Dioxins and related compounds in albatrosses from the Torishima Island, Japan: accumulation features by growth stage and toxicological implications.

Concentrations of dioxins and related compounds (DRCs), such as polychlorinated dibenzo-p-dioxins (PCDDs), polychlorinated dibenzofurans (PCDFs), and coplanar polychlorinated biphenyls (Co-PCBs), were determined in eggs, nestlings, and adults of black-footed albatross (BfA; Diomedea nigripes) and short-tailed albatross (StA; Diomedea albatrus) collected from the Torishima Island in Japan during 2002, which is one of their breeding grounds. Observed DRC concentrations, especially PCDFs and non-ortho Co-PCBs, in eggs of BfA and StA were relatively higher than those in other oceanic, coastal, and inland wild birds reported recently and were comparable to those in a pooled BfA egg from Midway Atoll in 1993, implying continuous input of these contaminants into open ocean and possible albatross-specific exposure. Concentrations of PCDDs except 12378-P5CDD and high-chlorinated PCDFs in 3-month-old BfA nestling were lowerthan those in 1-month-old nestling, indicating their developmental dilution. On the other hand, higher levels of T4-H6CDFs and Co-PCBs, especially low-chlorinated congeners such as 2378-T4CDF and T4CB77, were found in 3-month-old BfA nestling, suggesting specific exposure to these contaminants, possibly due to their higher transportability than high-chlorinated congeners. Estimated biomagnification factors of almost all the congeners in adults were apparently greater than those in nestlings, except 2378-T4CDF, T4CB77, and H7-O8-CDD/Fs. This could be due to preferential metabolism of 2378-T4CDF and T4CB77 and lower uptake efficiency of high-chlorinated congeners through the gastrointestinal tract in adults. Toxic equivalents in BfA and StA eggs estimated using WHO-avian toxic equivalency factors exceeded some toxicity thresholds for avian embryos, indicating possible adverse effects of DRCs to albatross embryos.

First detection of Rickettsia in soft-bodied ticks associated with seabirds, Japan.

Rickettsia was first detected in seabird soft-bodied ticks, Carios capensis and C. sawaii in Japan. According to sequence analysis, Rickettsia in Japan was identical to Rickettsia scc31 in C. capensis in the U.S.A. This suggested that an environmental circulation had consisted among microorganisms, ticks and long distance migratory seabirds around the Pacific Ocean.

First record of Leptospira borgpetersenii isolation in the Amami Islands, Japan.

In 2003, a Leptospira survey was performed on Yoroshima Island of the Amami Islands located in the southwestern part of Japan. Seven Leptospira strains were isolated from the field rat Rattus rattus, which were identified as L. borgpetersenii by flaB sequencing, 16S rDNA sequencing and gyrB sequencing, and serovar Javanica was determined by a microscopic agglutination test. NotI-long restriction fragment analysis indicated that these isolates were genetically indistinguishable from an isolate from the Okinawa Islands. The present results suggest that L. borgpetersenii is migrating into the Amami Islands in Japan.

Non-destructive monitoring of trace element levels in short-tailed albatrosses (Phoebastria albatrus) and black-footed albatrosses (Phoebastria nigripes) from Torishima Island, Japan using eggs and blood.

Concentrations of 18 trace elements (V, Cr, Mn, Co, Cu, Zn, Se, Rb, Sr, Mo, Ag, Cd, Sb, Cs, Ba, Hg, Tl and Pb) were determined in abandoned eggs of short-tailed albatrosses and abandoned eggs and blood of black-footed albatrosses from Torishima Island, Japan in 2002. Mercury concentration was highest among the toxic elements in egg content of both the two species. In some egg contents of black-footed albatrosses, Hg concentrations were higher than the threshold level that may cause decreased hatchability and aberrant nesting behavior in some avian species. Concentrations of Cd in egg content and Ba in egg content and shell increased with an increase in egg breadth/length ratio, whereas Hg in egg content showed an opposite trend in black-footed albatrosses. Since egg breadth/length ratio is known to increase with the age of mother bird, this result may imply that concentrations of these elements in eggs vary with age of mother bird. In blood of black-footed albatross chicks, concentration of Hg was highest among the toxic elements and the Hg concentration showed an increase in the later growth stages. In contrast, concentration of Sr in blood decreased with growth stage. Since the behaviour of Sr is similar to that of Ca in animals, it is conceivable that Sr was used along with Ca for the formation of bone.

Perfluoropentacene: high-performance p-n junctions and complementary circuits with pentacene.

We report the synthesis and characterization of perfluoropentacene as an n-type semiconductor for organic field-effect transistors (OFETs). Perfluoropentacene is a planar and crystalline material that adopts a herringbone structure as observed for pentacene. OFETs with perfluoropentacene were constructed using top-contact geometry, and an electron mobility of 0.11 cm2 V-1 s-1 was observed. Bipolar OFETs with perfluoropentacene and pentacene function at both negative and positive gate voltages. The improved p-n junctions are probably due to the similar d-spacings of both acenes. Complementary inverter circuits were fabricated, and the transfer characteristics exhibit a sharp inversion of the output signal with a high-voltage gain.

High-definition imaging system based on spatial light modulators with light-scattering mode.

We have developed a prototype high-definition imaging system using polymer-dispersed liquid-crystal (PDLC) light valves, which can modulate unpolarized light with high spatial resolution and exhibit a high optical efficiency, based on the light-scattering effect. We fabricated high-definition light valves with a fine polymer-matrix structure in a PDLC film by controlling the curing conditions used during the photopolymerization-induced phase separation and formation process. This device has excellent characteristics, such as a high resolution, with 50 lp/mm for a limiting resolution and greater than 20 lp/mm at the 50% modulation transfer function point, and a reflectivity of greater than 60%. An optically addressable full-color projection display was designed, consisting of three PDLC light valves, a schlieren optical system based on shift-decentralization optics with a xenon lamp illumination and input-image sources with 1.5 million pixels, including electrical image compensation of the gamma characteristics. We succeeded in displaying pictures on a 110-inch screen with a resolution of 810 TV lines and a luminous flux of 1900-2100 American National Standards Institute lumens.

Nucleotide sequence of equine erythropoietin and characterization of region-specific antibodies.

OBJECTIVE: To determine the full-length complementary DNA (cDNA) sequence of equine erythropoietin (EPO) and to develop region-specific antibodies to differentiate equine EPO (eEPO) and human EPO (hEPO). SAMPLE POPULATION: RNA and lysate extracted from renal tissues of an adult Thoroughbred. PROCEDURE: Full-length cDNA was determined by use of a reverse transcriptase-polymerase chain reaction assay and a rapid amplification of cDNA ends method. The deduced amino acid sequence was compared with sequences of EPO reported for other species. Furthermore, 4 synthetic peptides were designed in 2 distinctive parts of the eEPO and hEPO amino acid sequences to obtain antibodies specific for eEPO and hEPO. Specificity of the antibodies was tested against supernatant of homogenized equine kidney and recombinant hEPO (rhEPO) by use of western immunoblotting techniques. RESULTS: Analysis of the 1,181 bp in the nucleotide sequence revealed that eEPO was a residue of 192 amino acids. Similarity of eEPO with amino acid sequences of EPO from other species was 81.0% to 90.6%. Antibodies were specifically recognized by eEPO or rhEPO molecules. Anti-hEPO (161 to 165) antibody specifically recognized rhEPO. In contrast, anti-eEPO (133 to 144) antibody reacted with the equine kidney lysate. CONCLUSIONS AND CLINICAL RELEVANCE: We determined the cDNA and amino acid sequence of eEPO and developed region-specific antibodies that specifically recognized eEPO or rhEPO. These antibodies may be useful in distinguishing rhEPO from eEPO in a test to detect the misuse of rhEPO in racehorses.

Molecular cloning, nucleotide sequence and presence of multiple functional polyadenylation signals in the 3'-untranslated region of equine dopamine beta-hydroxylase cDNA.

Complementary DNA (cDNA) encoding equine dopamine beta-hydroxylase (DBH) was amplified with a combination of reverse transcription-polymerase chain reaction (RT-PCR) and rapid amplification of cDNA ends (RACE) method, and their nucleotide sequences (Accession No. AB029430: the DDBJ nucleotide sequence database) was determined. A total of 3842 bp cDNA sequence was consisted with 5 bp of 5' flanking untranslated sequence, 1833 bp of open reading frame encoding 610 amino acids, and 2004 bp of 3' flanking untranslated sequence. The deduced amino acid sequence of equine DBH was very similar to the known mammalian DBH sequences. The similarity between amino acid sequence of equine DBH to sequences of bovine, human, rat and mouse DBH were 86.3, 84.6, 82.2 and 81.2%, respectively. Northern blot analysis and in situ hybridization revealed three different sizes of mRNA expressions in equine adrenal medulla tissue. Then we found three putative polyadenylation signal sites in the 3' flanking untranslated sequence. These results indicate that alternative use of three polyadenylation sites generates the equine DBH mRNA that have different sizes of 3' flanking untranslated region. These results may provide further evidence for understanding DBH molecule and clues for the equine DBH gene analysis.

Immunohistochemical localization of chromogranin a in the acinar cells of equine salivary glands contrasts with rodent glands.

We investigated the existence of chromogranin A (CgA) in salivary glands of the horse by Western blotting and enzyme immunoassay (EIA) using an antiserum against a peptide sequence of equine CgA. We also compared its cellular distribution between the horse and rat salivary glands with a tyramide signal amplification immunofluorescence technique. Western blotting gave three significant immunoreactive bands (74, 56 and 48 kDa) in adrenal medulla and three major salivary glands of horses. Immunoreactivities for CgA measured by EIA in horses were 154.05 +/- 41.46, 20.32 +/- 5.59 and 4.43 +/- 2.23 pmol/g wet weight in the parotid gland, submandibular gland and sublingual gland, respectively, and 1.03 +/- 0.407 pmol/mg protein in the saliva. Immunohistochemically, the positive reactivity was mainly recognized at acinar cells in equine salivary glands. This exhibits a contrast to the finding in the rat salivary glands that the CgA immunoreactivity is localized at the duct cells of the submandibular gland. These results provide novel evidence that in the horse, CgA is stored in the acinar cells of salivary glands, and secreted into saliva.

Familial aggregation of uterine myomas in Japanese women.

To assess the familial aggregation of uterine myomas in Japanese women with myomas, one hundred forty four women requiring surgery for myomas and 288 age-matched healthy controls were studied in Hokkaido, Japan. The incidence of positive first-degree family history of myomas among women aged 45-54 years with myomas was greater than that among controls (31.5% versus 15.2%, respectively, p < 0.01). Analyses categorized by the status of parity and familiality among subjects showed that the risk for myomas was the greatest in women who had both fewer births (parity = 0 or 1) and the positive family history of myomas as compared with those who had both more births (parity > or = 2) and the negative familiality of myomas (odds ratio = 5.8, 95% confidence interval = 2.3 - 14.6). The results of this study suggest that Japanese middle-aged women with myomas have the familial predisposition of uterine myomas. Furthermore, nulliparous women with the familial aggregation of myomas may be at increased risk of the disease.
