RCM-mediated synthesis of trifluoromethyl-containing nitrogen heterocycles.

A ring-closing metathesis mediated pathway to trifluoromethyl-containing piperidines is detailed. This involves the development of a synthetic route to a new (trifluoromethyl)allylating reagent via a Diels-Alder/retro-Diels-Alder strategy, its application in the synthesis of a series of trifluormethyl-substituted diolefin precursors for ring-closing metathesis, and eventually the successful cyclization of these precursor molecules into the corresponding functionalized piperidines.

Chlorofluorocarbons, Methane↗

A sulfitylation-oxidation protocol for the preparation of sulfates.

A novel, high-yielding method for sulfation of alcohols has been developed, proceeding via sulfite- and sulfate diester intermediates. Sulfite diesters serve as versatile sulfate monoester precursors, allowing for transformations that are difficult or impossible with the latter compounds.

An in-depth study on ring-closing metathesis of carbohydrate-derived alpha-alkoxyacrylates: efficient syntheses of DAH, KDO, and 2-deoxy-beta-KDO.

Novel, efficient synthetic pathways to DAH, KDO, and 2-deoxy-beta-KDO are described. Ring-closing metathesis (RCM) of highly functionalized alpha-alkoxyacrylate fragments resulted in a series of synthetically versatile oxygen heterocyclic intermediates. Further functionalization of the resulting enol ether double bond and subsequent deprotection provided the natural products in high overall yields, starting from commercially available protected sugars.

Catalyst recycling via hydrogen-bonding-based affinity tags.

[Structure: see text] A novel procedure for catalyst recycling is described. Copper(I)-based catalysts, equipped with an affinity tag, are isolated from crude reaction mixtures on the basis of quadruple hydrogen-bonding interactions using a resin functionalized with complementary affinity tags. Recycled catalysts were successfully used to catalyze a tandem Sonogashira coupling/5-endo-dig cyclization and a Cu-catalyzed [3+2] Huisgen cycloaddition reaction in high yields.

Novel approach to 5-substituted proline derivatives using a silver-catalyzed cyclization as the key step.

A novel synthetic approach to the synthesis of enantiomerically pure 2,5-disubstituted pyrrolines is described. The methodology involves a Ag-catalyzed 5-endo-dig cyclization of enantiopure aryl-substituted acetylene-containing amino acids. It has also been shown that the obtained pyrrolines can be efficiently transformed into the corresponding saturated 5-aryl-substituted proline derivatives.

Expedient synthesis of triazole-linked glycosyl amino acids and peptides.

[structure: see text] An expedient, high-yielding synthesis of two types of triazole-linked glycopeptides is described. These novel and stable glycopeptide mimics were prepared via Cu(I)-catalyzed [3 + 2] cycloaddition of either azide-functionalized glycosides and acetylenic amino acids or acetylenic glycosides and azide-containing amino acids.

Efficient preparation of a 1,3-diazidocyclitol as a versatile 2-deoxystreptamine precursor.

A synthesis route toward 2-deoxystreptamine, a common structure in many of the clinically important aminoglycosides, is presented. Starting from p-benzoquinone and cyclopentadiene, 2-deoxystreptamine is synthesized with key steps involving Pd(0)-catalyzed rearrangement, a retro-Diels-Alder by flash vacuum thermolysis, and Yb(III)-directed regioselective epoxide opening. The obtained diazidocyclitol 17 is a suitable 2-deoxystreptamine precursor, conveniently protected for incorporation in new aminoglycoside entities.

Palladium-catalyzed synthesis of novel optically active tryptophan analogues.

[reaction: see text] Both unsaturated proline derivatives and optically active tryptophan analogues have been obtained via Pd-catalyzed cyclization of aniline-containing acetylenic amino acids. The side chain length of the cyclization precursor determines which one of the two possible products will be formed.
