H+/e- stoichiometry of mitochondrial cytochrome complexes reconstituted in liposomes. Rate-dependent changes of the stoichiometry in the cytochrome c oxidase vesicles.

The H+/e- stoichiometry of protonmotive cytochrome c oxidase, isolated from bovine heart mitochondria and reconstituted in liposomes, has been determined by making use of direct spectrophotometric measurements of the initial rates of e- flow and H+ translocation. It is shown that the ----H+/e- ratio for redox-linked proton ejection by the oxidase varies from around 0 to a maximum of 1 as a function of the rate of overall electron flow in the complex.

The cytochrome chain of mitochondria exhibits variable H+/e- stoichiometry.

A study is presented of the ----H+/e- stoichiometry for H+ pumping by the cytochrome chain in isolated rat liver mitochondria under level-flow and steady-state conditions. It is shown that the ----H+/e- stoichiometry for the cytochrome chain varies under the influence of the flow rate and transmembrane delta microH+. The rate-dependence is shown to be associated with cytochrome c oxidase, whose ----H+/e- ratio varies from 0 to 1, whilst the ----H+/e- ratio for the span covered by cytochrome c reductase is invariably 2.

Angiosarcoma of the bladder: a case report with regard to histologic and immunohistochemical findings.

Bladder angiosarcoma is an extremely rare neoplasia whose aggressive biological behavior follows that of other angiosarcomas. The second case of primary angiosarcoma of the bladder is reported pointing out the immunohistochemical characteristics of these neoplasias. Using immunoperoxidase staining techniques, Ulex lectin has proved to be a more sensitive marker for endothelial cells than factor VIII-related antigen, and it should be used as an additional marker for endothelial cell-derived tumors.

Is there a relationship between benign prostatic hyperplasia and prostatic cancer?

No definite relationship can be demonstrated between benign prostatic hyperplasia (BPH) and prostatic cancer on the basis of various anatomic and morphologic features. However, interesting recent observations suggest a link between BPH and prostate cancer. Clinically, the simultaneous presence of benign and malignant disease is observed quite often, and this coexistence is bound to increase with the age of patients. Possible intermediate forms between BPH and early cancer have been investigated. Two premalignant lesions have been identified: atypical adenomatous hyperplasia and prostatic intraepithelial neoplasia.

Uterine benign-appearing smooth muscle tumor "metastatic" to the lungs: a case report.

A case of pulmonary fibroleiomyomatosis secondary to a uterine "metastasizing" leiomyoma, with aspects of vascular invasion, is reported. Although we cannot exclude the existence of primitive pulmonary forms, which we do not consider to be of a hamartomatous nature, we believe that a careful examination of the uterus for the presence of angioinvasive leiomyoma is mandatory.

Protonmotive activity of cytochrome c oxidase: control of oxidoreduction of the heme centers by the protonmotive force in the reconstituted beef heart enzyme.

This paper contributes to the characterization of partial steps of electron and proton transfer in mitochondrial cytochrome c oxidase with respect to their membrane arrangement and involvement in energy-linked protonmotive activity. It is shown that delta psi controls electron flow from cytochrome c to heme a is consistent with the view that the latter center is buried in the membrane in a central position. The pressure exerted by delta psi on oxidation of heme alpha 3 by O2 indicates also that this center is buried in the membrane at some distance from the inner side and is consistent with observations showing that protons consumed in the reduction of O2 to H2O derive from the inner space. Electron flow from heme alpha to heme alpha 3 is shown to be specifically controlled by delta pH and in particular by the pH of the inner phase. Analysis of the effect of DCCD treatment of oxidase vesicles reveals that concentrations of this reagent which result in selective modification of subunit III (Prochaska et al., 1981) produce inhibition of redox-linked proton release. Higher concentrations of DCCD which result also in modification of subunits II and IV (Prochaska et al., 1981) cause inhibition of the pH-dependent electron-transfer step from heme alpha to heme alpha 3.

Basilar arterio-venous pseudoparallelism due to persistence of embryonal venous pattern.

A vascular malformation, consisting of a venous vessel bridgeing the right inferior petrosal sinus and the anterior spinal veins, was found in the posterior fossa. The vessel presented a ring-like course around the right trigeminal root, and it was parallel and dorsal to the basilar artery. The malformation was associated with cutaneous and hepatic angiomas and peri-osteal lipomas. It had been clinically silent for 52 years, when it thrombosed causing death. The authors think that, within a general mesenchymopatic state, this is a result of the persistence of an embryonal cerebral venous pattern.

Role of supernumerary subunits in mitochondrial cytochrome c oxidase.

Role of supernumerary subunits of bovine heart cytochrome c oxidase has been investigated by examining the influence on the enzymatic activity of their removal by chromatographic procedures or controlled digestion by trypsin. Is has been shown that partial proteolytic cleavage of subunit IV results in depression of respiratory activity and of redox-linked proton translocation. Selective removal by gel-filtration of subunit Vlb has no significant influence on the redox and protonmotive activity of the oxidase.

Scanning electron microscopy of 'normal' urothelium in patients with infiltrating bladder carcinoma.

'Normal' dome biopsies from 20 cystectomies for infiltrating bladder carcinomas were studied with scanning electron microscopy (SEM). All cases showed alterations ranging from minimal changes to numerous pleomorphic microvilli, and were classified according to Herd and Jacobs. The SEM findings and grades were correlated with dome histology and the results were quite good. However, in our opinion, the Herd and Jacobs classification and subclassifications are poorly reproducible without morphometric analysis. Nevertheless the SEM study is useful to strengthen the diagnosis of dysplasia/carcinoma in situ in 'healthy'-looking urothelium.

[Cystic adventitial disease (histopathological contribution of a case in the ilio-femoral region)].

Cystic adventitial disease is a rare vascular pathology mostly affecting the popliteal artery in disease is a rare vascular pathology mostly affecting the popliteal artery in men. An unusual case, located in the ilio-femoral region of a young woman, is reported. The authors claim that the cystic adventitial formations are similar to the ganglia that originate from tendon-sheaths, as structure, configuration and, probably, histogenesis are concerned. Moreover the authors suggest that cystic adventitial disease can be located among "histogenic dystrophic arteriopathies" in the pathokinetic classification of angiopathies.

[Myxoid liposarcoma and pleomorphic liposarcoma: cyto-histological correlations].

Myxoid liposarcoma and pleomorphic liposarcoma: cito-histological correlations. A correlative cytologic and histologic study of a myxoid liposarcoma of the shoulder in a 72 year-old man and a pleomorphic liposarcoma observed in the retroperitoneum of a 84 year-old woman, are presented. A preoperative FNAB cytology performed in both cases showed necrotic material containing spindle-stellate shaped cells, interspersed in a myxoid matrix, with rare classical monovacuolated lipoblasts and fragments of plessiform vessels were seen in the first and scattered pleomorphic and multinucleated cells, with prominent nucleoli and numerous atypical mitosis in the second. A malignant mesenchimal spindle-cells tumor, with myxoid matrix and pleomorphic cells, consistent with liposarcoma, respectively were suspected. Gross and histological specimens confirmed the cytological suspect. Authors discuss main cyto-histological differential diagnoses of myxoid tumors, and point out the importance and a correct differentiation between myxoid liposarcoma and intramuscular myxoma. The cytologic appearance of pleomorphic liposarcoma is similar to histologic type and therefore the problem of a differential diagnosis with soft tissue tumors is analogous. When mono or plurivacuolated lipoblasts are absent, differential diagnosis between pleomorphic histiocytoma and liposarcoma is impossible. Nevertheless this is not a important problem at cytological level because both tumors have a had prognosis and must be treated with radical surgery. Definition of correct histologic type will be more suitable on histologic specimens.

Comparison between western blotting, immunohistochemical and ELISA assay for p185neu quantitation in breast cancer specimens.

The protein encoded by erbB2 oncogene was evaluated by three different methodological approaches (Western blotting, ELISA and immunohistochemistry) in 147 breast cancer specimens. A highly significant correlation was found between Western blotting and ELISA results (p < 0.001). The data from ELISA and Western blotting were categorized as negatives (-), low-overexpressing (+) and high-overexpressing (++). Immunohistochemical results were classified as (-) or (+) according to the absence or the presence of specific staining. Overall positive cases (+ and ++) were 42.2% by Western blotting and 51% by ELISA, while (++) cases were 23.8% and 25.2% respectively. Histochemical staining was found in 29.8% of cases. The two by two evaluation of the assays showed a close association (chi2, p < 0.0001). In particular, the comparison between both ELISA and Western blotting with immunohistochemistry showed concordance rates of 78.9% for ELISA and 83.1% for Western blotting considering the + and ++ cases as a single group. When only the ++ cases were considered as positive, the overall agreement rises to 93.3% and 89.1% respectively. From these preliminary data we conclude that p185 values obtained with the three evaluated methods are possibly superimposable. Nevertheless, the biochemical methods seem to identify an intermediate p185 expression group, whose clinical meaning should be investigated.

Intrauterine insemination with husband's semen as alternative to other assisted reproduction techniques.

The objectives of this study was to assess the efficacy and clarify how many intrauterine insemination cycles can justifiably be proposed, in terms of cost and benefits for the patient, as alternative to other assisted reproduction procedures. Two hundred and twenty infertile couples were referred to our centre and six intra-uterine insemination cycles in combination with controlled ovarian hyperstimulation were proposed for them. A total of 650 treatment cycles were completed. The indications of intra-uterine insemination were male factor (147 couples), cervical factor (31 couples), male and cervical factor (30 couples) and unexplained infertility (12 couples). Superovulation was induced in 92 patients with human menopausal gonadotrophins, in 90 with clomiphene citrate, in 22 with FSH HP, while in 16 patients the intra-uterine inseminations were carried out during spontaneous cycles. Considering the entire population, a mean monthly pregnancy rate equal to 8.9% was obtained and a cumulative pregnancy rate equal to 39% at the end of six intra-uterine insemination cycles. In particular, in the patients treated with HMG+hCG, a mean monthly pregnancy rate equal to 10.9% and a cumulative pregnancy rate to 46% at the end of the six cycles were obtained. In conclusion, when the indications allow, we believe it is justified to propose six cycles of intra-uterine insemination in combination with controlled ovarian hyperstimulation with HMG and hCG. The theoretical probability of success at the end of six cycles has proved comparable and advantageous in terms of cost and risks for the patient, if compared with other assisted reproduction techniques.
