[High efficient generation of recombinant adenovirus containing human Fas gene using a method of homologous recombination in bacteria].

OBJECTIVE: To generate recombinant adenovirus with human Fas gene and transfect the Fas gene into keloid-derived fibroblasts to take place of the dysfunctional Fas gene, reconstruct the blocked Fas signal. METHODS: We used a new bacterial homologous recombination system (Ad-Easy system) to construct recombinant adenovirus vector. Fas gene was cut down from the PMD-T-Fas plasmid and then transferred to track plasmid. Recombination was successfully completed in bacteria BJ5183 and recombinant adenovirus was produced in 293 cells. Then we infected keloid derived fibroblasts and compared the expression of Fas protein. Finally, we detected the function of newly produced Fas protein. RESULTS: We successfully constructed the recombinant adenovirus with Fas gene and detected its highly expressed Fas protein in infected keloid derived fibroblasts. Obvious apoptosis was also detected in infected keloid derived fibroblasts exposed to FasMcab. CONCLUSION: (1) The recombinant adenovirus with Fas gene can transfect the Fas gene into keloid-derived fibroblasts and highly improved the expression of Fas protein. The newly expressed Fas gene can reconstruct the blocked Fas signal. (2) The correlation between keloid and Fas gene was further proved and it may paves a sound foundation for further gene therapy in keloid.

[Effect of jiawei yupingfeng powder on T-lymphocyte subsets in patients with senile chronic bronchitis in acute onset stage].

OBJECTIVE: To explore the effect of Jiawei Yupingfeng Powder (JYP) on T-lymphocyte subsets in patients with senile chronic bronchitis in acute onset stage (SCB-AOS). METHODS: Patients were divided, according to the randomized controlled principle, into two groups, the 44 patients in the treated group and the 40 in the control group. The conventional western medicinal therapy was given to both groups, but to the treated group, JYP was administered additionally. The therapeutic course to them was 14 days. Changes of CD3, CD4, CD8 and the ratio of them were observed. RESULTS: Increase of CD3, CD4 and CD4/CD8 (P < 0.05) and decrease of CD8 (P < 0.01) were significantly shown in the treated group after treatment, but no change in the control group. Besides, comparison of the total effective rate in the two groups also showed significant difference (P < 0.05). CONCLUSION: Cellular immune function disturbance exists in patients with SCB-AOS, JYP could enhance the efficacy by way of modulating cellular immune function.

Dispersion of ventricular mRNA of RyR2 and SERCA2 associated with arrhythmogenesis in rats.

AIM: To investigate the effect of CPU86017 on the changes of mRNA abundance of different calcium handling system in infarcted heart. METHODS: Rats were subjected to left coronary ligation to induce myocardial infarction (MI). The treatment with either propranolol (Pro) 5 mg/kg ip or CPU86017 1, 2, and 4 mg/kg ip was initiated on the next day of operation and continued for 20 d. Medication with isoproterenol (Isop) 3 mg/kg sc started on the d 17-21. Ventricular mRNA abundance of ryanodine receptor 2 (RyR2), sarcoplasmic reticulum Ca(2+)-ATPase (SERCA2), L-type Ca(2+) channel, and Na(+)/Ca(2+)exchanger (NCX1) were measured. RESULTS: Arrhythmic scores (AS) in the Isop group was raised up to 5.27+/-1.75 (P<0.01) vs myocardial infarction group 2.25+/-2.04 and sham group 1.50+/-1.73. The AS was depressed by Pro (1.63+/-1.53, P<0.01 vs Isop), and CPU86017 2 and 4 mg/kg (3.00+/-1.24, and 1.70+/-1.85, P<0.01 vs Isop). The significant dispersion of depressed mRNA abundance of RyR2 and SERCA2 was associated with an increase in AS in Isop group, and it was much depressed in the left than the right ventricle. The dispersion and depression of mRNA were restored significantly by Pro and CPU86017, associated with suppression on AS. In Isop group, the mRNA abundance of L-type Ca(2+) channel was not changed; and a moderate increase in the mRNA of NCX1 was seen, the changes were regressed by Pro and CPU86017. CONCLUSION: Isop-induced arrhythmogenesis in MI heart was correlated mainly with a dispersion of depressed mRNA abundance in ventricle likely due to the consequence of PKA over-phosphorylation. A suppression of arrhythmia by Pro and CPU86017 resulted from a regression of the dispersion and depression of RyR2 and SERCA2.

[Experimental study on apoptosis of keloid-derived fibroblast transducted with Fas gene by recombinant adenovirus].

OBJECTIVE: To generate recombinant adenovirus with human Fas gene and transfect the Fas gene into keloid-derived fibroblasts to replace the dysfunctional Fas gene for reconstruction of the blocked Fas signal. METHODS: Fas gene was cut down from the PcDNA3.1-Fas plasmid. The recombinant adenovirus vector was constructed using PDC 315 system. Then the expression and function of the transfected Fas gene was detected. RESULTS: We successfully constructed the recombinant adenovirus vector with Fas gene and detected its expression and function in the infected keloid-derived fibroblasts. CONCLUSION: We have successfully generated recombinant adenovirus (Ad-PDC315-Fas), which can transfect the Fas gene into keloid-derived fibroblasis to replace the dysfunctional Fas gene and reconstruct the blocked Fas signal. The correlation between the keloid and Fas gene was confirmed. The result may set up a sound foundation for gene therapy of keloid.

[Convolution spectrometric determination of the effect of hydroxyl radical scavenger on UV radiation-induced DNA aberrance].

Convolution spectrometry was applied to determining the DNA aberrance induced by UV-A, UV-B and UV-C radiation, and the extent of minute aberrance was quantitatively expressed by the differential value of convolution spectra. The differential values of DNA solutions irradiated by UV-A, UV-B and UV-C increase in turn, indicating an increscent DNA aberrance. The addition of dimethylsulfoxide or mannitol to DNA solution decreases the differential value in respective way under UV-A, UV-B and UV-C radiation, representing different inhibitory effects. Dimethylsulfoxide decreases the UV-A-induced DNA aberrance by 60% (approximate value), UV-C-induced aberrance by 35% (approximate value), and UV-B-induced aberrance by 20% (approximate value), while mannitol shows similar inhibitory effects (40%, approximate value) for UV-A, UV-B and UV-C-induced DNA aberrance. Although the extent of DNA aberrance varies, the distribution of differential points of convolution spectra reveals that the types of UV radiation-induced DNA aberrance are similar, and the inhibitory mechanism of two hydroxyl radical scavengers is possibly the same.

Comparison of different approaches for pediatric congenital heart diseases.

To compare the clinical results of different surgical approaches for congenital heart disease in pediatric patients, 1669 cases of atrial septal defect, ventricular septal defect, or tetralogy of Fallot, which were corrected from January 1999 to December 2001, were classified according to approach (sternotomy, ministernotomy, or minithoracotomy). In cases of ventricular septal defect, the incidence of pulmonary complications was significantly higher in the minithoracotomy group than in the full sternotomy or ministernotomy groups. In patients with tetralogy of Fallot, hemoglobin concentration was higher, oxygen saturation was lower, and more patients required a transanular patch in the sternotomy group than in the other groups, but the clinical results were similar. Patients with complex defects or severe pulmonary hypertension should undergo a full sternotomy.

Immunohistochemical analysis of Fas protein expression in hypertrophic scar and keloid.

OBJECTIVE: To find an efficient diagnostic method of hypertrophic scar and keloid. METHODS: Immunohistochemistry was employed to detect the expression and distribution of Fas protein in keloids, hypertrophic scars and their surrounding normal skins. Image analysis was performed to quantitatively compare the differences of the protein expression in these tissues. RESULTS: The expression level of Fas antigen was much higher in the keloids and their surrounding normal skins than in hypertrophic scars, and no significant difference in the expression level between the former 2 tissues was found. In hypertrophic scars, however, much lower Fas protein expression was detected in comparison with that in their surrounding normal skins(P < 0.01). CONCLUSION: Immunohistochemical detection of Fas protein is a simple method that is well applicable in the differential diagnosis of pathological scars.

Graft patency in off-pump and conventional coronary artery bypass grafting for treatment of triple vessel coronary disease.

OBJECTIVE: To compare graft patency in off-pump and conventional coronary artery bypass grafting by using the transit time flow meter in the treatment of triple vessel coronary artery disease. METHODS: Between June 2000 and April 2001, 60 patients with triple vessel coronary artery disease underwent coronary artery bypass grafting. They were divided into two groups: off-pump and conventional coronary artery bypass. All completed grafts were tested intraoperatively using Transit Time Flow Measurement (TTFM). Preoperative and postoperative variables of the two groups were also compared. RESULTS: There were no significant differences in sex, age, weight, acute or remote myocardial infarction, hypertension, diabetes and type of bypass grafts between the two groups. The number of bypass grafts and the assisted respiratory time of the off-pump coronary artery bypass grafting (OPCAB) group were significantly less than those of the conventional coronary artery bypass grafting (CCABG) group. The flow and pulsatile index (PI) of the left anterior descending artery bypass grafts and the right coronary artery bypass grafts were not significantly different between the OPCAB and CCABG groups. The flow of OM in the CCABG group with the multiple anastomosis site of sequential grafts was higher than that in the OPCAB group. Diffused narrow coronary artery bypass grafts in both groups had less flow. CONCLUSION: No significant differences in graft patency were observed in patients with triple vessel coronary artery disease who had undergone OPCAB or CCABG.

[Identification of Aaron's beard with clustering analysis by attenuation reflection-Fourier transform infrared spectrometry].

OBJECTIVE: To establish a new method to discriminate Aaron's beard by Fourier transformation infrared spectrometry. METHODS: Attenuation Reflection-Fourier transformation infrared spectrometry with clustering analysis was used to the identification of Aaron's beard. RESULTS: There were obvious differences in Aaron's beard. The results are consistent with that of morphologic study. CONCLUSION: This method is rapid, simple and economical, and can be used to the quality control.

[Effect of esculentoside A on cellular adhesion].

AIM: To investigate the anti-inflammatory mechanism of esculentoside A (EsA) and to observe the effects of EsA on cellular adhesion between human umbilical vein endothelial cell (VEC304) and human neutrophil and to further observe the mRNA expression of intercellular adhesion molecule-1 (ICAM-1) and cluster of differentiation 18(CD18). METHODS: The hemocyte counting method was used for assaying the adhesion rate between VEC304 and neutrophil. The RT-PCR method was used for measuring the mRNA expression of ICAM-1 and CD18. RESULTS: The adhesion rate between VEC304 and neutrophil was increased with treatment of lipopolysaccharide(LPS). EsA (3 - 12 x 10(-6) mumol.L-1) was shown to inhibit the high cellular adhesion induced by LPS. A further investigation of adhesion molecules mRNA expression was undertaken using semi-quantitative reverse transcribed polymerase chain reaction (RT-PCR). The results of RT-PCR from VEC304 and human neutrophil treating with LPS showed that ICAM-1 and CD18 mRNA expressions were higher than those of normal cells, while this increased expression of ICAM-1 and CD18 mRNA was remarkably attenuated by the addition of EsA. CONCLUSION: EsA was found to inhibit the increased adhesion rate induced by LPS. Moreover, LPS induced high expression of ICAM-1 and CD18 was inhibited with treatment of EsA. It might be involved in the mechanisms of anti-inflammation of EsA.

[Study on the role of glucosamine hydrochloride in the pathogenesis of osteoarthritis].

The role of glucosamine hydrochloride in the pathogenesis of osteoarthritis in animal models of sponge granuloma, Kaolin arthritiaand and adjuant arthritis was studied. The results showed that glucosamine hydrochloride inhibited granulation, delayed immunoreactivity, immunological-reative arthritis in daily doses of 0.25-0.75 g/kg BW orally and vascular oozing, tissue swelling and cell isolating in daily doses of 0.5-1.5 g/kg BW orally, indicating that glucosamine hydrochloride might have a preventive and theraputic effect on osteoarthritis.

[Surgical treatment of coronary artery aneurysm].

OBJECTIVE: To describe the diagnosis and surgical management of coronary artery aneurysm. METHODS: Between October 1996 and May 2000, 6 patients with coronary artery aneurysm underwent surgical treatment. Of these patients, 3 had Kawasaki disease and 3 coronary artery fistula. All patients underwent coronary artery bypass grafting. Three patients had aneurysms resected and fistulous ostium closed. One patient received aortic valve replacement. RESULTS: There were no deaths and later death, nor major complications during the hospital stay. The results of follow-up were satisfactory. CONCLUSIONS: Coronary artery aneurysm is rare and its prognosis is poor. Early diagnosis and operation are necessary. The aneurysm should be resected, the coronary artery should be bypassed, and other associated diseases should be treated properly.

[Studies on NaNO2-induced DNA mutation by convolution spectrometry].

AIM: To study the sodium nitrite induced DNA mutation by convolution spectrometry (CS). METHODS: The spectra of sodium nitrite-induced mutative calf thymus DNA was compared with ego criteria based on Spectra of the primary DNA within the wavelength range from 200 to 340 nm. Distilled water served as blank and normal saline served as negative control. Any difference was quantitatively expressed by differential value (delta) of convolution spectra. Near-infrared spectroscopy was employed as the reference method. RESULTS: The differential value was positively correlated with the increasing time and concentration of sodium nitrite. delta values increased to 1.37%, 2.41% and 5.44% respectively within 2-hour's reaction between calf thymus DNA and 0.5, 0.05 and 0.005 microgram.mL-1 sodium nitrite, while on the contrary, changes could be hardly observed on the corresponding UV absorption spectra. The results were also confirmed by their corresponding near-infrared spectra. CONCLUSION: The delta values can be used to represent the compound's strength of mutagenesis. Every convolution procedure takes less than one minute, so CS provides a fast, simple and inexpensive alternative method to determine chemical or medicinal DNA mutation.
