. After precipitation of the protein with acetonitrile, an aliquot of the supernate is injected directly onto a radially compressed, reversed-phase column. The aminoglutethimide is isocratically eluted with a mobile phase of acetonitrile/water/tert-butyl ammonium phosphate. The method is both accurate and precise and has been in routine use in our laboratory for more than 12 months.

Chronic lymphatic leukemia developing in a patient with multiple myeloma: immunologic demonstration of a clonally distinct second malignancy.

A patient with multiple myeloma who subsequently developed chronic lymphocytic leukemia is reported. Initial studies demonstrated clinical and hematological features of multiple myeloma with an IgM lambda paraprotein. Skeletal disease was a significant presenting feature, although relapse occurred in extraosseous sites, particularly the pleura. He developed chronic lymphatic leukemia 31 months later and immunological studies showed the malignant lymphocytes to have kappa (Kappa) light chain surface immunoglobulin, demonstrating separate clonal origin of this patient's two B-cell malignancies.

Monoclonal antibodies to murine cell surface antigens. I. Lyt-1.1.

The production and properties of a monoclonal anti-Lyt-1.1 antibody are described. The monoclonal antibody is of the IgG2a subclass, is cytotoxic, and reacts with Staphylococcal protein A. The tissue culture supernatant has the same activity (titer on thymus > 1/1000) as an anti-Lyt-1.1 produced by a standard immunization procedure, and after in vivo passage of the hybridoma cell line, the serum titer was > 1:10(6). The monoclonal antibody has the same strain and tissue distribution as the conventional antibody, including reactions on Lyt-1 congenic strains, and it precipitates a cell surface molecule of m.w. approximately 67,000. In nonprimed mice, there were similar numbers of Thy-1+ and Lyt-1+ cells in spleen, lymph node, and thymus. By using the monoclonal reagent, T-helper and T-killer cells in an allogenic system were shown to be Lyt-1.1.

Multiple myeloma and monoclonal IgA with anti-actin reactivity.

Serum containing a monoclonal IgA protein from a patient with multiple myeloma gave intense immunofluorescent staining of smooth muscle fibres and the striations of skeletal muscle, cardiac muscle and thymic myoid cells. It also gave a weaker reaction with hepatocytes in a 'polygonal' pattern, and with renal glomeruli in a diffuse pattern. In culture fibroblasts, the serum stained long, parallel cytoplasmic filaments. Specificity of the staining reactions for actin was established by their prevention on serum absorption with skeletal muscle actin, but not by skeletal muscle myosin, tropomyosin or troponin, and by the demonstration that eluates obtained by acid dissociation of the serum-actin precipitates gave the same staining reactions as the original serum. Localization of the anti-actin reactivity to the monoclonal IgA protein was established by the observation that the same staining reactions were obtained with a monospecific IgA conjugate, with the eluate derived from the gamma globulin band of a serumel ectrophoretic strip, and with a purified euglobulin (IgA) fraction; also, the eluates obtained by acid dissociation of the serum actin precipitates contained monoclonal IgA.
