Asymmetric heterogeneous catalysis.

Limited natural resources and an increasing demand for enantiomerically pure compounds render catalysis and especially heterogeneous asymmetric catalysis a key technology. The field has rapidly advanced from the initial use of chiral biopolymers, such as silk, as a support for metal catalysts to the modern research areas. Mesoporous supports, noncovalent immobilization, metal-organic catalysts, chiral modifiers: many areas are rapidly evolving. This Review shows that these catalysts have more to them than facile separation or recycling. Better activities and selectivities can be obtained than with the homogeneous catalyst and novel, efficient reaction mechanisms can be employed. Especially fascinating is the outlook for highly ordered metal-organic catalysts that might allow a rational design, synthesis, and the unequivocal structural characterization to give tailor-made catalysts.

Asymmetric hydrogenation of aromatic compounds.

The asymmetric hydrogenation of aromatic and heteroaromatic compounds arguably presents one of the most attractive methods for the synthesis of six-membered cyclic compounds. In recent years, a number of promising stereoselective methods for the asymmetric hydrogenation of pyridines and related heterocycles have been reported.

Complex aldol reactions for the construction of dense polyol stereoarrays: synthesis of the C33-C36 region of aflastatin A.

[reaction: see text]. Facial selectivity in the addition of boron enolates of alpha-oxygenated ketones to anti-disposed alpha,beta-bisalkoxy aldehydes is controlled by the aldehyde vicinal diol protecting group. Protection of the diol as an acetonide results in the exclusive formation of the anti-syn-anti stereoarray found in the C(33)-C(36) region of aflastatin A.

Sterically demanding, bioxazoline-derived N-heterocyclic carbene ligands with restricted flexibility for catalysis.

A unique family of N-heterocyclic carbenes derived from bioxazolines (IBiox) suitable for application in transition-metal catalysis is described. The ligands are electron rich, sterically demanding, and have restricted flexibility. Their usefulness has been demonstrated in the Suzuki-Miyaura cross-coupling of sterically hindered aryl chlorides and boronic acids. For the first time, tetraortho-substituted biaryls with methyl and larger ortho-substituents have been synthesized from aryl chlorides using the Suzuki-Miyaura method.

Catalysis-based enantioselective total synthesis of the macrocyclic spermidine alkaloid isooncinotine.

A concise and efficient total synthesis of the spermidine alkaloid (-)-isooncinotine (1) incorporating a 22-membered lactam ring is outlined. The approach is largely catalysis-based, involving a selective iron-catalyzed alkyl-aryl cross-coupling reaction of a difunctionalized pyridine substrate, a heterogeneous asymmetric hydrogenation step to set the chiral center of the target, and a highly integrated ring-closing metathesis/hydrogenation sequence to forge the saturated macrocyclic edifice in a single operation.
