Selected autoimmune diseases in the dog.

In summary, previous attempts to explain the role of inheritance in autoimmune disorders through family studies have been hampered by the variable degree of phenotypic expression. Our evidence utilizing dogs in family studies is consistent with the concept that genetic factors play some role in determining disease susceptibility. Recognizing the complexity of the genetic components involved in these studies, we have proposed a hypothesis of two classes of genes, one that relates to immunoregulation and another that specifies pathologic lesions (and thus the clinical signs). This dual system of interacting genes provides a rational explanation for many of the observations previously encountered in both human and canine studies.

Acepromazine-ketamine anesthesia in the rhesus monkey (Macaca mulatta).

Six adult male rhesus monkeys (Macaca mulatta) were anesthetized using a combination of acepromazine maleate and ketamine hydrochloride administered by intramuscular injection. This combination produced a smooth induction to anesthesia requiring less than 5 minutes. The average duration of anesthesia was slightly less than 1 hour and was safely prolonged with additional doses of ketamine. The depth of anesthesia was sufficient for minor surgical procedures and for precise radiological studies. No deleterious side effects were noted, and animals recovered completely in a short time.

Ulcerative dermatitis in the rat.

Ulcerative dermatitis observed in several shipments of rats was found to be limited to one strain (Sprague Dawley) provided by one supplier from two specific breeding areas. The incidence of lesions was higher in males than in females. Rats receiving an agar base diet also appeared to have a higher incidence of the lesions. The lesion consisted of a moist ulcer usually involving the lateral aspect of the shoulder and often the sides of the neck. Staphylococcus aureus was isolated from lesions of affected rats and from the skin of unaffected rats. Inoculation of S aureus into susceptible animals resulted in lesions in 25-40% of the rats, but usually at sites other than the site of inoculation. Serum neutralization tests for sialodacryoadenitis virus indicated that about half the rats with lesions and half the rats without lesions were positive. Toenail clipping of the hind feet of affected rats reduced the severity of the lesions. Efforts to isolate fungi and ectoparasites were unsuccessful. The selective procurement of rats from specified breeding areas, eliminated the population susceptible to or harboring the etiologic agent or agents responsible for the development of the ulcerative dermatitis described.

Cellular basis of tolerance in neonatally induced mouse chimeras.

In mice, thymectomized as young adults, neonatally induced tolerance persists in the putative absence of cell chimerism. The finding provides evidence that a selective deficiency of specific clones of lymphocytes exists in transplantation tolerance when induced under the conditions of these experiments.

Inheritance of renal amyloidosis in Chinese Shar-pei dogs.

Renal amyloidosis (RA) and recurrent fever of unknown origin (RFUO) are characteristics of familial Mediterranean fever (FMF), a human disorder inherited as an autosomal-recessive trait. Although no animal model has been established for FMF, a similar syndrome of RFUO and RA has been reported in Chinese Shar-pei (CSP) dogs. This report addresses two questions: (1) Is RA inherited in CSP dogs? (2) If it is, is it possible to hypothesize the type of inheritance involved? Two studies were conducted to answer these questions: a historical cross-sectional comparison, which included CSP and non-CSP dogs with RA; and a prospective study that included CSP dogs with RA, RFUO, or both. The cross-sectional comparison resulted in an odds ratio of 10 for RA in CSP dogs under 7 years of age. The prospective study of 28 dogs with RA or RFUO identified 20 that had RFUO and RA, three with RA alone, and five with RFUO alone. RFUO preceded RA in all cases with both conditions. The RFUO/RA combination was observed in both sexes. Four dogs with RFUO with or without RA were born to parents that either were alive at age 7 or had died because of conditions other than kidney failure/RA. When one parent was known to express one of these conditions, the prevalence of RA was between 25% and 50% among littermates.(ABSTRACT TRUNCATED AT 250 WORDS)
