A simple method for estimation of gastric mucus and effects of antiulcerogenic agents on the decrease in mucus during water-immersion stress in rats.

In the present study Corne's technique that has been commonly used to quantify the gastric mucus in situ using the dye, Alcian blue, has been improved. Furthermore, the effects of antiulcerogenic agents on the decrease in gastric mucus gel during water-immersion stress were also studied. In order to increase the extraction of dye-recovery, we used 30% dioctyl sodium sulfosuccinate (docusate sodium, DSS) instead of 0.5 mol/l MgCl2 used in Corne's technique. The control values of the dye-recovery extracted with DSS were about 5 times higher than those with MgCl2. Indomethacin (20 mg/kg s.c.) failed to influence dye-recovery with DSS and MgCl2, while reserpine (5 mg/kg i.p.) significantly decreased the dye-recovery in both extractions. Water-immersion stress for 2 h decreased the dye-recovery with DSS but the dye-recovery with MgCl2 decreased only 15 min after water-immersion. These results suggest that extraction with DSS reflects the mucus gel secretion, and that dye-recovery with DSS is more sensitive to alteration of mucus gel than that with MgCl2. Bombesin (10 micrograms/kg), secretin (100 U/kg) and mild irritants increased the dye-recovery with DSS. Cimetidine remarkably reduced the mucus secretion and failed to improve the decrease in mucus during water-immersion stress. Pirenzepine and atropine dose-dependently prevented the decrease in the dye-recovery during water-immersion stress.

ATP-dependent [3H]Met-enkephalin uptake by bovine adrenal chromaffin granule membrane.

Uptake of [3H]Met-enkephalin by purified chromaffin granule membrane isolated from bovine adrenal medulla was investigated. Addition of a single divalent cation, such as Mg2+ or Ca2+, did not affect the uptake. The presence of 3 mM ATP in the incubation medium stimulated the uptake to two times that of the control. The same stimulation effect was also found in the presence of ATP plus a single divalent cation. The ATP-dependent uptake reached its half-maximal level within 5 min after initiation of the reaction at 25 degrees C, and reached a plateau within 10 min. The apparent Km for [3H]Met-enkephalin uptake by the chromaffin granule membrane was about 2.3 X 10(-6) M. GTP, CTP, UTP and ADP did not stimulate enkephalin uptake. Several calcium inhibitors such as trifluoperazine, verapamil and 3,4,5-trimethoxybenzoic acid 8-(N,N-diethylamino)octylester (TMB-8) markedly inhibited enkephalin uptake. The inhibition exceeded 80%, although these chemicals inhibited Ca2+-ATPase activity in chromaffin granule membrane only 30% under the same conditions. Ethyleneglycol-bis-(beta-aminoethylether)N,N'-tetraacetic acid (EGTA), at 3 mM also inhibited uptake about 30%. The results indicate that uptake of Met-enkephalin by chromaffin granule membrane ghosts was driven not only by ATPase activity, but also by some other Ca2+-ATP-mediated mechanism(s).

Management of polyuria subsequent to pituitary surgery based on the diurnal pattern of urinary excretion.

Polyuria subsequent to pituitary surgery was studied in 64 cases. Most cases of postoperative polyuria were due to diabetes insipidus. These cases showed a triphasic pattern in daily urinary volume. Observation of hourly urinary volume in polyuria revealed four diurnal patterns of urinary excretion: rhythmic, continuous, transient, and unspecific. Clinical observation of diurnal patterns has an advantage, in terms of simplicity of procedure, in immediately determining the nature of the polyuria, prognosticating diabetes insipidus, and eliminating inappropriate procedures in treatment. Indomethacin suppository is considered to be a favorable agent in reducing polyuria without disturbing the diurnal pattern in diabetes insipidus.

Gastric cytoprotection by pirenzepine in rats: evaluating method for cytoprotective activity by antisecretory agents.

The effects of antisecretory agents, pirenzepine, atropine and cimetidine, on gastric mucosal lesions induced in rats by ethanol, HCI (0.6 N), HCI-acidified 50% ethanol and HCI-acidified 50 mM sodium taurocholate (TCA) were comparatively studied with PGE2. The involvement of gastric acid in the formation of ethanol-induced necrosis was also studied. PGE2 and pirenzepine inhibited necrosis induced by all necrotizing agents at the non-antisecretory doses, and the cytoprotective effect of pirenzepine was not abolished by indomethacin. Atropine and cimetidine did not inhibit HCI-induced necrosis even at the antisecretory dose. Atropine and cimetidine at the antisecretory dose inhibited necrosis induced by ethanol, but did not inhibit the red streaks. The ethanol-induced necrosis was also inhibited by neutralizing intragastric H+ with Tris buffer. In gastric fistula rats, alkalinization of the lumen was observed by exposure to ethanol, but necrosis was not produced. There is a close relationship between the necrosis and intragastric acid. Thus it is assumed that gastric acid is involved in the formation of ethanol-induced necrosis. It was suggested that pirenzepine possesses cytoprotective action which is not related to endogenous PGs. On the other hand, the antiulcer actions of atropine and cimetidine may be due, in a part, to antisecretory effects.

Inhibition of sympathetic nervous system by mexiletine, an antiarrhythmic agent, and its antagonism against ouabain.

The effects of mexiletine on the sympathetic nervous system and the antiarrhythmic action were studied and compared with those of lidocaine. In isolated blood vessels, mexiletine inhibited the contractile responses to nicotine, tyramine and electrical transmural stimulation, but did not affect the contractile responses to exogenous norepinephrine and KCl. Lidocaine also inhibited the contractile responses to nicotine and electrical transmural stimulation, but such an inhibitory activity was weaker than that of mexiletine. Lidocaine did not affect the contraction induced by tyramine and enhanced significantly contractile responses to exogenous norepinephrine and KCl. Mexiletine and lidocaine inhibited the release of 3H-norepinephrine from the isolated rabbit pulmonary artery induced by transmural electrical stimulation. Mexiletine increased the dose of ouabain required for the occurrence of arrhythmia and cardiac arrest. These effects of mexiletine did not occur after reserpine. Though lidocaine increased the dose of ouabain required for cardiac arrest, the development of arrhythmia was not prevented. The increase in the dose of ouabain required for cardiac arrest induced by lidocaine was not affected by reserpine. On the other hand, mexiletine and lidocaine prevented the epinephrine-induced arrhythmia. These results suggested that the antagonism of mexiletine against ouabain may be due not only to the previously reported quinidine-like direct action on the myocardium, but also partially to the inhibitory action on releasing norepinephrine from the sympathetic nerve terminals.

A highly sensitive determination of individual serum bile acids using high-performance liquid chromatography with immobilized enzyme.

We have developed a highly sensitive method for the simultaneous determination of individual bile acids in serum using high-performance liquid chromatography (HPLC) combined with immobilized 3 alpha-hydroxysteroid dehydrogenase. Both the HPLC column and the immobilized-enzyme column are suitable for use with alkaline solutions necessary in working with this enzyme system. With this method we were able to determine simultaneously fifteen different serum bile acids.

Divalent cation, ATP-dependent [3H]leu-enkephalin uptake by synaptic vesicle fraction isolated from bovine caudate nucleus.

The mechanism of [3H]Leu-enkephalin uptake into synaptic vesicle fraction from bovine caudate nucleus was investigated. The simultaneous addition of 2 mM MgCl2, 2 mM CaCl2, and 2 mM ATP stimulated the uptake 22 times over the control, whereas the separate addition of these agents augmented the uptake 2.4 times at most. The addition of 2 mM MGCl2 plus 2 mM ATP and of 2 mM CaCl2 plus 2 mM ATP increased the uptake 6.6 times and 4.5 times, respectively. The cation, ATP-dependent uptake reached its half-maximal level within 10 min after the initiation of incubation at 25 degrees C, and little Leu-enkephalin was taken up at 0 degree C. The apparent Km for the uptake of [3H]Leu-enkephalin was 1.8 x 10(-7) M. Guanosine triphosphate stimulated the uptake as well as ATP, whereas CTP and ITP were only one-fourth effective of ATP. The cation, ATP-dependent uptake was inhibited by 25% and 20% in the presence of 0.1 mM colchicine and 1 microM reserpine, respectively.

A suprasellar embryonal carcinoma producing alpha-fetoprotein and human chorionic gonadotropin; treated with combined chemotherapy followed by radiotherapy.

A case of primary suprasellar embryonal carcinoma that produced alpha-fetoprotein and human chorionic gonadotropin in a 12-year-old girl is reported. Partial removal of the tumor and cis-platinum-based combined chemotherapy followed by local radiotherapy resulted in a definite regression of the tumor and improvement from clinical signs and symptoms. After these treatments, the concentrations of alpha-fetoprotein and human chorionic gonadotropin in both serum and cerebrospinal fluid decreased to undetectable levels. Serum basic fetoprotein, another tumor marker, can be a tool for evaluating the clinical course following postoperative chemotherapy.
