[Preparation and identification of polyclonal antiserum against heat shock protein 72].

OBJECTIVE: To express fusion protein of histamine (His) and human heat shock protein 72 (hHSP72) in Escherichia coli (E. coli), and to prepare hHSP72 antiserum in rabbit. METHODS: hHSP72 gene was inserted into pPROEX-1. The recombinant vector was identified by restriction endonuclease digestion analysis and sequence. Fusion protein His-hHSP72 was expressed in E. coli under isopropyl-beta-D-thiogalactoside (IPTG) induction. The rabbit antibody against His-hHSP72 was prepared by using purified His-hHSP72 protein as immunogen, and the specificity and sensitivity of polyclonal antibody were identified by Western blot. RESULTS: The restriction endonuclease digestion analysis of recombinant plasmid and sequence demonstrated that the hHSP72 gene had been exactly inserted into pPROEX-1. Sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) analysis showed that the relative molecular mass of the fusion protein was about 73 ku. Western blot result proved that the rabbit polyclonal antiserum could fuse with over 20 ng hHSP72 protein when diluted to 1:100,000. CONCLUSION: The polyclonal antibody against hHSP72 can be prepared in E. coli, it is a new reagent with high specificity and sensitivity for the research of hHSP72.

Mechanistic features for hydroxyl anion emission from the modified 12CaO.7Al2O3 surface.

OH(-), O(-), and H(-) emissions from the [Ca(24)Al(28)O(64)](4+).4(OH(-)) (defined as C12A7-OH(-)) surface were investigated by time-of-flight (TOF) spectrometry. The emission intensities were sensitive to surface temperature and extraction field. The apparent activation energies of anions decreased with the increase of applied extraction field. At an extraction field of 800 V/cm, the emission ratio of OH(-) to total anions is 0.98-0.65 in the temperature range of 870-1075 K. The OH(-) emission from C12A7-OH(-) was described by the following kinetic processes: the OH(-) anions in the cages migrated onto the sample surface by field enhanced thermal diffusion, and then desorbed to form the gas-phase anions of OH(-). The O(-) emission originated from the dissociation of O(2-) and OH(-). Similarly, H(-) emission was also attributed to the dissociation of OH(-) on the C12A7-OH(-) surface.

Diminution of toxic copper accumulation in toxic milk mice modeling Wilson disease by embryonic hepatocyte intrasplenic transplantation.

AIM: To observe the therapeutic effect of intrasplenic transplantation with embryonic hepatocytes on amelioration of hereditary copper accumulation in toxic milk (TX) mouse modeling Wilson disease. METHODS: Donor hepatocytes were harvested from 14-d fetal liver of a pregnant homogeneous DL mouse. These cells were successively cultured, labeled with fluorescein dye Hoechst 33342 for 24 h, and sequentially infused into the spleen parenchyma of the recipient TX mice. No host immunosuppression measures were taken. Two and four weeks after transplantation, the recipients were killed for routine histologic investigation and immunohistochemistry study up to 4 wk after transplantation. The serum copper and ceruloplasmin concentrations of the recipient mice were determined by graphite furnace atomic absorption spectroscopy. RESULTS: In the following 2nd and 4th wk after transplantation, the donor hepatocytes could be visualized in the livers of 47.3% recipients. The serum ceruloplasmin and copper concentrations increased by 1.6-fold after 2 wk and 2.0-fold times after 4 wk respectively, which ultimately rose from about 30% of the normal level to nearly 60% (P<0.01). The hepatic copper concentration decreased 7.2%, 4 wk after transplantation. Pathologic examination showed that there were many actively proliferative hepatocyte precursor cells with specific embryonic hepatocyte marker AFP migrated into hepatic sinusoids of the recipients. A large number of cells carrying hepatocytes marker and albumin were observed in the recipient spleen tissues. CONCLUSION: Embryonic hepatocytes are capable of differentiating into mature hepatocytes in vivo. After transplantation, the hereditary abnormalities of copper metabolism in TX mice could be corrected partially by intrasplenic transplantation of homogeneous embryonic hepatocytes.

Features and mechanism of H- anion emission from 12 CaO x 7 Al2O3 surface.

The hydrogen anion (H-) and other anionic species (O-, OH-, e-) in the gas phase, emitted from the synthesized crystal surface of 12 CaO x 7 Al2O3-H- (C12A7-H-), have been observed. The emission intensity of all the anionic species strongly depends on the sample surface temperature and the extraction field. H- has the highest emission branch ratio, and the extraction field can reduce its apparent activation energy. H- emission current at a microA/cm2-level has been achieved, which is about 4 orders of magnitude higher than that obtained from the thermal desorption process of CaH2. The observed anions of H- and OH- are attributed to their migration from the C12A7-H- cages onto the surface [i.e., Y-(cages) --> Y-(surface) --> Y-(gas phase) (Y = H, OH)]. The weak O- and electron emission would both arise from the dissociation of O2-: O2-(surface) --> O-(surface) + e-(surface) --> O-(gas phase) + e-(space).

One-step synthesis of phenol by O- and OH- emission material.

A novel approach to the direct synthesis of phenol from benzene was obtained with high benzene conversion (30%) and phenol selectivity (approximately 90%) by using a microporous material [Ca24Al28O64]4+.4O-(C12A7-O-) as catalyst with oxygen and water; active O- and OH- anions are proposed to play important roles in the formation of phenol by hydroxylating the aromatic ring of benzene.

Intranasal immunization strategy to impede pilin-mediated binding of Pseudomonas aeruginosa to airway epithelial cells.

Prevention of pulmonary Pseudomonas aeruginosa infections represents a critical unmet medical need for cystic fibrosis (CF) patients. We have examined the tenet that a mucosal immunization approach can reduce interactions of a piliated form of this opportunistic pathogen with respiratory epithelial cells. Vaccinations were performed using ntPEpilinPAK, a protein chimera composed of a nontoxic form of P. aeruginosa exotoxin A (ntPE), where the C-terminal loop amino acid sequence of the PAK strain pilin protein was inserted in place of the ntPE Ib domain. Intranasal (i.n.) immunization of BALB/c mice with ntPEpilinPAK generated both serum and saliva immune responses. A series of in vitro studies showed that diluted samples of saliva obtained from immunized mice reduced pilin-dependent P. aeruginosa binding to polarized human tracheal epithelial cells, protected human pulmonary epithelial cells from cytotoxic actions associated with bacterial challenge, and reduced exotoxin A toxicity. Overall, i.n. administration of ntPEpilinPAK induced mucosal and systemic immune responses that may be beneficial for blocking early stage adhesion and/or infection events of epithelial cell-P. aeruginosa interactions at oropharyngeal surfaces.

[Effect of the mutation of promoter region in Wilson disease ATP7B gene on the expression of reporter gene].

OBJECTIVE: To find out the relationship between mutation of ATP7B gene promoter region and pathogenesis of Wilson disease(WD). METHODS: Two of 48 WD patients presented C-->T base substitution mutations at the position -183. DNA sequences of the promoter region from normal and mutant samples were separated. The fragments containing the promoter region were cloned upstream of the luciferase. Luciferase activity was analyzed. RESULTS: The luciferase activity of reporter gene containing normal sequence of ATP7B gene promoter region did not show significant difference as compared with that of reporter gene containing mutant promoter(n=3, P > 0.05). CONCLUSION: No influence of C-->T base substitution mutations on the activity of promoter was observed in study. The results suggest that WD pathogenesis relates little to the mutations of the promoter region in Chinese.

[Redox-dependent changes in structure and function of hHSF1].

To evaluate the effects of cysteine-SH-directed regents on the redox status, structure and function of human heat shock transcription factor 1 (hHSF1), treatment in vitro of hHSF1 with 0.3 0.5 mmol/L oxidizing reagent diamide (DM) and treatment in vivo of HeLa cells with 1 mmol/L buthionine sulfoximine (BSO), an inhibitor of gamma-glutamylcysteine synthetase, promoted the formation of a compact, intramolecularly disulfide-crosslinked, stable monomeric form of ox-hHSF1, and blocked the trimerization and activation of HSF1. The effects of diamine were dose-dependent and readily could be completely reversed by adding 0.4 0.5 mmol/L reducing reagent dithiothreitol (DTT) to the samples prior to gel electrophoresis. Computer modeling of the alpha-helical coiled-coil domains of the HSF1 monomer and trimer showed that the alignment of the N- and C-terminal hydrophobic repeats of HSF1 monomer could bring C(3)(Cys(153))close to C(4) and C(5)(Cys(373) and Cys(378), respectively), in positions permissible for disulfide bond formation under appropriate experimental conditions. The results suggest that redox-dependent thiol-disulfide exchange can provide a mechanism for regulation the conformation and activity of hHSF1.
