The wrap-around problem and optimal padding in the exit wave reconstruction using HRTEM images.

A free fast Fourier transform software FFTW (www.fftw.org) is used in the exit wave reconstruction and shown to be able to handle images of any size effectively. The wrap-around problem commonly encountered in the exit wave reconstruction is investigated, and a new padding scheme is proposed and its performance is investigated. It is shown that while the usual wrap-around problem may be solved via padding the input high-resolution transmission electron microscopy (HRTEM) images with zero or averaged image intensities, the reconstructed exit wave at its boundary is inevitably distorted by the discontinuity of image intensities at the boundary. The proposed new padding scheme is shown to be able to provide a good estimate of the image intensities immediately outside the boundaries of the HRTEM images, reducing the estimated image intensity discontinuity across the boundaries and therefore providing a larger undistorted view of the exit wave.

Quantitative analysis of defects and domain boundaries in mesoporous SBA-16 films.

Some quantitative structural analyses on defects and domain boundaries observed in SBA-16 films were performed using the lattice concept and geometric phase method. These analyses show that there exist low angle, high angle and translational anti-phase domain boundaries in SBA-16 films. While some of the domain boundaries bear analogue to those found in normal solid crystals, others are similar to that found in the liquid crystals. In particular near Sigma11 and Sigma13b high angle boundaries were observed. On the one hand the Sigma11 boundaries were found to exist with or without steps (ledge) associated with them depending on whether or not the boundary plane is parallel to the densely packed lattice plane. On the other hand segments of the boundary plane in the Sigma13b boundary were found being always associated with densely packed lattice plane, with the {011} type of lattice plane in one domain being parallel to the {112} type of plane in the other domain. The translational domain boundary was observed to have a translation vector having a projected component of (1/2) <110> on the (111) plane. The bending deformation similar to that found in the nematics liquid crystal was also observed and quantitatively analyzed using the geometric phase method, and rotational field associated with the deformation was identified.

Orbital susceptibilities of PbSe quantum dots.

Different sizes of three-dimensional PbSe quantum dots have been synthesized for the study of orbital magnetic susceptibilities. Two types of orbital susceptibilities have been found, including the Curie susceptibility and finite-size corrections to the Landau susceptibility. The Curie term of a quantum dot manifests itself in the temperature dependence of magnetic susceptibility at low temperatures, while the field dependence of differential susceptibility at high temperatures shows finite-size corrections to the Landau susceptibility. Both of the two kinds of orbital susceptibility, estimated per quantum dot, show linear dependence on the size.

X-ray absorption spectroscopy investigations on oxidized Ni/Au contacts to p-GaN.

X-ray absorption spectroscopy was used to investigate the electronic structure of as-deposited and oxidized Ni/Au contacts to p-GaN and to elucidate the mechanism responsible for low impedance. X-ray absorption near edge spectra of Ni K- and L3,2-edges clearly indicate formation of NiO on the sample surface after annealing. The reason for low impedance may be attributed to increase in hole concentration and existence of p-NiO layer on the surface.

Four-dimensional dielectric property image obtained from electron spectroscopic imaging series.

We have demonstrated a new quantitative method to characterize two-dimensional distributions of energy-dependent dielectric function of materials from low loss electron spectroscopic image (ESI) series. Two problems associated with extracted image-spectrum from the low-loss image series, under-sampling and loss of energy resolution, were overcome by using fast Fourier transformation (FFT) interpolation and maximum entropy deconvolution method. In this study, Black Diamond/Si3N4/SiO2/Si-substrate dielectric layer designed for copper metallization was used as the sample. We show that the reconstructed (FFT interpolated and maximum entropy deconvoluted) image-spectrum obtained from ESI series images can be quantified with the same accuracy as conventional electron energy-loss spectroscopy spectra. Since the analysis of the dielectric function is sensitive to the local thickness of the specimen using Kramers-Kronig analysis, we also developed a new method to quantitatively determine the dielectric constant for low-k materials. We have determined the thickness of the Black Diamond using the extrapolated thickness method from the materials of known dielectric constants. Using Kramers-Kronig formula, the dielectric function map can be deduced from two-dimensional reconstructed single scattering spectra with providing the information of thickness. We proposed a four-dimensional data presentation for revealing the uniformity of the energy dependent property. The accuracy of our methods depends on the thickness determination and on the quality of the reconstructed spectra from the image series.

Extension of HRTEM resolution by semi-blind deconvolution method and Gerchberg-Saxton algorithm: application to grain boundary and interface.

A generalized maximum entropy method coupled with Gerchberg-Saxton algorithm has been developed to extend the resolution from high-resolution TEM image(s) for weak objects. The Gerchberg-Saxton algorithm restores spatial resolution by operating real space and reciprocal space projections cyclically. In our methodology, a generalized maximum entropy method (Kullback-Leibler cross entropy) dealing with weak objects is used as a real space (P1) projection. After P1 projection, not only are the phases within the input spatial frequencies improved, but also the phases in the next higher frequencies are extrapolated. An example of semi-blind deconvolution (P1 project only) to improve the resolution in SiC twin boundary is shown. The nature of the bonding in this twin boundary is Si-C but it was rotated 180 degrees along the boundary normal. The optimum solution from P1 projection can be further improved by a P2 projection. The square roots of diffraction intensities from a diffraction pattern are then substituted to complete a cycle operation of the Gerchberg-Saxton algorithm. Application examples of Gerchberg-Saxton algorithm to solve the atomic structure of defects (2 x 1 interfacial reconstruction and dislocation) in NiSi2/Si interfaces will be shown also.

[Effect of songling xuemaikang capsule combined with captopril on quality of life in primary hypertension patients].

OBJECTIVE: To compare the hypotensive effects of captopril and combined medication of Songling Xue-maikang Capsule (SXC) with captopril on quality of life (QOL) in treating primary hypertension patients. METHODS: For 166 hypertension patients, a perspective randomized double blind study was performed with both sexes, aged 42-78 years, blood pressure (BP) ranged > or = 140/90 mmHg and < or = 200/110 mmHg. Patients were randomly divided into two groups: captopril group and combined medication group. Each patient should answer the QOL questionnaire independently before and after experiment. 60 age-matched normotensive subjects without chronic diseases served as control. RESULTS: Both hypertension groups showed an improvement of BP and QOL after treatment, but the combined medication group had a higher score in sense of well being, physical symptom-signs, work performance and life satisfaction than the captopril group had. CONCLUSION: Combined medication of captopril and SXC had better effect than captopril alone either in lowering BP or in improving QOL of primary hypertension patients.

An evaluation of chromogenic substrates for characterization of serine protease produced by pathogenic Vibrio alginolyticus.

Four chromogenic substrates for characterizing serine protease of Vibrio alginolyticus were evaluated. The protease activity of bacterial extracellular products, or the fractions of 33 kD protease purified by the AKTA purifier system with various columns, was completely inhibited by ethylenediamine tetra-acetic acid, ethylene glycol-bis(beta-amino-ethyl ether) N,N,N',N'-tetraacetic acid (EGTA), antipain and phenylmethylsulphonyl fluoride (PMSF) using water-soluble substrates (azoalbumin and azocasein). It was only completely inhibited by antipain and PMSF using water-insoluble substrates (azocoll and hide powder azure). The protease activity was not, or only partially, inhibited by 1,10-phenanthroline and sodium dodecyl sulphate (SDS) using all four substrates. Since chelating agents and 1,10-phenanthroline are commonly employed as inhibitors to identify metalloprotease, the two water-soluble substrates may not be appropriate for this purpose, except for using 1,10-phenanthroline as an inhibitor. Chelating agents may be still applicable as inhibitors using water-insoluble substrates and 1,10-phenanthroline is highly recommended in the characterization for metalloprotease to avoid confusion. In the present study, the 33 kD protease was further confirmed as an SDS-resistant serine protease and not a metalloprotease.

Purification and partial characterization of a toxic serine protease produced by pathogenic Vibrio alginolyticus.

An extracellular lethal toxin produced by Vibrio alginolyticus strain Swy originally isolated from diseased kuruma prawn (Penaeus japonicus) was purified using the AKTA purifier system with hydrophobic interaction chromatography, anion exchange and gel filtration columns. The toxin is an alkaline serine protease, inhibited by phenyl methylsulphonyl fluoride (PMSF), antipain and shows maximal activity at pH 8 to 11, having a pI of 4.3 and a molecular weight of approximately 33 kD. The toxin was completely inhibited by FeCl2 but partially inhibited by 3,4-dichloroisocoumarin (3,4-DCI), ethylenediamine tetraacetic acid (EDTA), ethylene glycol-bis(beta-amino-ethyl ether) N,N,N',N'-tetraacetic acid (EGTA), CuCl2 and ZnCl2. The purified protease was lethal for kuruma prawn at an LD50 of 0.29 microgram protein/g body weight. The haemolymph withdrawn from the moribund prawns injected with the toxic protease was unable to clot. The coagulogen in the kuruma prawn plasma showed an increased migration rate after incubation with this serine protease, and a plasma colour change from blue to pink was recorded. The addition of PMSF completely inhibited the lethal toxicity of the purified protease, indicating that this serine protease was a lethal toxin produced by the bacterium. The 33 kD protease was therefore a toxic protease produced by V. alginolyticus strain Swy.

Interaction between obesity and genetic polymorphisms in the apolipoprotein CIII gene and lipoprotein lipase gene on the risk of hypertriglyceridemia in Chinese.

To understand the effects of the interaction between genetic polymorphisms and obesity on the risk of hypertriglyceridemia (HTG), two polymorphisms, an SstI polymorphism on the apolipoprotein CIII gene and a HindIII polymorphism on the lipoprotein lipase gene, were analyzed in 339 Chinese subjects with (82 cases in the HTG group) or without HTG (257 cases in the control group). Our data revealed that the frequencies of obesity, the SstI minor allele (S2), and the HindIII major allele (H+) in the HTG group were significantly higher than in the control group. Subgroup analysis revealed that the association between these two polymorphisms and HTG occurred predominantly in nonobese subjects and in subjects with the less hypertriglyceridemic genotype of another polymorphism. Multivariate logistic regression analysis showed that all three risk factors (obesity, S2-containing chromosome, and H+ homozygosity) were associated with HTG, and an interaction was found between obesity and H+ homozygosity for the occurrence of HTG. The risk of HTG increased significantly with combinations of risk factors. Subjects can be divided into low or high risk groups for HTG using such combinations. These results provide evidence of interaction between obesity and the HindIII polymorphism of the lipoprotein lipase gene on the risk of HTG.

Effects of extracellular products of Vibrio alginolyticus on penaeid prawn plasma components.

The effects of both crude extracellular products (ECP) and a partially purified protease of Vibrio alginolyticus on the plasma components of kuruma prawn (Penaeus japonicus) and tiger prawn (P. monodon) were studied using crossed immunoelectrophoresis (CIE). A component of the plasma, tentatively identified as coagulogen, apparently disappeared after incubation with the ECP, while the amount of a component tentatively identified as haemocyanin decreased. The coagulogen and an unknown component (component 1) in the penaeid plasma showed an increased migration rate after incubation with a partially purified 33 kDa protease of the bacterium. In contrast, incubation with protease had no detectable effect on the amount of haemocyanin. These events may significantly contribute to the pathogenicity of Vibrio alginolyticus in penaeids.

Virulence of Vibrio alginolyticus isolated from diseased tiger prawn, Penaeus monodon.

Outbreaks of mass mortality among cultured tiger prawns (Penaeus monodon) with white spotted syndrome (WSS) in the carapace occurred in the summer of 1994 in I-Lan, Taiwan. A swarming strain Val was isolated from hemolymph of the moribund prawns with tryptic soy agar (TSA, supplemented with 1% NaCl, Oxoid) and/or thiosulfate citrate bile salt sucrose (TCBS, Difco) agar. This strain was characterized and identified to be Vibrio alginolyticus. The strain was susceptible to antibiotics such as chloramphenicol, ciprofloxacin, doxycycline hydrochloride, nalidixic acid, oxolic acid, and oxytetracycline while resistant to ampicillin, novobiocin, penicillin G, sulfisoxazole, and sulfonamide. The bacteria and their extracellular products (ECP) were lethal to both tiger prawns (P. monodon) and kuruma prawns (P. japonicus) with LD50 values of 1.13 x 10(5), 2.46 x 10(5) CFU/g, and 0.23, 0.63 micrograms protein/g prawn body weight, respectively.

[Pregnancy after cardiac valve replacement: analysis of 21 cases].

A retrospective analysis of 21 cases with 23 pregnancies after cardiac valve replacement from August 1979 to March 1992. In 11 cases mechanical and in 10 biological valves were used, and in 18 cases with mitral valve replacement was done (MVR), in 1 case aortic valve replacement (AVR) and in 2 cases, both MVR and AVR were carried out 22 pregnancies in 20 cases with cardiac function grading I and II (New York Heart Association, NYHA) before pregnancy resulted in normal term deliveries and 1 case with grading III prior to pregnancy died of heart failure in the third trimester. All the cases treated with mechanical valve replacement received warfarin derivatives throughout pregnancy, and no malformation was found in their neonates. Issues related to pregnancy after cardiac valve replacement were discussed.

Effects of pulse excimer laser irradiation on the human left ventricular wall.

This study was to assess the potential application of excimer lasers in the ablation of myocardium in vitro for the treatment of constant ventricular tachycardia or hypertrophic cardiomyopathy. A fresh human heart and EMG model 103 XeCl pulse excimer laser machine were used. The pulse repetition rate varied from 1 to 7 Hz. Irradiation directly on the left endocardial and epicardial walls lasted for 10 seconds and was repeated 3 times, creating 3 craters. The histological changes were examined by light microscope. Results showed very close relations between the depth or volume of vaporized craters and the pulse repetition rate on the endocardial (r = 0.9674, P less than 0.001 and r = 0.8962, P less than 0.01, respectively) and epicardial walls (r = 0.9602, P less than 0.001 and r = 0.9612, P less than 0.001, respectively). A sharp, clear border without debris or coagulating necrosis was seen under the microscope. We concluded that the pulse excimer laser, differing from Ar+ or Nd:YAG lasers, might be a powerful tool for manipulating the human ventricular wall, but more work needs to be done before it can be widely applied in the treatment of cardiovascular disease.
