Protein kinases in rat testes. Subcellular distribution of the enzyme and endogenous substrates.

The subcellular distribution of cAMP dependent and independent protein kinases (Pr K) has been studied in rat tests. The enzymatic activity was preferentially located in the cytosol. In all fractions mature animals have higher specific activity. The localization of endogenous protein substrates of Pr K was investigated using cytosol as an enzymatic source. The best endogenous substrate for the cAMP dependent Pr K seemed to be localized in the microsomal fraction. The rate of protein phosphorylation in the testicular subcellular fractions was examined by in vitro incubation with 32P-orthophosphate. The nuclear fractions have the highest specific radioactivity.

Protein kinases in rat testes: evidence for different fractions of the enzyme.

Protein kinase activity of rat testis homogenate was separated into five fractions by means of pH 4.8 acidification and DEAE-cellulose chromatography. The five fractions showed a peculiar pattern of activity and cAMP dependency with the substrates used: casein, protamine, histone mixture, arginine-rich histone, lysine-rich histone, and phosvitin. The casein-sepharose substrate affinity column separated two fractions from the pH 4.8 precipitate. Peak number one phosphorylates histone preferently and is cAMP-dependent, while peak number tow has a strong affinity toward casein as substrate and is non cAMP-dependent.

Metabolism of steroids by left ovaries of Gallus domesticus during post-embryonic maturation.

Homogenates of left ovaries from laying hens incubated with labeled pregnenolone or progesterone produced slightly higher amounts of estrone than of estradiol-17 beta. Progesterone, 17 alpha-hydroxyprogesterone, androstenedione and testosterone are probable intermediates. Left ovaries from 1-, 21- and 40-days-old chicks incubated with progesterone as substrate formed increasing amounts of androgens and estrogens. Weight per weight, ovaries from one-day-old chicks have a greater steroidogenic capability than those from adults. The comparative steroidogenesis by testes and left ovaries from Gallus domesticus during growth is also reported.
