Electron microscopic evidence of persistent chlamydial infection following treatment.

Chlamydia trachomatis infections of the female and male genital tracts are often asymptomatic and, thus, tend to become persistent. In the persistent state the typical Chlamydia life cycle is arrested and standard antibiotic regimens do not always eradicate this infection. We sought to relate treatment failures in men and women with persistent chlamydial genital tract infections to electron microscopic evidence of chlamydial persistence and with atypical morphological forms of the organism. Of 16 patients with chlamydial persistence following azithromycin treatment, morphological variants of this organism were observed by electron microscopy from one endocervical sample and one male urethral sample. We document the presence of intracellular inclusions containing only reticulate bodies, extracellular monomembrane and polymembrane phagosomes containing elementary bodies and reticulate bodies with abnormal outer membranes in the process of dividing extracellularly. These observations parallel previous in vitro studies of chlamydial persistence under adverse conditions. This capacity of C. trachomatis to undergo atypical morphological alterations in vivo may contribute to its persistence and relative resistance to antibiotics.

Modern methods for diagnosis of Gardnerella infection.

Comparison of morphological, bacteriological, serological, and genetic methods for diagnosis of Gardnerella infection in vaginosis showed that the first three methods are preferable, while the genetic method (polymerase chain reaction) is more efficient for identification of Gardnerella vaginalis in culture.

[The problem of diagnosis and treatment of urogenital chlamydiosis in Russia].

The problems associated with the distribution, diagnosis, classification and treatment of urogenital tract chlamydiosis in Russia are discussed. Some arrangements for the improvement of the activities of the laboratory diagnostic services, the use of the International Classification of urogenital tract chlamydioses and the treatment optimization are offered. The drug of choice in the treatment of urogenital tract chlamydiosis is azithromycin (Sumamed, Pliva). Doxycycline, erythromycin and ofloxacin are recommended as the reserve drugs.

[Antimicrobial and antitrichomonal activity of 5-nitroimidazole derivatives in an in vitro experiment].

The comparative study on the antimicrobial activity of metronidazole and its complex with cobalt sulfate showed that the complex formation increased 2-4 times the metronidazole activity against elective anaerobes. The study of the drug action on Trichomonas spp. revealed that as a result of the complexing with cobalt sulfate the metronidazole efficacy against the Trichomonas strains isolated from the discharge of a female patient with metronidazole resistant trichomoniasis increased, the activating effect of the complex on the host cells being suggested.

Adenylate cyclase activity of cultural strains of Neisseria gonorrhoeae and Staphylococcus aureus.

Adenylate cyclase of gonococci and staphylococci from patients with gonorrhoeae and pyoderma was studied ultracytochemically. Adenylate cyclase activity was mainly revealed on the inner side of the plasma membrane and septa of dividing cells. The enzymic activity is markedly higher in penicillinase producing strains and dividing bacteria. These data may be useful for the diagnostics and treatments of the infections concerned.

[The ultrastructure of anaerobes isolated from the urogenital tract of women].

Electron microscopic (ultrathin sections) examination of culture strains of vaginal Gardnerella and bacteroids isolated in aerobic conditions from urogenital tract of women suffering from Gardnerella infection was carried out. Structural analysis of the bacteria is presented, i.e. their shape, size, morphology, and features of multiplication.

[The effect of intensified lipid peroxidation in patients with Devergie's lichen ruber pilaris on adenylate cyclase activity].

Studies with the use of Fe2(+)-induced chemiluminescence of apo-beta-lipoproteins and electron histochemistry have revealed that PUVA therapy and vitamin A high doses enhance lipid peroxidation and activate adenylate cyclase in patients with Devergie's disease. This finding permits a conclusion on the usefulness of enhancing lipid peroxidation in order to reduce cellular mitotic activity.

[Chronic pyoderma caused by beta-lactamase-producing strains of Staphylococcus].

Features of the agent structure have been revealed on a subcellular level in vitro and in vivo in the patients with chronic pyodermas induced by Staphylococcus beta-lactamase-producing strains, and the type of these strains interaction with phagocytes before and in the course of combined therapy detected. Regularities in morphologic manifestations of pyoderma development were singled out, explained by specific features of cellular exudate composition and interactions between bacteria and eukaryotes.

[Structuro-functional properties of staphylococci in patients with pyoderma].

Electron-microscopic and ultra-cytochemical studies have shown the structural and functional features of pyoderma agents--staphylococci isolated from patients with acute and chronic pyoderma. The studies have revealed magnesium-activated ATPase and adenylate cyclase in these bacteria; the authors have tried to trace a correlation between these enzymes' activities and location and the disease pattern. This may help assess the bacterial population status and choose the drugs for purposeful pathogenetic therapy.

[An ultracytochemical study of the hydrogenosomes of Trichomonas vaginalis].

The ultracytochemical study of cultural T. vaginalis has been carried out. The activity of magnesium-dependent ATPase, transport (sodium-potassium-dependent) ATPase and adenylate cyclase was detected in dense corpuscles, located on the membranes of hydrogenosomes having a rounded form. The authors suggest that the dense corpuscles are the zones of the active transport of ions through the membranes of hydrogenosomes.

[Ultracytochemistry of phosphohydrolase of the microorganisms and phagocytes in gonococcal infection].

The complex study of phosphohydrolases (Mg2+ATPase, Na+,K+ATPase) and adenylate cyclase in gonococci, both in vivo and in vitro, and in phagocytes in gonococcal infection has been carried out with the use of ultracytochemical techniques. The enzymatic activity in the cellular structures of micro- and macroorganisms under study has been visualized. The prospects of the ultracytochemical approach to the study of the enzymes of nucleotide metabolism, their role in the life of eu- and prokaryotes, as well as the ways for affecting the enzymatic systems of cells are discussed.

[Cultivation of the gonococcus in nutrient broths].

The dynamics of the multiplication of gonococci and the parameters of their growth have been studied in the process of batch cultivation in liquid culture media with different content of bovine blood serum. The protective and stimulating action of the serum on the growth of gonococci has been shown. The optimization of the process of cultivation has been carried out; as a result, the growth of test strains in a culture medium containing no serum has been achieved. Phasic changes in the ultrastructure of gonococci in the process of their cultivation in liquid media have been followed. 9- to 12-hour cultures of gonococci grown in liquid culture media have been found the most valuable and physiologically active.

[Differences in the ultrastructure of colored variants of colonies of pathogenic Neisseria].

Electron-transparent zones (vacuoles) of different size and localization have been detected in the cytoplasm of meningococci forming orange colonies. These vacuoles have been detected in cells from meningococcal cultures of different age, grown both in liquid and solid culture media. This phenomenon requires further deeper study. The presence of a relationship between newly detected vacuoles and the pathogenicity of meningococci is suggested.
