Detection of bile salt-dependent lipase, a 110 kDa pancreatic protein, in urines of healthy subjects.

Bile salt-dependent lipase (BSDL), a 110 kDa glycoprotein secreted by the pancreatic acinar cells, participates in the duodenal hydrolysis of dietary lipid esters. Recent in vitro and in vivo studies demonstrated that the BSDL reaches the blood via a transcytosis motion through enterocytes, suggesting that this enzyme may play a role in vascular biology. Once in the blood, BSDL should be eliminated. We address the hypothesis that BSDL may be filtered by the glomerulus and eliminated in urines. Immunological methods and proteomic were used to detect and to characterize BSDL in urine. The immunoreactive form of BSDL was detected in urines of 36 male subjects devoid of renal failure. Proteomic demonstrated that the immunoreactive protein is BSDL. Experiments using a monoclonal antibody to the oncofetal glycoform of pancreatic BSDL suggested that the protein is not expressed by renal cells but originates from the pancreas via circulation. We demonstrate that under normal physiological conditions, BSDL, a high-molecular weight blood glycoprotein, can be filtered by the renal glomerulus to be eliminated in urines.

[Study of HIV localization in sperm].

The aim of the study was to investigate the presence and the localization of HIV in human ejaculate and its different components. Sixty-three ejaculates from 19 HIV-positive patients have been studied. By using cellular as well as molecular biology methods, we never detected HIV in living and mobile spermatozoa although we sometime found the virus in seminal liquid and in nuclear fractions. Up to date, frozen sperms free of HIV particles from 11 HIV-positive male partners were used to inseminate safely and with success the 11 HIV-negative female partners (no female partner as well as babies contamination). In conclusion, our procedure of HIV-free spermatozoa screening allowed discordant couples to have HIV-negative descendants. In contrast to the natural coïtus in ovulated period or to insemination with washed spermatozoa that are really "Russian roulette", our procedure ensure a total security of no contamination for the pregnant mother.

Effects of thymus humoral factor gamma-2 (THF gamma 2) on lymphohaematopoietic progenitor cells: an in vitro study.

Thymus humoral factor gamma-2 (THF gamma 2), an immunomodulatory factor, was previously shown to exert a regulatory effect upon human haematopoietic development. In the present study, we were primarily interested in determining whether THF gamma 2 has an immunorestorative effect after HIV infection. Although no inhibition of viral production by T-cluster-forming cells and their mature progeny was observed, this thymus-derived compound prevented the dramatic decrease in CD4+ cells observed in cultures of immature T cells from normal bone marrow infected with HIV 1.
