Bioactive galactose-branched polyelectrolyte multilayers and microcapsules: self-assembly, characterization, and biospecific lectin adsorption.

We describe the fabrication of multilayers and microcapsules with biologically designed targeting activity using chemoenzymatic synthesized carbohydrate-branched polyelectrolytes. A novel cationic d-galactose-branched copolymer [poly(vinyl galactose ester-co-methacryloxyethyl trimethylammonium chloride), PGEDMC] is alternated with poly(styrene sulfonate) (PSS) to form thin multifilms by the layer-by-layer (LbL) technique on such different solid surfaces as quartz slides, poly(ethylene terephthalate) (PET) films, silicon wafers, and polystyrene (PS) microparticles. The experimental protocols were first optimized on flat, smooth silica substrates using UV-vis, contact angle, and atomic force microscopy (AFM) measurements. The film properties of PGEDMC/PSS multilayers are modified by varying polyelectrolyte concentration, ionic strength, and counteranion types. Hollow capsules were formed after the removal of colloidal templates; transmission (TEM) and scanning (SEM) electron microscopy were used to verify the LbL process integrity. PGEDMC/PSS planar films and capsules carrying beta-galactose as recognition signals have specific recognition abilities with peanut agglutinin (PNA) lectin rather than concanavalin A (Con A) lectin observed by fluorescence spectroscopy.

Cloning, chromosome mapping and expression analysis of an R2R3-MYB gene under-expressed in maize hybrid.

R2R3-MYB transcription factors play important role in transcriptional controls during higher plant metabolism and development. In this study, an R2R3-MYB gene was isolated from maize according to an EST, which expressed differentially between a hybrid and the two parents on a cDNA chip. The full-length cDNA, designated by ZmMYBL1 (GenBank accession no. AY365033) consists of 1417 nucleotides and contains an open reading frame of 828 bp. The deduced amino acid sequence contained two conserved MYB domains near its N-terminus, a conserved E1 motif and an acidic Ser/Thr rich region toward its C-terminus. Southern blot analysis revealed ZmMYBL1 could be a single copy gene belonging to a multi-gene family in the maize genome. Expression analysis showed ZmMYBL1 transcripts accumulated in various tissues examined, with strong level in tassel and weak level in leaf. Also it was under-expressed in root, stem, and leaf of hybrid as compared with that of the two parents. ZmMYBL1 was mapped on maize chromosome bin7.03 between two SSR markers, bn1g339 and umc1865 using Yuyu22 recombinant inbred line population. A QTL for root average diameter in maize seedlings was also localized on the corresponding region of chromosome 7 within the interval ZmMYBLI-bnIg1805. A possible role of ZmMYBL1 and its relation to maize heterosis were discussed based on these results.

[Observation the electroporation of blood cells in vitro by scanning electron microscope].

OBJECTIVE: To observe changes on cell membrane in blood cells after they were been electrified. METHODS: Blood were electrified for 5, 10, 20, 30 s, 1 min respectively, and Scanning electron microscope was used to detect the changes on their cell membranes. RESULTS: Pores were detected both on electrified erythrocytes and leukocytes with round or ellipse shapes. The erythrocytes often have one or more pores while the leukocytes often have more pores looked like cribble. The rates of perforated cells were increased with the prolonging time of electrification, 5 s with 6% and 1 min increased to 40%. CONCLUSIONS: Alternating current can cause the cell perforating, and the rates of perforated cell were increased with the prolonging time of electrification.

Reactive coupling of poly(ethylene glycol) on electroactive polyaniline films for reduction in protein adsorption and platelet adhesion.

Poly(ethylene glycol) (PEG)-coupled polyaniline (PANI) film surfaces were prepared by incorporating the chlorinie end-capped methoxy PEG (mPEGCl) of molecular weight of about 2000 onto the emeraldine (EM) base form of PANI via N-alkylation. The microstructure and composition of the mPEG-coupled PANI (mPEG-c-PANI) surfaces were characterized by atomic force microscopy, contact angle measurement and X-ray photoelectron spectroscopy. The concentration of surface-coupled mPEG increased with the increase in concentration of the mPEGCl solution. The mPEG-c-PANI film surfaces exhibited enhanced ability to repel protein adsorption, with only an moderate reduction in their electrical conductivity. The mPEG-c-PANI surface with a high concentration of coupled mPEG also exhibited good resistance towards platelet adhesion.
