The pathogenetic connection between Graves' disease and chronic lymphocytic thyroiditis. (The role and incidence of thyroid stimulating antibodies).

Antibody-positivity to thyroid specific antigens (Htg, microsomal) and/or lymphocytic infiltration of the gland's parenchyma were observed in 207 (55%) of 377 patients with Graves's disease. Only in 48 (12.7%) of the cases were the findings in agreement with the criteria of chronic lymphocytic thyroiditis. Human thyroid stimulating antibody (HTSab) was detected in 135 (65%) of these 207 patients. In cases of Graves' disease associated with chronic lymphocytic thyroiditis, this proportion was found to be as high as 89.6% and attained even 100% in cases of Hashitoxicosis (39 patients). The presence of HTSab thus seems to form one of the features of patients with Hashitoxicosis. Infiltrative ophthalmopathy also showed a remarkably high incidence (59%) in this porcess. The typical prevalence of Graves' disease in females in the present material attained a 15:1 female-to-male ratio when the disease was associated with chronic lymphocytic thyroiditis. The results of the present study suggest that chronic lymphocytic thyroiditis associated with Graves' disease promotes the formation of thyroid stimulating antibodies.

Neurotoxicological investigation of the new Hungarian pesticide Toxurazine.

The new total herbicide Toxurazine was subjected to general toxicologic and neurotoxicologic examinations. Rats were treated with 1/80, 1/40 and 1/20 of the acute oral LD50 for three months. The animals showed a decrease in body weight and a transitional anaemia. The product had a slight liver damaging effect. Renal function tests and urine analysis were negative. With time, the learning ability of animals tested in maze for 42 days became impaired dose dependently. At the end of the treatment, both at rest and on flashlight stimulation, as well as in complex EEG activity and in each frequency band there was a decrease in electric activity. In electroneuromyographic tests no effect damaging the peripheral nervous system could be detected. The hot plate method showed the reaction time of the animals to be slightly extended. The results suggest that the compound should only be used in uncultivated areas.

Stimulating effect of triiodothyronine on cell-mediated immunity.

The effects in vitro and in vivo of triiodothyronine on certain functions of human lymphocytes and polymorphonuclear granulocytes have been investigated. Triiodothyronine was able to enhance the phagocytic capacity and chemiluminescent activity of polymorphonuclear granulocytes. PHA-P-induced 3H-thymidine incorporation into lymphocytes was significantly increased in the presence of triiodothyronine. A concentration-dependent enhancing effect of triiodothyronine on antibody-dependent enhancing effect of triiodothyronine on antibody-dependent cell-mediated cytotoxicity was also observed.

Combined cyclosporin-A and methylprednisolone treatment of Graves' ophthalmopathy.

Twelve patients with Graves' ophthalmopathy (grade 1-6, ATA classification) were treated with Cyclosporin-A, systemically in combination with methylprednisolone. We observed slight or moderate favourable effect in 9 cases. Our data suggest that the benefit was due to the methylprednisolone, the effectivity of which was enhanced by the cyclosporin even in the glucocorticoid-resistant cases.
