[Characterization and simultaneous assay of polymers and additives composing multilayer plastic material for pharmaceutical use by FTIR].

Multilayer plastic materials provide useful solutions for primary pharmaceutical formulae (maintaining physical properties, compatibility with contents, protection against exterior agents). Fourier transform infrared spectometry is an ideal method to identify and quantify the different polymers and additives. Each functional group has a characteristic band which can be selected and rapidly identified for routine quality control. The proportional quantities of each polymers in the multilayer system can be determined directly using the characteristic bands for each functional group. The technique was applied to the analysis of heat-formed platelets made of polyvinyl chloride and polyvinylidene chloride.

Liquid chromatographic analysis of monosaccharides with phenylisocyanate derivatization.

Because of the lack of sensitivity in carbohydrate analysis, HPLC pre-column derivatization techniques which give strongly UV absorbing compounds have been reported. These techniques have the disadvantage of leading to several chromatographic peaks from each reducing sugar. To enhance both the sensitivity and the selectivity of such specific separation problems, a simplex procedure was applied to optimize the phenylisocyanate derivatization of monosaccharides: a major and stable derivative was formed under the optimal conditions established. The method was extended to deoxysugars and methylglycosides. The limit of detection was 0.2-1 ng for all sugars tested.

[Contribution of capillary electrophoresis in the analysis of molecules with pharmaceutical and biotechnological value].

Capillary electrophoresis entails a variety of separation modes which are: capillary zone electrophoresis, capillary gel electrophoresis, micellar electrokinetic capillary chromatography, isoelectric focusing and isotachophoresis. Many applications are already reported. The fundamentals of the various modes are discussed along and typical separations in the pharmaceutical and biotechnologic fields are presented. The advantages and disadvantages in comparison to other analytical techniques are also discussed.

Simultaneous determination of gluconolactone, galactonolactone and galactitol in urine by reversed-phase liquid chromatography: application to galactosemia.

A reversed-phase liquid chromatographic assay was developed for the specific evaluation of metabolic by-products in the urine of galactosemic patients and based on the simultaneous determination of gluconolactone, galactonolactone and galactitol. The procedure involved a lyophilization step and the formation of phenylisocyanate derivatives, followed by injection directly into the chromatograph. Analytical results showed good selectivity, linearity, precision and accuracy. The method enabled the detection of levels as low as 0.05-0.1 ng, and compared favourably with other published techniques for the estimation of aldonic acids in biological fluids.

[Quality control of cod liver oil, physicochemical methods of analysis].

The control of oils and fats requires analytical processes far surpassing the traditional framework of physical properties and chemical characteristics. The control of cod liver oil is improved by the use of these procedures. The present report is devoted to the measurement of vitamin A by ultraviolet spectrophotometry and of vitamin D3 by high performance liquid chromatography, to the determination of the profile of their fatty acids and polyunsaturated fats, as well as to their measurement by liquid gas chromatography after derivatisation. The results permit the definition of the cod liver oil's quality, the differentiation between fish oils and the detection of possible changes. A technique for verifying the absence of butylhydroxyanisol and of butylhydroxytoluen is also described.

[Determinants of left ventricular hypertrophy in hypertensive subjects that have never been treated: role of the sodium intake].

Many factors have been implicated in the pathogenesis of myocardial hypertrophy, and the role of sodium has recently been suggested. In the present study, we assessed the influence of dietary sodium on the degree of left ventricular hypertrophy (LVH) in 41 patients aged 38 +/- 10 (mean +/- SD) with mild essential hypertension (casual blood pressure 149 +/- 17/91 +/- 11 mmHg). Patients had never been given antihypertensive drugs before and ingested ad libitum sodium intake. Posterior wall thickness (PWT) and left ventricular mass (LVM) were measured by M-mode echocardiography and sodium intake was estimated from urinary sodium excretion rate (UNa, mmol/24h). Both PWT and LVM, and not telediastolic diameter or LV fractional shortening, were directly correlated with UNa (r = 0.47 and 0.46; p less than 0.02 and 0.002, respectively. A stepwise multiple regression analysis confirmed that UNa was a determinant of LVM independently of sex, age, body weight, blood pressure and duration of hypertension. No correlation was found between LVM and plasma renin activity, whilst a positive one existed between PWT and hematocrit (r = 0.42; p less than 0.007). These results suggest that dietary sodium may play a role in modulating left ventricular mass in untreated hypertensives, possibly in expanding volume or activating the adrenergic system.

[Rapid analysis of plastic materials for pharmaceutic use].

The use of plastic materials as containers for pharmaceuticals implies the knowledge of previous data and the constancy of their composition. These data and migration studies are acquired in the development phase and are the ground line in routine control. The molecular size distribution and the identification of principal additives are determined by gel permeation chromatography. Infra red spectrometry is applied to the determination of relative proportions of monomers in copolymers; HPLC and GLG head space are suitable for the identification of additives and for the studies of stability.

Nuclear magnetic resonance spectrometry and liquid chromatography of two bile acid epimers: ursodeoxycholic and chenodeoxycholic acid.

A specific reverse-phase high-performance liquid chromatographic (HPLC) technique is described for the analysis of bile acids and their conjugates in human serum. Precise quantitation is obtained using UV detection. 13C-NMR spectrometry suggests a structural explanation for the different HPLC retention times of chenodeoxycholic acid, its epimer (ursodeoxycholic acid), and their methyl esters.
