Opsin-immunoreactive outer segments in the pineal and parapineal organs of the lamprey (Lampetra fluviatilis), the eel (Anguilla anguilla), and the rainbow trout (Salmo gairdneri).

The pineal complex of Lampetra fluviatilis, Anguilla anguilla and Salmo gairdneri was studied by means of the indirect immunohistochemical antiopsin reaction. Opsin-immunoreactive material was demonstrated in the outer segments of the photoreceptor cells in the pineal organ of all three species investigated. In the lamprey, the opsin-positive outer segments were located in the lumen of the pineal vesicle and atrium. In the two teleost species, the immunoreactive outer segments were observed in abundance in the pineal end-vesicle and stalk. These structures were found to accumulate in the prominent initial portion of the pineal stalk of the eel. In the rainbow trout, immunoreactive outer segments occurred in the wide orifice of the pineal recess at the roof of the third ventricle. In addition, outer segments of photoreceptor cells of the parapineal organ ("parapinealocytes") displayed opsin immunoreactivity. In the lamprey, opsin immunoreactivity was restricted to the central portion of the ventral parapineal retina, while the parapinealocytes in the lateral portions did not bind the antibody. In the two teleosts, immunoreactive outer segments displayed a scattered pattern. These immunocytochemical results provide direct evidence that the photosensitivity of the pineal demonstrated electrophysiologically in lampreys and teleosts (cf. Dodt 1973) is based on an opsin-containing photopigment. The presence of opsin in cells of the parapineal organ strengthens the view that also this organ may be capable of direct light perception. In the lamprey, the exclusive opsin immunoreactivity of a circumscribed group of parapineal cells suggests the existence of two types of parapinealocytes. The significance of opsin-containing photoreceptor outer segments occurring in the most proximal portion of the teleost pineal stalk is discussed, especially with regard to the interpretation of results obtained from pinealectomy experiments.

Human papilloma virus 5-DNA in a carcinoma of an epidermodysplasia verruciformis patient infected with various human papillomavirus types.

An epidermodysplasia verruciformis patient suffering from generalized warts and a carcinoma at the forehead was found to be infected by at least six types and subtypes of human papillomaviruses. The central part of the carcinoma, however, harbored only human papillomavirus 5 DNA. The DNA persisted extrachromosomally in high genome copy number. In contrast to wart DNA preparations, a significant part of the viral sequences in the carcinoma was present as oligomers, at least part of them being concatemers as shown by S1 digest. The human papillomavirus 5 subtype from this carcinoma was compared with the two other carcinoma isolates described so far and proved to be rather similar.

[Peroral kallikrein therapy for male infertility--comparison between smokers and non-smokers].

101 male patients with idiopathic Oligo-, Astheno- and Oligoasthenozoospermia were given Kallikrein orally (3 X 200 KE daily) for 100 days. Sperm controls were performed before, during and after treatment. The evaluation of the results of the patient group showed, that sperm density, total sperm count, qualitative and quantitative motility during and after therapy had improved significantly. The patients were divided into 4 groups: smoker, non-smoker, group I and group II. Patients in group I had a sperm density equal to or less than 20 X 10(6) sperm/ml; those in group II had a sperm density greater than 20 X 10(6) sperm/ml. This calculation was determined by the average of two sperm controls before treatment. A significant improvement was practically seen only in the groups of smokers. The possible reasons are discussed.

[Prostato-urethritis due to chlamydia].

One of the etiological agents of urethritis and prostatitis is Chlamydia trachomatis which reveals obligatory intracellular parasitism. Patient specimens (smears of the urethra, urine, first portion, expressed prostatic fluid, ejaculate) are centrifuged onto special cell cultures, so-called McCoy cells, and induce characteristic cytoplasmatic inclusions. Giemsa staining or DNA-binding fluorochromes (DAPI or Hoechst 33258) are suitable procedures to identify chlamydia inclusions. Serologic tests to establish infections due to Chlamydia trachomatis are only of limited diagnostic value.

Characterization of a human papillomavirus from epidermodysplasia verruciformis lesions of a patient from Upper-volta.

A case of epidermodysplasia verruciformis in a patient from Upper-Volta is described. Slightly elevated, flat warts were observed on hands, feet, arms and legs, and pityriasis versicolor-like lesions were found mainly on the trunk. The patient showed no malignant tumors. Histological examination revealed hyperkeratosis, granulosis and moderate acanthosis with large, foamy, basophilic keratinocytes in stratum granulosum and stratum spinosum. Papillomavirus particles could be prepared from these lesions and were differentiated from known papillomavirus types by immune electron microscopy with monospecific antisera and by DNA-DNA hybridization. The viral DNA was characterized by cleavage with several restriction endonucleases and a physical map of the resulting fragments was established. The virus is designated as HPV 8. Preliminary seroepidemiologic studies with human sera indicate a rather wide distribution of HPV 8. Blot hybridization of DNA from human carcinomas with 32P-labelled virus DNA detected no HPV 8-specific sequences.

Oxytocin- and vasopressin-immunoreactive nerve fibers in the pineal gland of the hedgehog, Erinaceus europaeus L.

Oxytocin- and vasopressin-immunoreactive nerve fibers, apparently originating from a dorsal subunit of the paraventricular nucleus, were demonstrated in the pineal gland of the hedgehog. The majority of these fibers (pinealopetal projections) is intimately related to the capillaries of the pineal organ, whereas only a few elements are scattered throughout the pineal parenchyma. The number of peptidergic elements observed in the central portion of the pineal organ exceeds that of fibers located at the periphery. In relation to the functional state of the animals, the amount of immunoreactive material in these pinealopetal nerve fibers exhibits conspicuous variations. In hibernating hedgehogs (group 1), these nerve fibers were considerably richer in oxytocin than in non-hibernating or arousing winter animals (group 2 and 3). In contrast, only weak immunoreactivity for vasopressin was found in intrapineal nerve fibers of hibernating hedgehogs (group 1), whereas the fibers of arousing or non-hibernating hedgehogs (group 2 and 3) contained slightly larger amounts of vasopressin. In the pineal organ of animals sacrificed during the summer period (group 4), no immunoreactivity for both neuropeptides was found. The functional significance of the connection between the hypothalamic paraventricular nucleus and the pineal organ is discussed with special reference to the vascular terminals of the pinealopetal peptidergic nerve fibers.

[Adenoma of the nipples].

The adenoma of the nipple (syn.: erosive adenomatosis, papillary adenoma, florid papillomatosis) is a benign tumour of the major nipple ducts and presents chiefly as an eroded oozing and crusted lesion with serous or sanguineous discharge of the nipple or as an indurated nodule. The lesion can be mistaken clinically for Paget's disease of the nipple. The correct diagnosis, however, can easily be established by an incisional biopsy. Pathohistologically, the tumour displays adenomatous and papillomatous proliferation of the milk sinus epithelium. An outer myoepithelial and inner columnar epithelium layer can mostly be distinguished. Atypical cellular features are lacking. Keratin cysts near the mostly eroded epidermal surface are are additional characteristic signs. The resection of the nipple is the therapy of choice. The adenoma of the nipple is no precancerous condition. A typical case of adenoma of the nipple from a 54 year old woman is presented.

The ultrastructure of mycobacterium marinum granuloma in man.

3 biopsies of 3-5 week-old nodular lesions in 2 patients with so-called swimming-pool (aquarium-) granuloma have been examined by electron microscopy. The cytohistological spectrum simultaneously comprises acute exudative as well as chronic proliferative phenomena. Epithelioid cells and collagen producing fibroblasts are already conspicious in 3 week-old granuloma. According to the cytological composition the Mycobacterium marinum granuloma represents a high-turnover granuloma with immunogenic origin. It is comparable to mycobacterial diseases in the presence of well developed cell mediated immunity (Lupus vulgaris, tuberculoid leprosy). Degrading mycobacteria have been rarely detected in phagocytes and are compared with viable bacilli in macrophages of experimentally infected mice. Curved and annular parallel membranes ("worm-like structures") in the cytoplasm of transformed macrophages and in fibroblasts presumably originate from the membranes of endoplasmic reticulum. Cord-like structures with transverse bands (periodicity 170-180 A) in the lumen of RER of some fibroblasts are interpreted as intracellularly aggregated collagen precursors.

Actinic elastosis in black skin. A light- and electronmicroscopic study.

In 9 otherwise dermatologically normal South African Balck volunteers (1--72 years old), sunlight-exposed and non-exposed skin has been examined by light- and electronmicroscopy with special references to age-dependent and actinic alteration of dermal structures. Two 72 year old Blacks exhibited typical dermal elastosis: in 1 case to a marked extent and already detectable by lightmicroscopy, in the other case only to a mild degree. In contrast, only age-related elastic fibers were revealed in the lower dermis of sunlight-exposed skin and in all dermal layers of unexposed skin. These observations are contradictory to the general view that actinic elastosis does not occur in Blacks. The elastotic material is obviously a de-novo synthesized and secreted pathological product of chronically UV-altered fibroblasts.

So-called cellulite: an invented disease.

The anatomic basis of so-called cellulite, the hormonal basis for the clinical condition, the prevalence of it, the essential normality and inevitability of it in women, the supervention of it in hormonally feminized men, and the near futility of treating the non-disease are explored in this paper.
