Hyperdense intracranial epidermoid cysts: a study of 15 cases.

BACKGROUND: Although intracranial epidermoid cysts generally appear as hypodense lesions on CT scans, occasionally, they may appear hyperdense, making diagnosis difficult. The aim was to study clinical features and possible pathogenesis of hyperdense intracranial epidermoid cysts (HIECs). METHODS: Fifteen patients with HIECs were surgically treated in our department during a 21-year period (1985 up to 2005). We reviewed each patient's clinical records, radiological findings, operative reports, and pathological examinations, along with a review of the literature. FINDINGS: The incidence of HIECs in our series was approximately 3.02%, with female predominance. The average age was 32 years and the average duration of complaints was nine months. The most common location was the parasellar area (6 cases), followed by cerebellum (4 cases), clival region (3 cases), cerebello-pontine angle (1 case) and lateral ventricle (1 case). Gross-total excision of the tumor was achieved in ten patients (66.7%). Four (26.7%) patients underwent subtotal resection; and one (6.6%) patient underwent partial resection. Aseptic meningitis that occurred postoperatively in two cases was controlled by lumbar drainage. CONCLUSIONS: Recurrent leakage of the irritating cyst contents and subsequent chemical inflammatory response may be responsible for the high-density on CT scans and the cystic nature. A correct histological diagnosis is important because, in contrast to typical ECs and other lesions, HIECs are more prone to spread intra-operatively and result in severe aseptic meningitis.

Remanent zero field spin splitting of self-assembled quantum dots in a paramagnetic host.

We report on the observation of a finite spin splitting at zero magnetic field in resonant tunneling experiments on CdSe self-assembled quantum dots in a (Zn,Be,Mn)Se barrier. This is remarkable since bulk II-VI dilute magnetic semiconductors are paramagnets. Our experiment may be viewed as tunneling through a single magnetic polaron, where the carriers contained inside the dot act to mediate an effective ferromagnetic interaction between Mn ions in their vicinity. The effect is observable up to relatively high temperatures, which we tentatively ascribe to a feedback mechanism with the electrical current, previously predicted theoretically.

Direct evidence of oxidized gold on supported gold catalysts.

Supported gold catalysts have drawn worldwide interest due to the novel properties and potential applications in industries. However, the origin of the catalytic activity in gold nanoparticles is still not well understood. In this study, time-of-flight secondary ion mass spectroscopy (TOF-SIMS) has been applied to investigate the nature of gold in Au (1.3 wt %)/gamma-Al2O3 and Au (2.8 wt %)/TiO2 catalysts prepared by the deposition-precipitation method. The SIMS spectrum of the supported gold catalysts presented AuO-, AuO2-, and AuOH- ion clusters. These measurements show direct evidence for oxidized gold on supported gold catalysts and may be helpful to gaining better understanding of the origin of the catalytic activity.

Probiotic treatment using Bifidobacterium lactis HN019 reduces weanling diarrhea associated with rotavirus and Escherichia coli infection in a piglet model.

BACKGROUND: Diarrhea associated with rotavirus and Escherichia coli is one of the major gastrointestinal problems faced by human infants. Using a piglet model, the authors investigated the protective effectiveness of probiotic feeding against naturally acquired diarrhea in weanlings. METHODS: Seventeen piglets were allocated into two groups balanced for live weight and litter of origin. The test group was administered Bifidobacterium lactis HN019 (109 colony-forming units (cfu)/piglet/d) orally until the end of the experiment; the control group did not receive probiotic treatment. After 1 week, animals were penned individually and weaned onto a diet for a weaner. The degree of subsequent diarrheal disease was monitored in both groups of animals, feed intake and live weight gain of the piglets were assessed, and blood and fecal samples were collected to measure concurrent systemic and gastrointestinal tract immunologic activity. RESULTS: Compared with the controls, piglets that received B. lactis HN019 had a lower severity of weanling diarrhea and maintained a greater feed conversion efficiency during weaning. The protective effect of probiotic feeding was associated with lower concentrations of fecal rotavirus and E. coli, higher blood leukocyte phagocytic and T-lymphocyte proliferative responses, and higher gastrointestinal tract pathogen-specific antibody titers. CONCLUSIONS: These results show that dietary treatment using B. lactis HN019 can reduce the severity of weanling diarrhea associated with rotavirus and E. coli, possibly via a mechanism of enhanced immune-mediated protection. This study suggests that probiotic treatment may be an effective dietary means of preventing or limiting diarrhea in human infants.

[Treatment of dilute solution anilines-containing with displacement by extraction].

It was found that the biodegradability (indicated by BOD5/CODCr value of water solution containing anilire or chloroaniline declined with the increasing of their concentration. In order to bio-treat such water, decrease of the solute's concentration was necessary. Based on the research of organic solvents' and complexing agent's biodegradability, a series of extraction experiments were conducted for aniline and m-chloroaniline with complexing agent or physical solvent. The factors influencing the distribution ratio were discussed. The BOD5/CODCr value of raffinates showed that the raffinate can be treated by biological degradation without dilution if suitable solvent was choosed. It was proved that displacement by extraction had great potentiality for treating wastewater containing non-biodegradable organic contaminant.

[A comparative clinical study of carboplatin solution and carboplatin powder].

OBJECTIVE: To observe the efficacy and side effects of carboplatin solution in comparison with carboplatin powder. METHODS: A multicenter, open randomized controlled trial was carried out. A total of 121 patients with small cell lung cancer (SCLC), non-small cell lung cancer (NSCLC) and breast cancer (BC) were randomized into carboplatin injection group (study group) and carboplatin powder group (control group), treated with CE, CIE and CAF protocol, respectively. The same drugs and dosage were used in the two groups. RESULTS: The response rate of the study group and the control group was 94% and 67% in SCLC (P > 0.05), 12% and 22% in NSCLC (P > 0.05) and 71.4% and 67% in BC (P > 0.05), respectively. The major side effects were myelosuppression and gastrointestinal reactions, the frequency and intensity of which were statistically not different in the 2 groups of patients. CONCLUSION: Carboplatin solution is as effective as carboplatin powder for the treatment of cancer.

[Determination of coenzyme Q and alpha-tocopherol in mouse plasma and tissue mitochondria by liquid chromatography with electrochemical detection].

A convenient and reliable liquid chromatographic method with electrochemical detection was developed for the simultaneous determination of reduced coenzyme Q(ubiquinol), oxidized coenzyme Q(ubiquinone) and alpha-tocopherol in various tissues. The compounds of interest were separated on a reverse-phase C18 column under the mobile phase of 7 g/L NaClO4 in ethanol-methanol-70% HClO4 (800:200:1, volume ratio) and detected simultaneously by using a series of detection cells. Electrochemical guard cell (upstream of the injector) was used for eliminating interference for mobile phase. Ubiquinones were reduced in the -550 mV conditioning cell (downstream of the column). The resultant ubiquinols and original ubiquinols and alpha-tocopherol existing in the sample were easily oxidized in the 150 mV analytical cell. The linear range of the compounds were from 5 micrograms/L to 200 micrograms/L with correlation coefficients from 0.9976 to 0.9999. The detection limits were from 0.23 microgram/L to 0.68 microgram/L. The method was applied satisfactorily to the simultaneous determination of the amounts of ubiquinol-9, ubiquinol-10, ubiquinone-9, ubiquinone-10 and alpha-tocopherol in mice plasma and tissue mitochondria.

Nuclear localization of turnip crinkle virus movement protein p8.

Turnip crinkle virus (TCV) is a single-stranded positive-sense RNA virus of the Carmovirus genus. Two of its five open reading frames (ORFs), encoding proteins of 8 and 9 kDa, are required for cell-to-cell movement of the virus. These movement proteins (MPs) were fused to green fluorescent protein (GFP) to determine their cellular localization. In protoplasts, p9-GFP, like GFP itself, is found throughout the cytoplasm, as well as in cell nuclei. In contrast, p8-GFP was confined to the cell nucleus. Similar localization patterns were observed when specific small peptide epitopes were fused to p8 and p9 proteins instead of GFP. The cytoplasmic localization of p9-GFP and nuclear localization of p8-GFP were also detected in leaves after particle bombardment of DNA encoding these fusion proteins or after overexpression of p8-GFP in transgenic Arabidopsis seedlings. The expression of the GFP fusion proteins by recombinant TCV viruses in infected protoplasts or on inoculated Arabidopsis leaves produced similar patterns. Unlike TMV-MP and other MPs studied to date, no obvious punctuate expression in the cell wall or association with the cytoskeleton was detected. The sequence analysis of p8 revealed two unique nuclear localization signals (NLSs), which were not conserved within p8 homologues of other viruses in the genus Carmovirus. Mutation in either of these NLSs did not disrupt the nuclear localization of p8-GFP. However, when both NLSs were mutated, p8-GFP expression was no longer restricted to cell nuclei. The NLSs are not required for cell-to-cell movement; TCV recombinant viruses mutated in one or both NLSs could still facilitate cell-to-cell movement of the virus. The nuclear localization of p8 suggests a novel function for this protein in the cell nucleus.

HRT gene function requires interaction between a NAC protein and viral capsid protein to confer resistance to turnip crinkle virus.

An Arabidopsis protein was found to interact specifically with the capsid protein (CP) of turnip crinkle virus (TCV) through yeast two-hybrid screening. This protein, designated TIP (for TCV-interacting protein), was found to be a member of the recently recognized NAC family of proteins. NAC proteins have been implicated in the regulation of development of plant embryos and flowers. TIP alone was able to activate expression of reporter genes in yeast if fused to a DNA binding domain, suggesting that it may be a transcriptional activator. The TIP binding region in the TCV CP has been mapped to the N-terminal 25 amino acids. Site-directed mutagenesis within this region revealed that loss of the TIP-CP interaction in the yeast two-hybrid assay correlated with loss of the ability of TCV to induce the hypersensitive response and resistance in the TCV-resistant Arabidopsis ecotype Dijon (Di-0 and its inbred line Di-17). These data suggest that TIP is an essential component in the TCV resistance response pathway.

Anti-Epstein-Barr virus (EBV) activity of beta-L-5-iododioxolane uracil is dependent on EBV thymidine kinase.

beta-L-5-Iododioxolane uracil was shown to have potent anti-Epstein-Barr virus (EBV) activity (50% effective concentration = 0.03 microM) with low cytotoxicity (50% cytotoxic concentration = 1,000 microM). It exerts its antiviral activity by suppressing replicative EBV DNA and viral protein synthesis. This compound is phosphorylated in cells where the EBV is replicating but not in cells where the EBV is latent. EBV-specific thymidine kinase could phosphorylate beta-L-5-iododioxolane uracil to the monophosphate metabolite. The K(m) of beta-L-5-iododioxolane uracil with EBV thymidine kinase was estimated to be 5.5 microM, which is similar to that obtained with thymidine but about fivefold higher than that obtained with 2' fluoro-5-methyl-beta-L-arabinofuranosyl uracil, the first L-nucleoside analogue discovered to have anti-EBV activity. The relative V(max) is seven times higher than that of thymidine. The anti-EBV activity of beta-L-5-iododioxolane uracil and its intracellular phosphorylation could be inhibited by 5'-ethynylthymidine, a potent EBV thymidine kinase inhibitor. The present study suggests that beta-L-5-iododioxolane uracil exerts its action after phosphorylation; therefore, EBV thymidine kinase is critical for the antiviral action of this drug.

Cap-independent translational enhancement of turnip crinkle virus genomic and subgenomic RNAs.

The presence of translational control elements and cap structures has not been carefully investigated for members of the Carmovirus genus, a group of small icosahedral plant viruses with positive-sense RNA genomes. In this study, we examined both the 5' and 3' untranslated regions (UTRs) of the turnip crinkle carmovirus (TCV) genomic RNA (4 kb) as well as the 5' UTR of the coat protein subgenomic RNA (1.45 kb) for their roles in translational regulation. All three UTRs enhanced translation of the firefly luciferase reporter gene to different extents. Optimal translational efficiency was achieved when mRNAs contained both 5' and 3' UTRs. The synergistic effect due to the 5'-3' cooperation was at least fourfold greater than the sum of the contributions of the individual UTRs. The observed translational enhancement of TCV mRNAs occurred in a cap-independent manner, a result consistent with the demonstration, using a cap-specific antibody, that the 5' end of the TCV genomic RNA was uncapped. Finally, the translational enhancement activity within the 5' UTR of 1.45-kb subgenomic RNA was shown to be important for the translation of coat protein in protoplasts and for virulent infection in Arabidopsis plants.

Time-frequency representation of epicardial electrograms during ventricular fibrillation.

In the present study we quantified changes in dominant frequency, which is reciprocal of activation interval or cycle period, during ventricular fibrillation (VF). We used a Smoothed Pseudo Wigner Distribution (SPWD) to estimate time-frequency representations of epicardial electrograms recorded in swines. We used a sock with 64 electrodes spaced equally to record electrograms during 30 seconds of electrically induced VF. Results from 29 trials in three animals showed a mean dominant frequency of 6.64 Hz. We observed considerable variation in dominant frequencies during VF. Temporally, the frequencies varied by as much as +/- 1.24 Hz (2 standard deviations). Spatial variation in frequencies was +/- 1.20 Hz. Cycle periods were computed as the reciprocal of dominant frequencies obtained from the SPWD. These cycle periods were verified to be numerically similar to the cycle periods estimated using activation times detected from differentiated electrograms. Results of recent studies by others have shown that cycle periods during VF are correlated with refractory periods. Our results show that a non-stationary analysis technique such as the SPWD can be used to track spatio-temporal variation in cycle periods. These changes can be used to investigate spatio-temporal variation in cellular properties such as the effective refractory periods during VF. The substantial temporal variation in dominant frequencies that we observed suggest the possibility that the excitable gap at any epicardial location also varies considerably from one instance to another during a VF episode.

Time averaged spatial distribution of epicardial dominant frequencies during ventricular fibrillation.

Recent evidence suggests that the dominant frequencies during ventricular fibrillation (VF) may be used as indicators of dispersion in repolarization and in activation patterns. In the present study, we quantified dominant frequencies from multiple epicardial electrodes to investigate if there are differences in the averaged frequencies within the electrograms recorded from the left and the right ventricles. Further, we quantified whether the difference in average frequency between the two ventricles changed during 30 seconds of VF. Results from eighteen trials in two pigs showed that during the entire duration of VF the average dominant frequencies of all electrodes over the left ventricle were higher than those over the right ventricle (p < 0.005). The dominant frequencies are reciprocal of cycle periods or activation intervals during VF. Our results show that on average, activations in the left ventricle occurred at a faster rate than those in the right ventricle. Activation intervals at any site are determined by the refractory period at that site and the arrival time of next activation. Although differences in cellular properties may have contributed to the observed differences in activation intervals between the ventricles, it is possible that activation arrival times may be different as well. It is possible that the increased tissue mass of the left ventricle may increase the probability that any site will get excited at a faster rate after it is recovered from previous activation.

[Biotherapy and biochemotherapy of patients with malignant melanoma].

OBJECTIVE: To evaluate the efficacy of postoperative biochemotherapy on survival of patients with malignant melanoma. METHODS: One hundred and five patients with malignant melanoma received postoperative biotherapy/biochemotherapy or radiotherapy/chemotherapy. The median time of follow-up was 3 years (from 1 to 5 years). RESULTS: The median survival time (MST) in the whole series of patients was 27 months (range: 2-72 months). The MST in patients received postoperative biotherapy/biochemotherapy (57 cases) was 32 months with a 3-year survival rate of 36.8%. That in patients received postoperative radiotherapy/chemotherapy (54 cases) was 20 months. CONCLUSION: Biotherapy/biochemotherapy following surgery may significantly improve survival in patients with malignant melanoma.

Determination of aspartame by ion chromatography with electrochemical integrated amperometric detection.

In this paper, the separation and determination of the sweetener aspartame by ion chromatography coupled with electrochemical amperometric detection is reported. Sodium saccharin, acesulfame-K and aspartame were separated using 27.5 mmol/l NaOH isocratic elution on a Dionex IonPac AS4A-SC separation column. Aspartame can be determined by integrated amperometric detection without interference from the other two sweeteners. The method can be applied to the determination of aspartame in powered tabletop, fruit juice and carbonated beverage samples, and the results obtained by integrated amperometry were in agreement with those obtained using a UV detection method. A method for determining analytes with an NH2 group by ion chromatography with integrated amperometry was developed.

Ion chromatographic determination of taurine in medicine, nutrient capsule and human urine with electrochemical detection.

A very simple and sensitive method for the determination of taurine by ion chromatography with electrochemical integrated pulsed amperometry is firstly described. Taurine was determined using 160 mmol/l NaOH as eluent and a Dionex CarboPac PA1 separation column (250x4 mm I.D.) without the interference with ten kinds of common amino acids. The peak area response of taurine was linear in the range 0.1-20 microg/ml, the detection limit was 0.034 microg/ml. The method has been applied successfully in the determination of taurine in medicinal granule, nutrient capsule and human urine. The content determined in medicinal granule is consistent with that marked by the manufacturer.

Structure-activity relationships of L-dioxolane uracil nucleosides as anti-Epstein Barr virus agents.

A series of 1,3-dioxolanyluracil analogues was prepared from the dioxolane intermediates 2, and their anti-Epstein Barr virus (anti-EBV) activities were determined. The potency of L-dioxolane uracil nucleosides against EBV replication is dependent on the substituents at the 5-position in the following decreasing order: I > Br > Cl > CH3 > CF3 > F. The most active and selective analogue was the iodo derivative (L-I-OddU) with an EC50 value of 0.03 microM and an EC90 value of 0.16 microM. There was no cytotoxicity or depletion of mitochondrial DNA in cells after exposure to L-I-OddU at 50 microM. The action against EBV replication in H1 cells is time-dependent, and EBV DNA in cells treated with L-I-OddU could rebound to pretreatment levels once the drug was removed. In view of the potent antiviral activity plus favorable toxicity profiles, L-I-OddU may be potentially useful for the treatment of EBV-related infectious diseases as well as for delaying the onset or decreasing the incidence of EBV-associated cancers.
