Comparative effects of T-2 toxin and diacetoxyscirpenol on drug metabolizing enzymes in rat tissues.

The effects of T-2 toxin and diacetoxyscirpenol on tissue drug-metabolizing enzymes in young male rats were compared. Mycotoxicoses were produced by daily oral administration of toxins at 1.0 mg/kg body weight for 1, 4 or 8 days. Many hepatic, renal and pulmonary oxidative and conjugative enzymes were measured in animals killed 24 hr following the last administration. The effects of the two trichothecene mycotoxins were generally similar. In liver the decrease in microsomal and cytosolic proteins paralleled the decline in total plasma proteins or the increase in plasma GOT activity. Hepatic microsomal cytochrome P-450 decreased in rats receiving trichothecenes for 8 days. This effect was more marked when aminopyrine, benzphetamine, ethylmorphine and ethoxycoumarin dealkylations or aniline and benzopyrene hydroxylations were measured. p-nitrophenol glucuronyltransferase activity was enhanced in animals receiving at least one administration of trichothecenes, whereas there was no change in conjugation to glutathione or acetate. In other tissues, there was no change in any renal enzymes whereas a significant rise in pulmonary monooxygenase was observed in T-2 toxin administered to rats for 4 or 8 days.

Retrospective analysis of clinical data of normotensive and hypertensive pregnant women and their newborns.

The medical history of 256 hypertensive and 263 normotensive pregnant women was analysed retrospectively. There was a negative correlation (P less than 0.01) between the maximal pretreatment diastolic blood pressure and the birth weight of newborns in the hypertensive group. The prevalence of a hypertensive family history, pyelonephritis, proteinuria, delivery by Caesarean section, fetal asphyxia during delivery and death of the newborn during delivery was significantly higher in the hypertensive group than in the normotensive one. The gestational age at delivery was shorter and the birth weight of the newborn was lower in the hypertensive women than in the normotensive women.

Candida guilliermondii var. guilliermondii infection in infertile women.

The sera of 263 women--217 infertile and 46 pregnant--were examined by various serological methods (precipitation test, agglutination, indirect immunofluorescence) to detect Candida guilliermondii var. guilliermondii (C.g.) infection. The precipitation reaction was performed with extracellular C. guilliermondii antigen, the agglutination reaction was employed parallel with C. albicans. In the infertile group 122 (56.2%) proved to be C.g. positive, while in the fertile 11 women (23.9%) proved to be so, the level of significance being p less than 0.0001 between the two groups. A one-month ketoconazole treatment (one tablet, 200 mg/day) was adequate for eliminating the C.g. infection. In a few cases hystological examinations were also performed according to Gomori-Grocott and yeast cells could be detected in the stroma of the ovary. IgA, IgG, IgM, Gc-globulin, transferrin and ferritin determinations were carried out before and after the ketoconazole treatment, and there were significant differences in the IgM and transferrin levels between the infected and non-infected groups. The authors achieved 5 pregnancies of 56 treated women in 6 months.

The effect of terbutaline upon myosin preparations of the human uterus.

It has been perceived that the phosphate (P) content of the human uterus myosin was considerably lower (half or less) in patients treated with terbutaline before delivery than that of untreated patients. Therefore the in vitro effect of terbutaline was investigated upon uterine myosin. The results showed that the presence of terbutaline in the incubation medium without ATP decreased the performed (endogenous) P content of myosin slightly, whereas in the presence of ATP terbutaline facilitated release of the phosphoryl groups from myosin parallel with the increase of terbutaline concentration. Although at low concentrations of terbutaline (in the micromolar range) a few phosphoryl groups are still incorporated in myosin, the time reaching the maximum of P incorporation was prolonged.

Preparation and P content of non-pregnant and pregnant human uterine myosin.

Myosin content and phosphorus (P) concentration of myosin preparations were measured in non-pregnant and pregnant human myometrial tissue specimens. It was found that the amount of myosin gained from 1 g of minced myometrial tissue is 0.5 mg in the early follicular phase of the menstrual cycle, 0.6-0.7 mg in the late luteal phase, and 6-7 mg during pregnancy. Considering the different functional stages of the myosin sources and the performance characteristics of the methods, the estimated myosin content of non-pregnant myometrium is 1.0-1.5 mg, while 10-15 mg in pregnant myometrial tissue. A considerable amount of P is bound to the preparations. It is the smallest in the post-menstrual period and increases towards the end of the cycle. The largest amount of P is gained from fresh pregnant uterine samples. Analysis of the alkaline hydrolysate showed that the phosphate group was bound to amino acids, in the largest amount to arginine, less to histidine and the smallest amount to lysine and serine. As a function of the duration of storage, especially the P-Arg concentration was decreasing. The prolonged hydrolysis time decreases again the concentration of P-Arg with a consecutive increase of No. 1 and 2 P-containing peaks in the chromatographic profile of alkaline hydrolysate.

[Effect of short duration euphylline infusion on the uteroplacental blood pool].

In 19 late pregnant women with suspected placental insufficiency uteroplacental perfusion was measured and then the effect of a shortlasting infusion of aminophyllin on uteroplacental blood pool was examined. The results show evidence that in normal perfusion typ uteroplacental blood flow is not influenced by aminophyllin. But in pathologic perfusion typs there are two groups after aminophyllin. In the first one there is an increase of activity measured over the placenta (positive test). In these cases the cause of the lowered perfusion may be a functional one, treatment will be successful. In the second group there was no change or a decreasing activity following administration of aminophyllin (negative test). In these cases the reduced perfusion may be caused by morphological factors. Treatment will be without success with a high likelihood. The aminophyllintest following the isotope perfusion measurement is useful both for prognosis of newborn weight and appropriateness of therapy.

The clinical importance of bacterial urinary tract infections during pregnancy. I. Premature delivery, dismaturity.

Using computer analysis the authors studied the relationship between the various clinical forms of urinary tract infections and premature deliveries, i.e. dysmaturity in a population including several thousands of individuals. In the frameworks of uniform and monitored perinatal obstetric and neonatologic care system--the uniform diagnostics of bacteriuria was established. The standard birth-weights for the regions involved by our care were calculated, too. The basic observations of Kass were still found to be valid. In case of urinary tract infections, an increased rate of premature deliveries with respect to both birth-weights and gestational age of pregnancy should be reckoned with. Computer analysis proved not only a tendency for premature deliveries but also retarded intrauterine growth of the foetus.

[Changes in estriol excretion in the urine and kidney function during Partusisten infusion].

In a random controlled trial the effects of a betamimetic infusion (Fenoterol 2 micrograms/min) on the estriol excretion and the renal function were analysed in 30 patients in the 28.-34. weeks of pregnancy. It was stated that the estriol excretion in urine during the Partusisten infusion significant decreases (from 663 micrograms/2 h to 376 micrograms/2 h, i.e. to 56%). The diuresis and the creatinine-clearance showed a significant diminishing too (from 218 ml/2 h to 154 ml/2 h and from 131 ml/min to 88 ml/min). The decrease of estriol excretion is caused on the one hand by the diminution of GFR, on the other by a direct effect of betamimetics on the kidney. From the results obtained in this study the following consequences can be drawn: 1. The determination of urinary estriol during tocolytic treatment is not a suitable method for the monitoring the fetoplacental unit. 2. Applying the tocolysis in an infusion a possible reduction of water intake and a strict control of water balance is of great importance. 3. The betamimetic therapy in patients having an impaired renal function can be applied only with great precaution.

[Sodium, potassium and acid-base values of mother and newborn, following delivery by caesarean section under sombrevin (propanidid) anaesthesia (author's transl)].

Acid-base and electrolyte values of 24 newborns delivered by caesarean section are reported in this paper. Anaesthesia had been introduced by sombrevin-succinylcholine. No metabolic acidosis, which might have been attributable to sombrevin action, was recordable from any of the newborns. Acid-base values were normal, and somewhat increased potassium values returned to normal, soon.
