[Results of postoperative radiotherapy in breast cancer (author's transl)].

The results are presented of postoperative irradiation in the treatment of breast cancer at the Department of Radiotherapy and Radiobiology, University of Vienna. Our data compare well with the best results reported in the literature. Apart from the importance of high loco-regional freedom from disease for the quality of life of these patients, the related increase in survival prospects is stressed.

[The value of postoperative irradiation in the curative management of breast cancer (author's transl)].

A report is presented of the value of post-operative irradiation in patients with operable breast cancer. A combination of conservative surgery followed by irradiation provides optimal loco-regional results. Improved 10-year survival rates have already been reported with adequate irradiation of the peripheral lymphatics. The location and size of the primary tumour, as well as exact quantitative statements about histologically positive axillary lymph nodes are parameters of utmost importance.

Lipogenesis in Ehrlich ascites tumor cells under anaerobic culture conditions.

Anaerobic culture conditions (95% argon/5% CO2) caused a slightly greater increase in total lipids of Ehrlich ascites tumor cells than a gas phase of 20% O2, 75% N2, 5% CO2. Whereas the rate of [U-14C]acetate incorporation into total lipids and lipid-subclasses rose markedly in the absence of oxygen, a drastic decrease of [U-14C]pyruvate and [1-14C]octanoate incorporation as well as a 30% reduction of 3H incorporation into lipids from tritiated water were observed under these conditions. Since profound changes in the metabolic state of cells cause alterations in the specific activity of the acetyl-CoA pool but do not alter the specific activity of intracellular water, this precursor is considered to be an adequate monitor for lipogenesis under aerobic and anaerobic culture conditions. Therefore, it is concluded that Ehrlich ascites tumor cells are not able to reoxidize NADH/NADPH in the absence of oxygen by a stimulation of biosynthesis of fatty acids as is discussed to be the case in normal cells. The slight increase in total lipids of anaerobically cultured cells seems to be the result of an imbalance between normal uptake and impaired utilization of lipids from serum-supplemented culture medium.

Unexpectedly low incorporation of isotopic acetate into lipids of Ehrlich ascites tumor cells cultured in lipid-poor medium.

The present communication reports data on the lipid biosynthesis of Ehrlich ascites tumor cells grown in culture media supplemented with modified sera. Whereas the metabolisms of [14C]pyruvate and [14C]mevalonate are identical in all media tested, the incorporation of [14C] acetate is higher in medium with dialyzed serum than in medium with delipidized serum; it is suppressed in the absence of all lipids in culture medium. Cellular integrity is not impaired in modified media. The results indicate that acetyl-CoA synthetase of Ehrlich ascites tumor cells is not regulated by exogenous lipids as is known to be the case in nonmalignant cells.

[Control of parturition in swine. 2. Study results of parturition synchronization in swine with the prostaglandin analog cloprostenol (I.C.I. 80.996: Estrumate)].

Premature parturition was induced in 115 of 121 sows (95%) that had been given injections of 150 microgram or 200 microgram of "Estrumate", a prostaglandin analogues, on the 112th and 113th days of gestation. The average time that elapsed between injection and parturition of those 115 animals was 27.2 +/- 7.7 hours, with no difference being observed between gilts, on the one hand, and old sows, on the other. The time of gestation of the treated animals was shortened with significance from 114.6 +/- 1.6 days for the controls to 113.2 +/- 0.4 days. The average duration of parturition proper was 3.7 +/- 2.2 hours, the average litter being 11.2 +/- 3.3 piglets. The total number of piglets born was 1,320, with stillbirths accounting for 4.17%. The average weight at birth, 1,330 +/- 210 g, and the weight at weaning did not differ from weight figures recorded from the controls. The courses of parturition and puerperium were in agreement with normal biological standards. The sows treated were tested for their blood counts close to full term and for their endometrial histomorphology after weaning, with no side effects of "Estrumate" treatment being established. The behaviours of progesterone, 17-beta oestradiol, and cortisol in serum of "Estrumate"-treated sows were investigated, and congruence was established between spontaneous and prostaglandin-induced parturitions, which was in agreement with international literature. The conclusion is that 150 micrograms of 'Estrumate", administered to swine, after the 11th day of gestation has very good labour-inducing effects and ensures normal physiological parturition and piglet development.

Anaerobiosis and oxygen recovery: changes in cell cycle distribution of Ehrlich ascites tumor cells grown in vitro.

The cell cycle distribution of in vitro cultured Ehrlich ascites tumor (EAT) cells was analysed by pulse-cytophotometry to characterize the growth cessation observed under anaerobic conditions. DNA histograms provided evidence that in the absence of oxygen EAT cells accumulate in the G1 and early S phase of the cell cycle while in the presence of oxygen an increase in G2 was observed during 24h culture period. Cellular recovery from anaerobiosis was observed soon after transfer of the cells into fresh aerobic culture medium but occurred slowly if the cells were only resupplied with air. Cell cycle analyses as well as (14C)-thymidine incorporation suggest considerable synchronization results from the introduction of anaerobiosis.

Histological investigation of aortic wall in experimental sclerosis of rabbits.

Rabbit aortic changes were investigated after different sclerogenic diets. A subintimal chondroitin sulfate layer was characterized by toluidine blue metachromatic staining at pH 1--3, CEC value expressed in MgCl2 concentration: 0.4M, hyaluronidase sensitivity. This layer becomes disorganized during plaque formation and partly disaappears. A long-lasting sclerogenic diet, as well as a diet containing several sclerogenic factors, produced an increase of frequency and extension of the media necrosis, calcification, and chondroid metaplasia of the aortic wall. In these lesions, an increase of mucopolysaccharide secretion of modified smooth muscle cells was observed.

[A new diet for inducing early experimental atherosclerosis in rabbits].

Authors by a new combined diet consisting of cholesterol vitamin D3 and sour cream produced clinico-biochemical changes characteristic for atherosclerosis in a very short period in rabbits. This diet caused severe lipoproteinaemia, hypercholesterinaemia and microscopis and macroscopic changes in the aortic wall. Authors assume, that this diet is suitable for producin atherosclerosis in a short period.

Chemical investigations on pig kidney aminoacylase.

1. Preparations of purified pig kidney aminoacylase (N-Acylamino-acid amidohydrolase, EC 3.5.1.14) were obtained by Sephadex and DEAE-cellulose chromatography in homogeneous form as judged by polyacrylamide gel electrophoresis and immunoelectrophoresis. 2. The apparent molecular weight of the enzyme, determined by gel filtration, was about 86 000. After treatment with mercaptoethanol, performic acid or sodium dodecyl sulphate a band with an apparent molecular weight of approximately 43 000 was observed in polyacrylamide gels containing sodium dodecyl sulphate. Thus pig kidney aminoacylase seems to be composed of two subunits. 3. The amino acid composition of the enzyme was determined. Aminoacylase contains 772 amino acids, which corresponds to a molecular weight of 85 500. 12 tryptophan and 12 half-cystine residues were found. 4. Each subunit of the enzyme contains two -SH groups of different reactivity and two disulfide bonds one of which is easily cleaved by -SH compounds, the second only by performic acid oxidation. 5. Chemical modification of two -SH groups abolishes the catalytic activity of aminoacylase. Cleavage of two disulfide bonds also inactivates the enzyme. It is suggested that the enzyme has two active sites each containing an essential -SH group and disulfide bond. One active site is assumed to be part of each subunit.
