Differentiation of mast cells during postnatal development of neonatally estrogen-treated rats.

The accumulation of mast cells in the testicular interstitium of neonatally estrogen-treated rats was studied from 15 to 90 days of age. The maturation of these cells was assessed by ultrastructural analysis and their histochemical properties were examined with the sequential alcian blue-safranin staining method. The first identifiable mast cells appeared in the testis at 17-20 days of age, as immature cells with proliferative capacity. The density of mast cells increased up to 45 days of age, showing a slight decrease from 45 to 90 days of age. Before 45 days of age, most mast cells showed alcian blue-stained granules, whereas at 45 days of age, most cells presented a mixture of alcian blue and safranin-stained granules. From this age onward, most cells were stained with safranin. These maturational changes were well-correlated with their ultrastructural features. Mast cells presented few and heterogeneous immature granules up to 45 days of age, and many uniform electron-dense granules at 90 days of age. These results indicate that the testicular interstitium of neonatally estrogen-treated rats provides an advantageous environment for the recruitment, proliferation and maturation of connective tissue mast cells.

Cadmium-induced alterations of chlorpropham metabolism in isolated rat hepatocytes.

In order to investigate the various steps of chlorpropham (CIPC) metabolism which could be influenced by cadmium, isolated rat hepatocytes were incubated in the presence of CIPC (0.1 mM) and of increasing Cd concentrations (0-180 microM). The results showed that Cd accumulation in hepatocytes was in good correlation to its concentration in the incubation medium. At 90 microM Cd, hydroxylation of CIPC was only slightly decreased by 30%, while CIPC hydrolysis into 3-chloraniline was unaffected by the presence of Cd. Accordingly, unchanged CIPC increased in hepatocytes. At 27 microM Cd, free 4-hydroxychlorpropham (4-OHCIPC) increased in the intracellular medium as a consequence of a strong suppression of both sulfation and glucuronidation which was related to the strong depletion of the intracellular ATP level under the combined influences of both cadmium and free 4-OHCIPC. Acetylation of 3-chloroaniline, which represents a minor pathway of CIPC metabolism, was already markedly suppressed (43%) with the lowest Cd concentration (27 microM). These in vitro results suggest that Phase II reactions are more sensitive to Cd than Phase I processes and that Cd enhanced the CIPC cytotoxicity as shown by alterations of the membrane integrity.

Interaction of hexachlorocyclohexane (HCH) and chlorpropham (CIPC) in male rats.

Hexachlorocyclohexane (60.0 mg/kg/d) and chlorpropham (50.0 mg/kg/d) given to male rats daily orally for 30 d caused subtle biochemical lesions. The significant rise in the activity of alkaline phosphatase in liver and serum and of GOT and GPT in liver with substantial weight increase of liver has been suggestive of the initiation of biochemical lesions and adaptation of the organism. Very severe hepatocellular damage is forecasted if the pesticides are given for a prolonged period and in higher concentrations to animals.

Determination of 2-carboxy thiazolidine-4-carboxylic acid in biological fluids by ion-exchange chromatography.

A column ion-exchange chromatographic method for the determination of 2-carboxy thiazolidine-4-carboxylic acid (TDCA) in blood and urine is described. After elimination of endogenous thiols, alkaline hydrolysis of the compound yields cysteine which is determined spectrophotometrically. The described method is specific for intact TDCA. Using a 3-ml aliquot of sample it allows the linear determination of this hepatoprotective drug from 5 micrograms TDCA ml-1 with a detection limit of 3 micrograms ml-1. The method was applied to study the time course of plasma levels and urinary excretion. An in vitro metabolism study showed that TDCA has a potential as a cysteine donor for glutathione synthesis.

[Pulmonary edema. A rare complication of the percutaneous insertion of silastic central venous catheters].

Seven newborn infants (birth weight 920 to 1,900 g) who developed pulmonary oedema as a complication of the use of percutaneous silastic central venous catheters are described. Clinical symptoms occurred three to forty days after catheter placement. In each case, radiographic detection, performed by the injection of radiopaque dye, localized the tip of the catheter in the pulmonary artery or in its collateral branches. Clinical symptoms decreased after catheter replacement in the right atrium. The casistic examination allows the Authors to suggest some aetiopathogenetic hypothesis.

[Nutritional status of the rat in refeeding following massive mobilization of DDT by fasting].

Rats were subjected to fasting-induced mobilization of a quantity of DDT equivalent to 45% of the LD50. They were then fed for 1 week for a nutritional and toxicological study. Mobilization of DDT by starvation had no apparent effect on the activity of the various enzymes or the weight of the organs measured at the end of the refeeding period. The animals which had received DDT pretreatment showed higher protein efficiency than the control animals. Only the level of body lipids was lower in the animals pretreated with DDT, although the quantities of lipids stored during the study period were identical in both groups of animals. Overall, the results show that the nutritional state of the two groups of animals was very similar at the end of the refeeding period.

[Percutaneous silicone catheters in the newborn infant].

246 percutaneous insertions of silastic venous catheters were performed in 189 newborn infants. This technique makes perfusion in newborn easier and safer, with less complication risks and less work for nurses. Furthermore silastic venous catheters insure a better observation of neonatological care methods. As a consequence it improves the vital prognosis and the quality of newborn survival in the newborn intensive care unit.
