[Differentiation of strains of bovine infectious rhinotracheitis virus using restriction analysis].

The virus of infectious rhinotracheitis of cattle (BHV-1) causes the respiratory diseases, encephalitis, vulvovaginitis, abortions, etc. in sensitive animals. The attempts to differentiate the viral strains by virology and serology methods were unsuccessful. The restriction analysis makes possible to divide the strains into the three main groups designated BHV-1.1, BHV-1.2 and BHV-1.3. Majority of authors note the lack of correlation between the restriction pattern of DNA and the syndromes caused by the strains of the first and second groups while all the representatives of the third group were isolated in case of meningoencephalitis. The restriction analysis of BHV-1 strains isolated in the USSR by restriction endonucleases EcoRI, BamHI, HpaI and ClaI is presented. The obtained results show the absence of correlation between the restriction pattern of the strain and the syndrome caused by the viruses of all three groups. The representatives of the group BHV-1.3 that are not associated with the neurological syndrome are isolated for the first time.

Development of hybridization probes on the basis of recombinant DNA to identify porcine parvoviruses and rotaviruses.

Porcine parvovirus (PPV) and porcine rotavirus (PRV) infections can be diagnosed, and the diagnosis greatly contributes to their control. For the diagnosis of PPV and PRV of pigs recombinant DNA were obtained which could be used as hybridization probes. Homology between PPV-RNA and cloned cDNA was confirmed, and the ability of plasmid probes to detect PRV was demonstrated. Hybridization with preparations of both PPV-infected cells and purified PPV gave positive reproducible results. The results are useful as an experience for the development of a sandwich hybridization system for PPV detection.

[Isolation and properties of membrane-bound DNA of the chromatin peripheral layer in the blood of healthy cattle and cattle with chronic lympholeukemia].

The nuclei and the fraction of chromatin peripheral layer bound to the nuclear membrane were isolated from blood lymphocytes of healthy and chronic lympholeukosis (CLL)-stricken cows. The isolation procedure included treatment of nucleic with DNase I with subsequent extraction of chromatin by low ionic strength salt solutions and purification of nuclear membranes by sucrose density gradient centrifugation. The blood lymphocyte nuclei from cows suffering from CLL differ from nuclei of healthy animals by a higher degree of chromatin condensation and a lower RNA content. Amount of DNA in the peripheral chromatin layer in lymphocyte nuclei of cows suffering from CLL is two times as high as that in healthy animals. The GC-content in the peripheral chromatin layer DNA from lymphocyte nuclei of healthy and CLL-stricken animals does not differ from that of total nuclear DNA (about 45 mol.%). Both in healthy and CLL-stricken animals, the 5-methylcytosine content in the chromatin peripheral layer DNA is by about 20% less than in the total nuclear DNA of healthy cows. In the chromatin peripheral layer DNA of blood lymphocytes from CLL-stricken animals the methylation level is unaffected. Therefore, a certain decrease in the total nuclear DNA methylation in CLL-stricken animals observed is due to methylation changes in DNA which is not bound to the nuclear membrane.
