Theoretical study on the mechanism of the [2+1] thermal cycloaddition between alkenes and stable singlet (phosphino)(silyl)carbenes.

The mechanism and the origins of the stereocontrol observed in the reaction between differently substituted alkenes and stable (phosphino)(silyl)carbenes giving cyclopropanes have been studied computationally. These cyclopropanation reactions proceed via asynchronous concerted mechanisms involving early transition structures with a significant charge transfer from the carbene to the alkene moiety. The geometric features of these transition structures preclude a significant overlap between the orbitals required for secondary orbital interactions between the reactants. The stereoselectivity observed experimentally stems from favorable electrostatic and steric interactions between the reactants leading to the stereoisomers in which the phosphanyl and carbonyl or aryl groups are cis to each other.

Stereoelectronic effects on type I 1,2-dyotropic rearrangements in vicinal dibromides.

The effects of different substituents on type I-dyotropic rearrangements of open-chain and cyclic 1,2-dibromo hydrocarbons have been studied by means of DFT calculations. The activation energy (E(a)) of this transformation decreases with the pi-donor ability of the substituent attached to the reacting ethylenic system. This is due to donation of electronic density by conjugation or hyperconjugation. This donation leads to longer C--C and C--Br bond lengths in the corresponding four-membered transition states (TSs). Linear relationships between the E(a) and either the sigma(p) Hammett substituent constants and the C--C bond length of the TSs were also found. In all cases, the processes have a high value of synchronicity, which is mainly independent on the substituents. A model based on the second-order perturbational analysis for one ethylene unit with two apical bromine radicals accounts for all the computed results.

Reaction of N-vinylic phosphazenes with alpha,beta-unsaturated aldehydes. Azatriene-mediated synthesis of dihydropyridines and pyridines derived from beta-amino acids.

Aza-Wittig reaction of N-vinylic phosphazenes (1,2 addition), derived from diphenylmethylphosphine or derived from trimethylphosphine with alpha,beta-unsaturated aldehydes, leads to the formation of 3-azatrienes through a [2 + 2]-cycloaddition-cycloreversion sequence. The presence of an alkyl substituent in position 3 of N-vinylic phosphazenes increases the steric interactions, and [4 + 2] periselectivity (1,4 addition) is observed. Reaction of azatrienes with alpha,beta-unsaturated aldehydes yields pyridines.

Effect of the metal fragment in the thermal cycloaddition between alkynyl metal(0) Fischer carbene complexes and nitrones.

The thermal cycloaddition between alkynyl metal(0) Fischer carbenes and nitrones has been studied computationally within the Density Functional Theory framework. It is found that the [3 + 2] cycloaddition takes place via transition structures that are more asynchronous and less aromatic than their nonorganometallic analogues. These reactions are also found to be completely regioselective in favor of the cycloadduct possessing the Fischer carbene moiety and the oxygen atom of the nitrone in a 1,3-relative disposition. These results are consistent with the role of the Fischer carbene moiety as an electron withdrawing group that enhances the electrophilic character of the alkyne group acting as a Michael acceptor as a dipolarophile. In terms of the isolobal analogy model, it can be concluded that alkynylalkoxy metal(0) carbene complexes act in this reaction as organometallic analogues of organic alkyl-propiolates with enhanced electrophilic character.

General contribution of nonspecific interactions to fluorescence intensity.

Many chemical compounds, including nonfluorescent ones, induce changes in the fluorescence spectra of certain probes, such as berberine cation and Reichardt's betaine, both in the absence and the presence of solvent, that affect almost exclusively emission intensity. In this work, the application of fluorescence detection by intensity changes (FDIC) to HPLC and TLC chromatographic systems with fluorescence detectors has been studied. FDIC detection is of special interest in detecting nonfluorescent analytes, either in HPLC or in TLC mode. It does not involve covalent interactions, and the dielectric permittivity (epsilon) of the medium plays an important role. The balance between nonspecific and specific interactions produces either an increase or a decrease in fluorescence intensity. Therefore, the influence of chromatographic conditions and chemical structure of analytes on the sign and magnitude of fluorescence peaks for sample detection in HPLC and TLC systems has been discussed. In general, probe nature and concentration determine response and detection sensitivity for a given sample in TLC and HPLC. As solubility and fluorescence properties in solvents determine the operating conditions for a FDIC probe in HPLC mode, nature and flows of mobile phase and solvent are important for chromatographic response and detection sensitivity.

Mechanism and stereoselectivity of the aza-Wittig reaction between phosphazenes and aldehydes.

The mechanism of the aza-Wittig reaction between phosphazenes and aldehydes has been studied computationally, using DFT methods (B3LYP/6-31G level), and experimentally. It has been found that the reaction consists of a tandem [2+2] cycloaddition-cycloreversion sequence in which pi and sigma orbitals as well as lone pairs are involved. Both [2+2] processes take place via thermally allowed supra-supra mechanisms. P-trimethyl-lambda(5)-phosphazenes are predicted to be more reactive than their P-triphenyl analogues. The stereochemical outcome of the whole reaction depends only on the second step, because conformational changes in the intermediate 1,3,2-lambda(5)-oxazaphosphazetidines have a much lower activation energy than the second [2+2] cycloreversion reaction. Preferential or exclusive formation of the corresponding (E)-imines is predicted.

On the affinity regulation of the metal-ion-dependent adhesion sites in integrins.

Density functional theory and a polarizable continuum model are used to (i) understand the affinity modulating mechanisms of the interaction between the metal-ion-dependent adhesion site (MIDAS) of a selected integrin, lymphocyte function-associated antigen-1 (LFA-1) and a ligand mimetic acetate molecule and to (ii) propose a new, promising family of inhibitors to block the interaction of the integrin with intercellular adhesion molecule-1 (ICAM-1). We quantify the effect of isolated factors, such as the metal coordination, the nature of the ligand or the cation present on the MIDAS, and the effect of the permittivity of the media. We show that the affinity for ligand decreases when metal coordination changes from the open conformation to the closed conformation. In addition, Mn2+ and Zn2+ showed to be good competitors for the octahedrically coordinated Mg2+ and yielded excellent affinity values, whereas Ca2+ in an octahedric environment would decrease the affinity for the ligand. Our affinity studies of the open MIDAS showed that nitronate-derived or carboxylic acid-containing ligands may represent new promising scaffolds of future inhibitors. Finally, we show that affinities are always highly favored by low-dielectric environments, which explains the propensity of MIDAS motifs to be surrounded by hydrophobic residues in integrins and highlights the importance of including hydrophobic groups in the inhibitors.

Computational and experimental studies on the mechanism of the photochemical carbonylation of group 6 Fischer carbene complexes.

The photocarbonylation reaction of Group 6 Fischer carbene complexes has been studied by DFT and experimental procedures. The process occurs by intersystem crossing (ISC) from the lowest excited singlet state (S1) to the lowest triplet state (T1), the latter structure being decisive for the outcome of the reaction. Methylenepentacarbonylchromium(0) complexes, alkoxypentacarbonylchromium(0)carbene complexes, and alkoxyphosphinetetracarbonylchromium(0) carbene complexes have coordinatively unsaturated chromacyclopropanone T1 structures with a biradical character. The evolution of the metallacyclopropanone species occurs by a jump (spin inversion) to the S(0) hypersurface by coordination of a molecule of the solvent, leading to ketene-derived products in the presence of ketenophiles or reverting to the starting carbene complex in their absence. The T1 excited states obtained from methylenephosphinetetracarbonylchromium(0) complexes and pentacarbonyltungsten(0)carbene complexes are unable to produce the carbonylation. The reaction with ketenophiles is favored in coordinating solvents, which has been tested experimentally in the reaction of alkoxypentacarbonylchromium(0) complexes and imines.

Ellipticity: a convenient tool to characterize electrocyclic reactions.

The ellipticity of the electron density is proposed as a convenient tool to characterize electrocyclic reactions. The study of the electron density offers several advantages since it is an experimental observable. This simple topological index has proven powerful and robust for the characterization of a wide variety of electrocyclic and pseudoelectrocyclic processes. This property is sensitive to changes in the anisotropy of the electron density in the bond-forming region and provides an insightful description of the events occurring along the reaction coordinate.

Substituent effects in eight-electron electrocyclic reactions.

The main features of transition structures associated with eight-electron electrocyclic reactions have been studied with Density Functional Theory. It is found that conrotatory electrocyclization reaction of (3Z,5Z)-octa-1,3,5,7-tetraenes takes place via Möbius aromatic transition structures of helical conformation. The reaction is completely periselective. In general, transition structures having outward substituents are preferred with respect to the inward transition structures, irrespective of the pi-donor or pi-acceptor character of the substituent. In contrast with four-electron thermal conrotatory electrocyclic reactions, there is no satisfactory correlation between the difference in energy of activation between inward and outward substituents and the Taft resonance sigma(R) parameter.

Quantitative evaluation of the catalytic activity of dendrimers with only one active center at the core: application to the nitroaldol (Henry) reaction.

One reference tertiary amine and three families of structurally related trialkylamines and dendrimers have been synthesized, characterized, and studied by molecular dynamics simulations. The catalytic activity of these amines in the nitroaldol (Henry) reaction between 2-nitroethanol and benzaldehyde has been measured by FT-IR spectroscopy. It is found that, in this kind of molecule with only one catalytic center at the core, the efficiency of the catalytic process decreases with the size and/or the degree of ramification of the dendrimer. According to these results, there is a linear departure from the behavior predicted by the hard sphere collision theory (HSCT) as the size of the dendrimer increases. Therefore, the behavior of structurally related dendrimers can be quantified in terms of their molecular weight and reagent accessible surfaces.

Light-induced aminocarbene to imine dyotropic rearrangement in a chromium(0) center: an unprecedented reaction pathway.

1,2-Dyotropic rearrangement can be induced by irradiation of properly functionalized Fischer carbenes. This novel reaction takes place by a stepwise mechanism and with double inversion of configuration at the static scaffold. Good yields are obtained with both cyclic and acyclic structures, thus suggesting that this unprecedented transformation can be extended to other organometallic systems.

Enhancing stereochemical diversity by means of microwave irradiation in the absence of solvent: synthesis of highly substituted nitroproline esters via 1,3-dipolar reactions.

Microwave irradiation induces the 1,3-dipolar cycloaddition of imines derived from alpha-aminoesters with beta-nitrostyrenes in the absence of solvent within 10-15 min. The reaction proceeds to give yields in the range 81-86% and three isomeric pyrrolidines are obtained in the cycloaddition. Consequently, the use of three imines and two beta-nitrostyrenes gives rise to a library of 18 nitroproline esters. The use of classical heating with longer reaction times (24 h) gives lower yields of products (below 50%) and only two stereoisomers can be detected in each reaction.

[4+3] versus [4+2] mechanisms in the dimerization of 2-boryl-1,3-butadienes. A theoretical and experimental study.

The thermal dimerization of 2-boryl-1,3-butadienes and the scope of this reaction to prepare six-membered rings difficult to synthesize by other methodologies have been studied. In addition, the nature of this dimerization has been studied theoretically. It has been found that the reaction coordinate associated with the formation of the cycloadduct of lowest energy has significant [4+3] character. This behavior is caused by the favorable carbon-carbon overlap and the large values of the corresponding resonance integrals. However, beyond the transition structure, the [4+2] pathway becomes the preferred one thus leading to the exclusive formation of the [4+2] cycloadduct. Aside from this effect, donating groups at the boryl moiety favor the [4+2] mechanism.
