Molecular characterization of the genomes of actinophages SH3, SH10, SH11, and SH12 infecting Streptomyces hygroscopicus.

Some physico-chemical properties of the DNAs released from the actinophages SH3, SH10, SH11, and SH12 are described. The four phage DNAs have a linear double-stranded secondary structure and are unique with respect to their high G.C contents which, from melting studies and buoyant density experiments, were found to be in the range of 68-73 mol-%. The DNA molecular weights were determined by sedimentation velocity experiments and by electron microscopic length measurements, the mean values of the two corresponding data sets being 34.0 x 10(6) (SH3), 26.7 x 10(6) (SH10), 26.1 x 10(6) (SH11), and 28.7 x 10(6) (SH12) with a mean relative error of +/- 5%. From different observations it was concluded that SH10 DNA, and possibly also SH11 and SH12 DNA, have cohesive ends and can undergo intramolecular or intermolecular association to form ring-like monomers or linear and ring-like multimers. Cleavage of the DNAs of SH3, SH10, SH11, and SH12 by EcoRI restriction endonuclease delivered two, one, zero, and two cleavage sites, respectively, and by BamHI restriction endonuclease eight, zero, zero, and zero cleavage sites, respectively.

[Biochemical and hematological parameters during extracorporeal heterologous perfusion with a porcine liver in acute liver failure].

The authors report on biochemical and hematologic investigations during 6 extracorporeal heterologous perfusions by means of porcine livers in 4 patients suffering from acute hepatic failure. The findings stress the functioning of porcine liver in the extracorporeal system. All livers produced bile with higher values of bilirubin and pH than found in serum. At each perfusion the serum levels of bilirubin decreased. Activities of serum enzymes (GOT, GPT, GLDH, gamma-GT, alkaline phosphatase) and the levels of potassium and sodium behaved differently. This could be explained by different damages of the porcine livers during perfusion which always occur. In 3 perfusions decreased leucocyte counts were observed, explicable immunologically. Extracorporeal heterologous perfusion by means of an alien liver is considered a possible way for temporary replacement of liver function and for removing toxins from the host. Success in single cases with reasonable indication are considered possible. The patients endured the perfusions well. After transitory improvement of their general conditions they succumbed from their severe diseases, though.

[Animal model for the study of the anhepatic state].

Hepatectomy in the pig is impossible without simultaneous removal of intrahepatic caval vein. The authors covered the vascular gap using a siliconized glass prosthesis, completed with rubber tubes. 16 animals were operated upon in this manner and survived between 6 and 34 hours. In only two cases thrombi were found in the prosthesis.

Restriction endonuclease analysis of DNA from the Streptomyces phages SH3, SH5, SH10 and SH13.

Using 26 restriction endonucleases, a cleavage site survey was undertaken for DNAs of several unrelated Streptomyces phages SH3, SH5, SH10 and SH13. Only EcoRI was found to produce single cleavage in SH3 and SH10 DNA. The complete maps were prepared for the 2, 9 and 11 fragments of SH10 DNA, as generated by EcoRI, KpnI and BglII, respectively. The evidence is presented that SH10 DNA contains cohesive ends. Moreover, a clear-plaque mutant of SH10 was shown to contain a deletion of 790 bp in the right part of the genome, including two KpnI sites.
