Inhibitory effect of peptide vilon on the development of induced rat urinary bladder tumors in rats.

The effect of peptide vilon (Lys-Glu) on urinary bladder carcinogenesis in rats was studied. Urinary bladder tumors were induced with a selective carcinogen N-butyl-N-(4-hydroxybutyl)nitrosamine. The tumors developed in 56% vilon-treated animals and in 75.5% controls. Vilon 2-fold decreased the incidence of preneoplastic and early neoplastic changes in urinary bladder mucosa and significantly inhibited carcinogenesis.

[Effect of olipifate on the wound healing proces and relapse after resection of Pliss lymphosarcoma].

The investigation was concerned with the influence of preliminary injections of Olipiphat, irrigation of operative wound and combination of both procedures on healing and relapse processes, following resection of Pliss lymphosarcoma at different stages after transplantation into rats. The physical condition of the animals after tumor resection on days 12, 10 or 7 of tumor growth was better than in controls, as a result of irrigation of the operative wound with Olipiphat or in combination with preliminary injections of the drug: they came out from anesthesia quicker, tidied themselves up and moved about the cage. Irrigation of the wound with Olipiphat or in combination with preliminary injections followed by longer survival after surgery performed at all stages of tumor growth. Moreover, one animal out of 16 in each of the 4 Olipiphat-treated groups survived 60 days recurrence-free. The drug proved more effective in stimulating the healing of larger wounds but contributed to healing by first intention in all cases.

[Effect of vilon and epithalone on induction and growth of induced bladder neoplasms in rats].

Experimental data on the effect of peptides--Vilon (Lys-Glu) and Epitalon (Ala-Glu-Asp-Gly)--on induction of urinary bladder tumors in rats are presented. Treatment with Vilon was followed by a significant fall in tumor incidence in 56% of experimental animals, as compared with 75.5% in control, as well as inhibition of early-onset neoplastic changes in the bladder mucosa. No inhibitory effect of Epitalon was recorded.

[Methodological approaches to the study of the carcinogenic properties of substances].

Since the methods of investigation of carcinogenicity of different agents have changed drastically, relevant manuals need to be revised and supplemented. A new concept of carcinogenic agents evaluation, criteria for their selection and study are discussed. Working out of criteria for establishing priorities of hazardous substance selection is of vital importance.

[Experimental investigation of 2,2'-diamino-1,1'-dinaphthyl in rats and mice].

Carcinogenicity of 2,2'-diamino-1,1'-dinaphthyl has been investigated in chronic experiments on LIO rats and CC57 mice. Single doses of 20 mg were given to rats and 1-5 mg--mice. Subcutaneous injections were followed by sarcoma development on injection site in 90% of male rats and 31%--females. Female rats (46%) also revealed multiple benign tumors of the mammary gland. Such tumors alone were detected in females (44%) receiving the agent with feed. All mouse males and 71-100% of females developed hepato- and cholangiocellular tumors as well as hepatic hemangiomas following subcutaneous injection, oral or skin administration. 2,2'-diamino-1,1'-dinaphthyl proved a potent carcinogenic agent; it is suggested that it might metabolize to such carcinogen as 2-naphthylamine.

[Tissue distribution of orthotolidine and pathways of its elimination].

Distribution of the carcinogenic agent orthotolidine (OT) in the rat's body has been investigated by diazotizing technique. Levels of free amines were 1.5-2 times those in control 3 days after the last administration (total treatment duration-8 months). Zymbal gland, the target tissue, showed the highest concentrations. Also, bound amine concentrations in Zymbal gland soared up (8 times those in control). Similar results were reported 7 days after the last administration of OT: free amine level in the same site was 7 times that in control. Enhanced free and bound amine concentrations may point to its being withdrawn through Zymbal gland ducts. This may also promote carcinogenesis in them.

[A model of rat's urinary bladder tumor induced by subcutaneous injection of N-butyl-N-(4-hydroxybutyl)-nitrosamine].

A model of urinary bladder tumor was developed in the rat by subcutaneous injection of N-dibutyl-N-(4-hydroxybutyl) nitrosamine. A direct dose-tumor frequency correlation was established. Maximum effect (100%) was recorded with administration of 70 mg of the carcinogenic agent twice a week, for 3 months. Among neoplasms induced were transitional-cell and in situ carcinoma, and multiple papilloma of the bladder. Carcinogenesis was found to involve a significant rise in the activity of urinary beta-gluco-uronidase which may be used as a diagnostic testing procedure.

[A new way of preventing urinary bladder tumors].

An experimental study of urinary bladder tumor in rats has demonstrated that their development is suppressed and latency period is increased as a result of a prolonged inhibition of a urine enzyme, beta-glucuronidase by treatment with poglucar.

[New experimental oncologic studies on prevention of bladder cancer].

Experimental data connected with prophylaxis of vesical cancer are reviewed. The following prophylactic measures are discussed: influence on metabolism of carcinogens and inactivation of metabolites; forced clearance of carcinogenic metabolites; carcinogenesis promoters arrest. Specific inhibition of beta-glucuronidase in urine and forced diuresis appeared to be extremely effective.

[The lithium carbonate promotion of urothelial tumors in rats induced with N-butyl-N-(4-hydroxybutyl)-nitrosamine].

The effect of lithium carbonate on promotion of N-butyl-N-(4-hydroxybutyl)-nitrosamine-induced bladder tumors in rats was studied. Lithium carbonate acted as promoter in bladder carcinogenesis resulting in a 6-fold increase in the tumor occurrence as compared to controls. The experiments with sequential injection of the agent showed its promoting effect to be most apparent within 3-6 months following exposure to the carcinogen.

[The carcinogenic and anticarcinogenic properties of neozon D].

Continuous administration of neozone D to dogs and rats failed to reveal its carcinogenicity. Nor did the agent potentiate the carcinogenic effect of 2-naphthylamine and benzidine in rats. Conversely, neozone D was found to inhibit benzidine-induced carcinogenesis in female rats.
