Over-expression of hTERT in CHO K1 results in decreased apoptosis and reduced serum dependency.

The enzyme telomerase plays a crucial role in cellular proliferation. By adding hexameric repeats to the chromosome ends, it prevents telomeric loss and, thus entry into senescence of limited life span cells. It is unclear, however, what would be the effect of over-expressing telomerase in an immortalised cell line, characterised by unlimited life span and high levels of apoptosis under sub-optimal growth conditions. In order to address this question, we have transfected the immortal cell line CHO K1 with the human telomerase reverse transcriptase (hTERT) catalytic subunit. Differences in the growth profile and apoptosis levels between the cells over-expressing hTERT (Telo) and the cells containing mock vector were found under standard growth conditions. Similarly, the Telo cells showed lower levels of apoptosis, greater attachment tendency and higher viable cell density under serum-deprived conditions compared to the control cell line, suggesting a major role for hTERT over-expression in stressed cultures. Using a mouse cDNA microarray, the collagen type III and V genes were shown to have at least a 10-fold higher expression in the Telo cells than the control cells, suggesting a role of hTERT in the cell attachment pathways.

Chromatographic behavior of open-chain polyamines NH2-(CH2)2-[NH-(CH2)2]n-NH2 and their quantitative determination in sea water by high-performance ion-exchange chromatography.

The results are reported of a study using high-performance liquid chromatography for the chromatographic behavior of a homologous series of open-chain polyamines having the general molecular formula NH2-(CH2)2-[NH-(CH2)2]n-NH2 (n = 0-4). The exchange column is a carboxylate-functionalized resin. Amines are eluted with mixtures of HClO4 0.200 mol/L, NaClO4 from 0.100 to 1.40 mol/L, and acetonitrile (ACN) from 0% to 40% (v/v) and detected amperometrically. The dependence of the retention factor k' on sodium and ACN concentrations in the eluent and the total number of nitrogen atoms in the amines are considered and discussed. Suitable relationships are derived. Examples of polyamines separation are given, and a quantitative determination of analytes in spiked sea water is shown.

The retention of some open-chain diamines on a strong cation-exchange resin in ion chromatography as a function of their structure.

This work reports the results of a study of the influence of the structure of some open-chain diamines on their retention in high-performance ion chromatography (HPIC). We examine a set of eight diamines, in which the N atoms are separated by two C atoms and are differently N- and C-alkyl substituted; four of these diamines have the same formula weight. In the other two sets, the diamines are different from each other, either in terms of the length of the alkyl chain between the N atoms or in terms of the length of the substituent(s) on the nitrogen atom(s). The mobile phase is a mixture of HClO4 0.100 mol/L, 0.160 < C(NaClO4) < 1.50 mol/L, and 0 < acetonitrile% < or = 15 (v/v). The amines are detected by integrated amperometry. Interpretation of experimental data shows that (a) elution occurs for both ionic and hydrophobic interactions and is dependent on all the components of the mobile phase, (b) the diamines are eluted in any case as dications, and (c) steric hindrance significantly influences elution times. Some examples of separation are proposed.

Dioxouranium-carboxylate complexes. Formation and stability of acetate species at different ionic strengths in NaCl(aq).

The formation and stability of UO2(2+)-acetate complexes was studied potentiometrically, at T = 25 degrees C, at different ionic strength, 0 < I < or = 1 mol dm(-3), in aqueous NaCl solutions. Computer analysis allowed us to find the species UO2(ac)+, UO2(ac)2(0), UO2(ac)3- and UO2(ac)3(OH)2-. The dependence on ionic strength of formation constants was taken into account by using both a simple Debye-Hückel type equation and the SIT (Specific ion Interaction Theory) approach. A critical examination of the present results is given together with a comprehensive analysis of literature data.

The protonation of polyacrylate in seawater. Analysis of concentration effects.

The protonation of polyacrylate (PAA, MW 2 kDa) was studied potentiometrically at 25 degrees C, in mixed electrolyte aqueous solution simulating the composition of seawater, in the salinity range 30 < or = S < or = 40. The salt composition of different solutions was varied in order to study its effect on apparent protonation constants. Results were analysed using two fairly different approaches: by simple regression analysis on a combination of concentration parameters and salinity, and by canonical correlation analysis. Unexpectedly we found that variations on protonation constants, due to the different relative component concentrations, are fairly low, revealing a sort of buffering capacity of seawater respect to protonation properties of polyacrylate (and likely for other HMW and LMW polycarboxylates). The intrinsic protonation constant of polyacrylate 2 kDa at 25 degrees C and 35 salinity is log K(H*) = log K(int) = 4.399 +/- 0.004. The use of different pH scales and standard seawater compositions is also discussed.

Enhancement of hydrolysis through the formation of mixed hetero-metal species: dioxouranium(VI)-cadmium(II) mixtures.

In order to continue the investigation on the formation of hetero-metal polynuclear hydrolytic species, in this paper we report some results (at I = 0.16 mol L(-1) in NaNO3, at t = 25 degrees C by potentiometry, ISE-H+, glass electrode) on the hydrolysis of several mixtures (in different ratios) of the dioxouranium(VI) and cadmium(II) cations. The same experimental and calculation procedure of previously investigated systems was followed, and all measurements were performed by two different operators, using completely independent instruments and reagents. Many different speciation models were considered in the calculations, and a simple statistical analysis of obtained results was proposed too. UO2(2+) and Cd2+ form two hetero-metal polynuclear hydrolytic species, namely UO2Cd(OH)3+ and (UO2)2Cd(OH)4(2+), with logbeta(pqr) = -3.25 +/- 0.25 and -13.75 +/- 0.10, respectively. The formation of hetero-metal hydrolytic species is thermodynamically favored with respect to the homo-metal ones, and causes an enhancement of the percentage of hydrolyzed metal cations; comparisons with previously studied systems reveal that the hydrolytic behavior of UO2(2+)/Cd2+ mixtures is more similar to that observed for UO2(2+)/Cu2+ than for UO2(2+)/(C2H5)2Sn2+, and the tendency to form hetero polynuclear hydrolytic species with dioxouranium(VI) by other cations follows the trend (C2H5)2Sn2+ > Cu2+ > or = Cd2+.

Dioxouranium(VI)--carboxylate complexes. Interaction with dicarboxylic acids in aqueous solution: speciation and structure.

In this paper we report the results of an investigation performed by potentiometric (H+-glass electrode) and visible spectrophotometric measurements on the interaction of UO2(2+) ion towards some carboxylic ligands (acetate, malonate, succinate, azelate). The measurements were carried out at T= 25 degrees C in different ionic media (KNO3 and NaCl) at different ionic strengths (0.1 < or = I/mol L(-1) < or = 1.0, NaCl; I/mol L(-1) = 0.1, KNO3). The dependence on ionic strength of formation constants was taken into account by using both a simple Debye-Hückel type equation and the SIT (Specific ion Interaction Theory) approach. Different speciation models (depending on concentration of reagents, ionic strength, pH-range) both for different carboxylates and different ionic media have been obtained. Linear combinations between formation constants, stoichiometric coefficients and length of alkyl chain of dicarboxylates have been observed and predicted formation constants at I= 0 mol L(-1) are reported for the interaction of UO2(2+) with HOOC-(CH2)n-COOH with 1 < or = n < or = 7. Finally, a visible absorption spectrum for each complex reaching a significant percentage of formation in solution (KNO3 medium) has been calculated to characterise the compounds found by pH-metric refinement.
