Influence of perftoran on structural and metabolic disturbances in the liver during experimental atherosclerosis.

We evaluated whether perftoran can be used for the correction of structural and metabolic changes in the liver during alimentary atherosclerosis. Perftoran in a dose of 0.3 g/kg normalized the contents of phosphatidylserine, phosphatidic acid, and phosphatidylcholine in the plasma membranes of liver cells in rabbits feeding an atherogenic diet for 2 months. Architectonics of hepatic trabeculae returned to normal, the degree of granular and vacuolar degeneration in hepatocytes decreased, and phagocytic activity of macrophages increased. Individual peculiarities of the effect of perftoran on structural and metabolic changes in the liver were revealed.

Changes in phospholipid composition of synaptic membranes in medulla oblongata and frontal lobes of the cerebral hemispheres in cats with hemorrhagic shock.

We studied phospholipid composition of brain synaptic membranes isolated from cats with severe hemorrhagic shock. Changes in the medulla oblongata were most pronounced and manifested in decreased content of phosphatidylcholine. Changes in the phospholipid composition of synaptic membranes in the frontal lobes included an increase in phosphatidylinositol content and reduced content of phosphatidylserine. Accumulation of phosphatidylethanolamine in synaptic membranes was found in both the medulla oblongata and frontal lobes. These data help to understand the mechanisms underlying exhaustion of compensatory reserves in brain cells during severe hemorrhagic shock.

[Effect of phosphatidylcholine liposomes on lipid peroxidation in the liver and phospholipid metabolism in hepatocyte plasma membrane in hemorrhagic shock].

The possibility of protecting the liver from lipid peroxidation (LPO) by phosphatidylcholine liposomes was studied in a model of hemorrhagic shock. Augmentation of LPO in hemorrhagic shock correlated with a decrease in phosphatidylcholine levels in the plasma membranes of hepatocytes and an increase in phosphatidylinositol content. With the use of liposomes the activation of LPO in the liver was removed and the level of phosphatidylcholine and phosphatidylinositol in the hepatocyte plasma membrane was restored.

[Characteristics of periodic motility of the upper portion of the jejunum in rabbits].

Proximal jejunal motility was studied balloonographically in rabbits devoid of food and in properly fed animals. Three phases of jejunal motor activity were revealed: the resting phase, irregular contraction phase and rhythmic phase. It was found that the regularity of rhythmic phase onset was not stable. The rate of continuous contractions during rhythmic phase and the rate of interrupted contractions during irregular phase coincided and were relatively constant. The feeding did not affect the rate of contractions during rhythmic and irregular phases, but increased the duration of the rhythmic phase.

[Compensatory and adaptive changes in the small intestine and liver of rabbits after short-term cholesterol diet].

Functional and morphological changes in the intestinal wall and liver were studied in rabbits on short-term cholesterol diet. It was established that with a rapid increase of cholesterol concentration in the general blood flow, the synthesis of high density lipoproteins in the intestinal wall was intensified. Enhanced hepatic elimination of cholesterol and chylomicrons from blood circulation contributes to cholesterol level stabilization in peripheral blood. With high density lipoprotein accumulation in the intestinal wall, cholesterol consumption did not change its concentration in the general blood flow. Structural changes in jejunal and liver mucosa were shown to depend on the degree of hypercholesterolemia and functional damage of these organs.

[Interrelation of the changes in the blood rheological properties and the microcirculatory disorders in the early and late stages of experimental hyperlipoproteinemia].

Experiments were made on rabbit fed an atherogenic diet (0.5 g/kg cholesterol) singly for 15 and 24 h and repeatedly for 3, 9 and 30 days. At early stages of lipid metabolism distress the interrelationship was established between blood rheological disorders and microcirculatory abnormalities. The dependence of the initial reaction of some rheological characteristics on their initial level was marked.

[Functional and structural changes in the small intestine during experimental hypercholesteremia].

Functional and structural changes that occurred in the small intestine and liver of rabbits with alimentary hypercholesterolemia have been studied. Maximal rise of peripheral blood cholesterol after a single exposure to cholesterol was coupled with an increased penetration of chylomicrons and low and very low density lipoproteins from the intestine to the circulation. The decrease of cholesterol excretion along the whole length of the small intestine was accompanied by activation of the release of high density lipoproteins into the blood outflowing from the intestine. The decay of chylomicrons in the liver was restricted during the first days of hypercholesterolemia. Cholesterol concentration in the bile was decreased. The microvessels of the small intestine demonstrated the dilatation of the postcapillary-venular component erythrocyte aggregates and capillarostases.

[Lipoprotein metabolic characteristics in the liver and intestinal wall of rabbits after a single exposure to sunflower oil and cholesterol].

Lipoprotein metabolism in the rabbit liver and intestinal wall was studied by an alimentary action on the cholesterol blood content. The data obtained indicated that the diet including cholesterol intensifies the release of chylomicrons into the lymph of the intestinal lymphatic trunk. Single addition of sunflower-seed oil to the diet leads to the increased deposition of high, low and very low density lipoproteins in the intestinal wall. Upon adding cholesterol to the rabbit diet the retention of low and very low density lipids in the intestine is followed by the increased release of high density lipoproteins into the blood of the portal vein. Single addition of sunflower-seed oil stimulates the synthesis of high density lipoproteins by the rabbit liver.

Protective effect of various forms of superoxide dismutase on phospholipid composition of hepatocyte and adipocyte plasma membranes and blood lipoprotein spectrum during hemorrhagic shock on cats.

Using a model of hemorrhagic shock, the possibility of protection by Cu,Zn-superoxide dismutase (SOD) of the phospholipid bilayer of plasma membranes of hepatocytes and adipocytes as well as of the blood lipoproteins was studied. SOD (5 mg/kg), injected 30 min after the onset of bleeding, efficiently prevented changes in the phospholipid bilayer of hepatocytic plasma membranes in cats. Simultaneous injection of SOD partly restored the concentration of different classes of phospholipids in plasma membranes of mesenterial fatty tissue adipocytes altered by shock. When incorporated into liposomes, SOD exerted a weaker corrective effect on the phospholipid composition of hepatocytic plasma membranes in animals with hemorrhagic shock and simultaneously produced a membrane-stabilizing effect on adipocytes. In contrast to pure SOD (which had no effect on the lipoprotein composition of the blood in animals with hemorrhagic shock), SOD-containing liposomes decreased the amount of chylomicrons and very low density lipoproteins in the venous blood of central vessels.

[Comparative effect of partial opiate agonists buprenorphine and butorphanol on the phospholipid composition of hepatocytes plasma membranes during hemorrhagic shock in cats].

Comparative effects of the partial opiate agonists buprenorphine and butorphanol on the phospholipid composition of liver cell plasma membranes were investigated in cats under conditions of hemorrhagic shock. Buprenorphine administration (0.03 mg/kg) normalized the level of phosphatidylinositol. The higher dose (0.3 mg/kg) induced additional disturbances in the phospholipid composition of liver cell plasma membranes by increasing the content of phosphatidylserine and by decreasing the contents of sphingomyelin and lysophosphatidylcholine. The protective effect of butorphanol on liver cells was more pronounced than that of buprenorphine.

[Dose-dependent effect of diltiazem on some lipid metabolic parameters in hemorrhagic shock in cats].

The possibility of modulating the phospholipid composition in the plasma membranes of hepatocytes and adipocytes by the calcium channel blocker diltiazem was studied on a model of hemorrhagic shock. Diltiazem injection facilitated normalization of phosphatidylinositol metabolism in the hepatocyte plasma membranes. Injection of a high dose of diltiazem was attended by a rise of the phosphatidylethanolamine level. Only low doses of diltiazem caused a normalizing effect on the phospholipid composition of adipocyte plasma membranes. A low dose of diltiazem caused a reduction in the content of very low density lipoproteins in venous blood, while a high dose reduced the content of low density lipoproteins.

[Effect of perftoran on microcirculation and target organs in early atherogenesis].

Effects of perfluoran, a plasma substitute with a gas-transport function, were studied at the stage of dyslipidogenic microangiopathy in rabbits. Changes in microcirculation, hemorheology, morphology of the liver, phospholipid composition of hepatocytic membranes were examined histologically, histochemically, electron microscopically and biochemically. Perfluoran produced a positive effect on microcirculation, phospholipid composition of hepatocytic membranes. Individual features of these parameters' dynamics were determined for animals.
