[Bacterial biofilms and resistance to disinfectants].

When bacteria grow in close association with solid surface, they constitute a microbial community tight included in the exopolymer glycocalyx. Many laboratory studies have shown that these bacteria are 10 to 100 folds more resistant to disinfectants than the bacteria of the same strain in suspension. Several factors are responsible for this resistance: the glycocalyx which limits the diffusion and reacts with the disinfectant, the more or less dense repartition of the bacteria inside the biofilm, their physiologic state with reduced metabolism, and the surface on which is the biofilm. The activity assessment of disinfectant agents is achieved with standardized methods. They must take into account not only the conditions in which the disinfectants are employed, but also the micro-organism state. Experimental results showing the resistance of biofilm bacteria must lead to elaborate methods allowing the assessment of bactericide activity of disinfectants against biofilm bacteria.

Docosahexaenoic and eicosapentaenoic acids inhibit in vitro human endothelial cell production of interleukin-6.

The interaction between lymphocytes, cytokines, and endothelial cells (EC) is a key step in the inflammatory process. Interleukin-6 (IL-6) a pleiotropic cytokine in its effects, seems to be an early indicator of acute systemic inflammation. In this study, we have examined the effects of polyunsaturated fatty acids (PUFAs) on the production of IL-6 by human unstimulated EC or EC stimulated with TNF-alpha (100 U/ml); IL-4 (100 U/ml); LPS (1 ug/ml); or allogeneic peripheral blood lymphocytes (PBL). Twenty-four hour culture supernatants of immunoreactive IL-6 were measured by Sandwich ELISA. We have shown that the production of IL-6 was potentiated when EC were stimulated with TNF-alpha; IL-4; LPS; or monocyte-depleted PBL in comparison to unstimulated EC. The addition of n-3 PUFAs in culture medium (100 ug/ml DHA or EPA) significantly reduces the production of IL-6 by unstimulated EC; or stimulated with TNF-alpha; IL-4 pg/ml); LPS or depleted PBL respectively for DHA and EPA, whereas the n-6 PUFAs (Arachidonic acid), even used at the highest concentration, was ineffective. This inhibitory effect is PUFA dose dependent but is more potent with EPA than DHA. Regardless of the mode of action, since IL-6 is known to be involved in hematopoiesis, in the regulation of the immune response and in the inflammatory reaction, these results suggest that n-3 PUFAs may play a role in suppressing inflammation. Further studies are needed to elucidate the mechanism involved and the choice between the two fatty acids for clinical and therapeutic purposes.

Burkholderia pseudomallei Requires Zn(sup2+) for Optimal Exoprotease Production in Chemically Defined Media.

The effects of various medium components on exoprotease production by Burkholderia pseudomallei were studied in order to understand the production of virulence factors by this pathogenic organism. FeSO(inf4), NaCl, and MgSO(inf4) do not significantly affect exoprotease activity, and CaCl(inf2) has only a slight influence. Conversely, ZnCl(inf2) plays a fundamental role since it drastically increases exoprotease production.

No major role for the dopamine D2 receptor Ser-->Cys311 mutation in schizophrenia.

A new structural polymorphism (Ser311/Cys311) in the dopamine D2 receptor (DRD2) gene has recently been reported to be associated with schizophrenia, particularly in patients with a positive family history of schizophrenia (Arinimi et al., 1994). However these findings remain controversial (Asherson et al., 1994; Nanko et al., 1994; Nöthen et al., 1994; Shaikh et al., 1994). Thus we investigated the role of the Cys311 mutation in schizophrenia using both association and family studies. First, we screened for the Cys311 mutation in 113 unrelated Caucasian schizophrenics (mean age 42 +/- 0.6; 34 females and 79 males) including 25 familial cases, and 184 unrelated controls (mean age 49 +/- 0.5, 74 females and 110 males) free of any psychiatric disorders. Diagnoses were ascertained according to DSM-III criteria (Campion et al., 1994). All patients and controls were native to the area of Rouen.

Internal architecture of the parotid gland at MR imaging: facial nerve or ductal system?

A radiologic-anatomic study was performed to determine the nature of the low-signal-intensity curvilinear structures currently seen in the normal parotid gland on axial T1-weighted magnetic resonance (MR) images. These structures are considered by some to represent the intraparotid facial nerve. After cannulation of the Stensen duct, the authors imaged the parotid gland of two cadavers in situ before and after intraductal injection of gadolinium tetrazacyclododecanetetraacetic acid. Retrograde filling was obtained in one gland. The same sections were used throughout a subsequent anatomic study, allowing correlation of the MR findings with the macroscopic and histologic appearance of the gland. Comparison of MR images and gross and histologic sections established that two intraparotid facial nerve segments, although identified from the histologic study, were not visible on corresponding MR images. Many areas of low signal intensity seen within the gland were found to represent the main duct and some afferent ductal branches.

[Biocontamination, European standardization and pharmaceutical industry].

The coming to light of new concepts in environmental microbiology, particularly the notions of bacterial adhesion and of biofilm or of cells in starvation, conducted to an evolution of the standardization of the controlled contamination area. It is necessary to match to the biocontamination of hazard area the principles governing the quality insurance used in alimentary industry named HACCP (hazard analysis of control critical point). This method will have to be applied wilfully to all the fields where a patient or a product risk a biocontamination.

[Biological safety in the laboratory. Biological risk, standardization and practice].

Working with pathogens or genetically engineered micro-organisms is a potential hazard for scientists, health care workers, employees of pharmaceutical industry, and also for the environment. Carelessness, poor technique in the handling of infectious materials, needle sting or infectious aerosol exposure are the cause of laboratory acquired infection. Biosafety, corollary of biocontamination, is based on the combination of good microbiological techniques, facility design of the laboratory and safety equipment. So, four biosafety levels are appropriate for the operations performed and the hazard posed by the infectious agents.

Pre-nucleation crystallization studies on aminoacyl-tRNA synthetases by dynamic light-scattering.

Dynamic light-scattering (DLS) studies on solutions of proteins approaching their precipitation point were made with asparaginyl- (NRSEC), leucyl- (LRSEC) and valyl- (VRSEC) tRNA synthetases from Escherichia coli. The three aminoacyl-tRNA synthetases have not been crystallized previously. As a control system, we used E. coli polypeptide elongation factor Tu (EF-Tu). Apart from the different proteins used here, the methods we employed differed from previous studies in that (1) instead of making a series of measurements on individual samples at various concentrations, the protein solutions were titrated with the precipitants, and (2) the results of the light-scattering measurements were analysed by a new maximum entropy procedure that calculates a particle size distribution in a highly reproducible way. The particle size distributions of protein solutions titrated with precipitants showed two major peaks in most cases. For both peaks, relative areas and mean diffusion coefficients were determined. The diffusion constants were corrected for the viscosity of the solutions. From comparing the results on the proteins known to crystallize (EF-Tu) with the amorphously precipitating systems (LRSEC, NRSEC) we find two necessary, but not sufficient, conditions for the formation of crystals: the diffusion coefficient of the monomer peak stays constant until very close to the precipitation point; the percentage of large aggregates stays small (less than 10% of the scattered light intensity) during the titration. For VRSEC, both ammonium sulphate and sodium citrate showed a low percentage of large aggregates and a constant diffusion coefficient of the main (protein monomer) peak below the precipitation point. This indicates that both would be possible precipitants for the crystallization of this enzyme. Crystallization trials using both these salts were carried out, and although no condition could as yet be found for obtaining crystals with ammonium sulphate solutions, crystals of the enzyme have been obtained with sodium citrate.

[Is facial nerve visible on magnetic resonance imaging?].

The internal architecture of the parotid gland was studied by nuclear magnetic resonance (NMR) imaging, particular attention being paid to the intra-parotid portion of the facial nerve. Currently available apparatuses provide high resolution images of previously poorly elucidated anatomic details. Certain authors consider that images in the axial plane in T1 allow direct visualization of the facial nerve and its branches in the form of low frequency linear signals within the gland. The present study, based on radio-anatomic correlations and date determined in a healthy volunteer, demonstrated that these structures correspond in fact to segments of the excretory canal tree, the facial nerve and its branches being invisible on NMR imaging. It is not possible, therefore, to determine the location of a parotid tumor in relation to these nerve elements using this imaging technique.

[Enteroclysis in chronic radioenteritis. Study of 43 cases].

Enteroclysis were practiced for 43 patients with radiation induced injuries of the small bowel. The radiological data revealed mucosal lesions in 23 cases, submucosa lesions in 21 cases, parietal thickening in 20 cases, mesenteric involvement in 32 cases. Narrowings and stenosis were present in 24 cases and dilated loops in 15 cases. Fistula formation existed in two cases. Twenty four patients were operated and the diagnosis of radiation enteritis was confirmed. In 12 cases, the enteroclysis succeed to a per os technique. The authors showed the superiority of the enteroclysis.

[The diagnosis of occupational tuberculosis of hospital personnel using the 2-stage intradermal Mantoux reaction].

Tuberculin skin testing was undertaken among employees of Cité de la Santé in contact with patients when the hospital was first opened in 1978. Since January 1980, two-step testing was done. The rate of the booster phenomenon was 6.9%. In well-documented BCG vaccinated personnel, almost 80% had easily interpretable reactions, either negative or significant. From our data and other data in the literature, we conclude that two-step tuberculin testing should be used if tuberculin skin testing of hospital employees is done in order to detect the booster phenomenon and not to conclude falsely in tuberculin conversion in a subsequent testing.
