Analysis of a viral integration event in a CG-rich region at the 1p36 human chromosomal site.

The preinsertion site of an adenovirus-5/simian virus 40 recombinant construct (Ad5/SV40) has been cloned and sequenced. Our data suggest that viral integration has occurred in a genomic region which has been the target of multiple events of Alu element retropositions within a TAA minisatellite. Extensive homologies between the left viral end and the host cellular DNA were also observed. The compositional similarity between Adenoviridae and the region of viral integration is consistent with the observed insertion of exogenous DNA in isochores of similar composition [G. Bernardi, Annu. Rev. Genet. 23 (1989) 637-661].

Chromatin structure, DNA methylation, and gene expression at sites of viral integration in human fibroblasts. Implications for chromosomal fragility.

We analyzed the structural and functional properties of a chromosomal region in which a recombinant hybrid virus adenovirus 5/SV40 preferentially integrates. Our results demonstrated that the structure of the cellular targets for DNA and RNA viruses is very similar and that the cellular sequence flanking the integrated virus possesses, simultaneously, all the features postulated to be the molecular basis for chromosomal fragility.

Core histone acetylation during lymphocyte activation.

Histone acetylation has been followed in cultures of human lymphocytes, in PHA-stimulated lymphocytes and in mixed lymphocytes obtained from identical twins and from unrelated donors. A computer assisted analysis of two-dimensional gels and autoradiograms revealed that in cultured lymphocytes only H3 and H4 core histones incorporate labeled acetate and that two H3 variants greatly differ in their rate of acetate uptake.

Establishment and characterization of a new human cloned myelomonocytic cell line (ZC-1.6).

A human cell line, ZC, derived from the blood of a patient with acute myelomonocytic leukemia, was established and cloned. One of the clones, ZC-1.6, was subsequently characterized. As for its morphology and cytochemistry, ZC-1.6 clone shares a number of features with immature cells of the monocytic or myelocytic lineages. Surface marker analysis shows positivity for 4F2 (100% of the cells), and OKM1 (38%) monoclonal antibodies, and presence of surface HLA-D/DR antigens (100%) and Fc (11%) and complement (C3b) receptors (100%). Functional capabilities of ZC-1.6 cells include adherence, spreading, and phagocytosis of latex and opsonized zymosan particles. Despite its morphological immaturity, the ZC-1.6 clone produces relevant amounts of O2- in the presence of different stimuli (phorbol myristate acetate, opsonized zymosan, or latex particles). The production of O2- by ZC-1.6 cells is the first evidence that reactive oxygen intermediate production may represent an early feature of cells of the myelomonocytic lineage.

Progesterone enhances reactive oxygen intermediates production by cultured human monocytes.

Progesterone at placental tissue concentrations (5-20 micrograms/ml) markedly increases in vitro PMA-stimulated O-2 and H2O2 production by human cultured monocytes. This appears to contrast the well-known suppressive action of the hormone on the other cell mediated defence mechanisms. We suggest that these findings could cast a new light on progesterone's multiple and differentiated functions in the placental environment.
