Isolation and partial characterization of rat lymphoid cell surface histocompatibility antigens and immunoglobulins.

Cell surface molecules of rat normal lymphoid cells were selectively labelled by lactoperoxidase catalysed iodination or by a galactose oxidase tritiated sodium borohydride technique, subsequently detergent solubilized, isolated by indirect immunoprecipitation and analysed on SDS-polyacrylamide gel electrophoresis. Four polypeptide chains were isolated by using the alloantiserum DA anti-Lewis. The molecular weights of the antigens were calculated as 41,000, 33,000, 27,000 and 12,000. Based on functional in vitro characteristics of the antiserum used and on the physiochemical properties as well as genetics of inheritance and tissue distribution, the polypeptide chains were identified as being subunits of Ag-B and Ia antigens. Two types of immunoglobulin heavy chains exhibiting the molecular weight 70,000 and 64,000 were isolated from unfractionated normal spleen cells by use of a polyvalent rabbit anti-rat immunoglobulin serum and tentatively identified as mu and delta chain. Using the same anti-immunoglobulin serum, no molecules could be precipitated from the lysated of Lewis thymocytes or peripheral T cells.

Thyroid carcinoma: presentation of a clinical material with special aspects on the classification and operative treatment.

During the years 1969-1975, 58 patients were treated for thyroid carcinoma. 48 patients had differentiated and 10 had anaplastic carcinomas. The material is presented with special attention given to diagnostic methods and treatment for the differentiated forms. The value of cytological examination of fine needle biopsy is clearly documented. 40 patients with differentiated carcinomas were treated with total thyroidectomy, with low complication rate. Accidental persistent recurrent nerve paralysis as well as persistent hypoparathyroidism occurred in less than 3%, Subdivision of papillary and follicular carcinomas on the basis of their local growth pattern is emphasized as being of value in the determination of subsequent therapy.

Propulsion and mixing of small bowel contents during exogenous gastrin infusion. An experimental study in rat.

The effect of intravenous infusion of Synthetic Human Gastrin I (SHG) on the propulsion and mixing of small bowel contents was studied quantitatively in conscious rats. Radioactive test substances were infused continuously through a permanent duodenal catheter for totally five hours. Two differently labelled, but otherwise identical, test substances were used and one of them (125I-PVP) was infused during the first three hours, the other (131I-PVP) during the remaining two hours. The activity distribution of each isotope along the excised bowel specimen was measured immediately after the end of the infusion. The bowel length passed by the transition point between the nuclides and the region of overlap in the border zone between them were taken as measures of propulsion and mixing, respectively. A significant increase of the serum gastrin level during infusion of SHG was verified by radioimmunoassay. The total small bowel transit time was significantly prolonged during gastrin infusion secondary to a retarded propagation velocity in the distal small bowel. The slower transport rate during gastrin infusion was coupled to a significant increase of the mixing of small bowel contents. These changes in small bowel motility were not apparently due to a concomitant increase of gastrict acid secretion, since pretreatment with a potent H2-receptor blocker, cimetidine, did not abolish them.

Propulsion and mixing of small bowel contents in rat during varied levels of endogenous gastrin.

The propulsion and mixing of small bowel contents were studied in conscious rats 14 days after antral resection, antral exclusion and gastroentero-anastomosis in order to evaluate the influence of hypo-, hyper- and normogastrinemia, respectively, on the net result of small bowel muscular activities. The quantitative measures of propulsion and mixing did not differ significantly between the groups. There was, however, a significant correlation between high serum gastrin and increased intestinal mixing after antral exclusion and gastroentero-anastomosis. This finding supports earlier studies on gastrin effects on the small bowel obtained by exogenous gastrin infusion.

Gastric emptying in duodenal ulcer patients before and after truncal vagotomy with pyloroplasty and parietal cell vagotomy.

The gastric emptying of a liquid test meal containing 99Tcm-sulphur colloid was studied with a gamma camera in 24 duodenal ulcer patients. Studies were made before surgery, randomly chosen between truncal vagotomy with pyloroplasty and parietal cell vagotomy, and six weeks and one year after. Large variations in the emptying patterns were observed both between patients and in the same patients. No consistent change in the distribution of patterns were found between the three observation times and between the two surgical procedures. An initial rapid emptying was found in several studies and was observed both before and after operation and did not seem to be related to type of surgical procedure or to individual patients. The method applied did not allow a detailed study of this initial phase, which appears to be of a higher significance for the over-all emptying characteristics than the late exponential emptying, on which earlier investigators have been focused.

Effects of vasoactive intestinal peptide and pancreatic polypeptide on small bowel propulsion in the rat.

Vasoactive intestinal peptide (VIP) and the newly discovered pancreatic polypeptide (PP), both having been proposed as pathogenetic factors in the watery diarrhea syndrome (WDRA), were intravenously infused into rats in order to study the effect on small bowel propulsion. Simultaneously with the infusion of hormone, radioactive test substance was continuously infused into the duodenum of the conscious animal through a permanent catheter. By recording the distribution of radioactivity along the excised gastrointestinal tract, analyses of small bowel propulsion were possible. After infusion of VIP small bowel transit time was significantly prolonged, and the local propagation velocity was retarded both in the proximal and the distal part of the small bowel. PP did not alter small bowel populations. The heavily retarded transport rate after VIP is a propulsive dysfunction and not inconsistent with the clinical sign of diarrhea.

Heterotopic bone formation in upper midline abdominal incisions.

Five cases of heterotopic bone formation in upper abdominal incisions within one year are presented. The patients were male, with a mean age of 52 years. The bone formation was diagnosed by palpation and X-ray and in 3 cases verified by operation and histology. The condition is said to be very rare.

Simultaneous measurement of the propulsion and mixing of small bowel contents in the rat.

A method is presented for the simultaneous measurement of propulsion and mixing of small bowel contents in conscious rats. Through a permanent duodenal catheter two differently labeled test substances (125I-PVP and 131I-PVP) were infused continuously one after the other. The radioactive label was changed once during the total infusion period of 5 hours. Studies were performed in six groups of animals which differed with respect to time for interchange of test substance. At the end of the total infusion period the border zone had travelled to six different locations in the small bowel. The animals were then killed and the two radioactivities were recorded from the excised bowel specimen. The resulting activity densities along the small bowel were peaky and the overlap between the two labels in the border zone was minute, indicating that the intestinal contents were transported in isolated portions with only minor exchange of luminal contents between adjacent regions. The transport apparently proceeded steadily in a mainly aboral direction with no indications of abrupt changes in transport velocity.

Acetylsalicyclic acid and gastrointestinal propulsion in the rat.

The possible influence of moderate amounts of acetylsalicyclic acid (ASA) on gastric emptying, duodeno-gastric reflux and small bowel propulsion was studied in rats with permanent gastric and duodenal tubes. The ASA-containing or the control solution was introduced intra-gastrically half an hour before the simultaneous administration of differently labeled radioactive test meals into the stomach and the duodenum. ASA was given as 7.5 or 15.0 mM solution in 100 mM hydrochloric acid or in 100 mM sodium chloride. After 15 minutes the gastrointestinal propulsion was examined. No effect of the ASA treatment was noted. No increase in duodeno-gastric reflux was found in the ASA-treated animals.

Non-occlusive enteric gangrene associated with severe upper gastrointestinal bleeding.

Six cases of non-occlusive enteric gangrene in association with severe upper gastro-intestinal haemorrhage are described, the incidence of which corresponds to approximately one per cent of the patients with serious gastric bleedings. A brief discussion of the pathophysiology of ischaemic gut lesions is given. In order to reduce the incidence of this disease rapid and adequate treatment of all massive gastro-intestinal bleedings is emphasized. Patients with unexplained abdominal pain in association with haemorrhagic shock should especially be regarded as being at risk of developing the enteric gangrene.

The adreno-medullary effect on gastrointestinal propulsion after laparotomy. An experimental study in the rat.

Gastrointestinal propulsion after abdominal surgery is impaired during the postoperative phase. Adrenalectomy as well as only adrenal demedullation prevented the impairment of gastric emptying and small bowel propulsion after laparotomy. In non-laparotomized rats gastric evacuation was significantly increased after adrenalectomy and adrenal demedullation. The results indicate that adrenomedullary activity retards gastrointestinal propulsion not only postoperatively but even in intact rats.

Influence of ether anaesthesia and oro-gastric intubation on gastric evacuation and small bowel propulsion after laparotomy. A methodological study in the rat.

A model for the simultaneous study of gastric evacuation and small bowel propulsion in the rat is presented. By means of a modification, with both indwelling duodenal and gastric tubes, studies on fully conscious animals were possible. Gastric evacuation and small bowel propulsion were analysed with respect to the effects of ether anesthesia and gastric intubation. The gastric intubation in ether anaesthesia greatly retarded gastric evacuation but left small bowel propulsion unaffected one day after laparotomy. The main factor retarding gastric evacuation appeared to be the oro-gastric intubation procedure. Ether anaesthesia neither affected gastric evacuation nor small bowel propulsion after laparotomy when the test meal was deposited into the stomach and into the duodenum, respectively, via the permanent tubes.
