Interactions of radio-detoxified Escherichia coli endotoxin preparations with the complement system.

Escherichia coli O89 lipopolysaccharide (LPS) was treated with different doses of gamma irradiation (5, 10, 15, and 20 Mrad). Various biological activities such as lethal effect, decrease in arterial blood pressure in dogs, and interaction with the complement system were determined for the parent and irradiated preparations. Irradiation of LPS significantly and in a dose-dependent manner decreased its lethal and blood pressure-depressing effects along with its ability to activate the complement system. In contrast, radio-detoxified LPS fixed more strongly the isolated human C1 than did the parent LPS. The possible connection between the toxicity of endotoxin and endotoxin-induced complement activation is discussed.

Role of C1 in the complement activating effect of Pseudomonas aeruginosa lipopolysaccharide preparations.

The complement consumption of endotoxin preparations extracted by the trichloroacetic acid or phenol-water method from different Pseudomonas aeruginosa strains was measured in normal human and guinea pig serum and in serum chelated with Mg2+-EGTA. In the chelated serum, which was essentially Ca2+-free, the first component of complement (C1) could not exert its function. All preparations tested consumed considerably less complement activity in chelated than in normal serum. The proportion of CH50 units fixed in Mg2+-EGTA and in normal serum was always higher in the tricholoracetic acid extract than in the phenol-water extract of the same strain. The part of LPS molecule that was able to activate the complement system in Ca2+-free serum was partially separated from the C1-requiring part by the combination of different extraction methods. The results suggest that on the LPS molecules two different sites are responsible for the complement activating effect through the classic and the alternative pathways.

The c 1 fixation and transfer test for studying immunity in melanoma patients.

Sera from 27 melanoma patients and from 30 healthy persons were investigated for humoral immunity by C 1 fixation and transfer test. The number of C 1 molecules fixed to serum treated, sensitized tumor cells and C 1 molecules fixed to these cells in vivo were calculated and the quotient of these values was determined. On the basis of the quotient the patients could be divided into four groups. In Group 1 the serum treatment of cells decreased the C 1 fixation; in these cases the presence of blocking factors can be supposed. In Group 2 the serum treatment did not cause any difference: the patient serum does not contain humoral antibody. In Groups 3 and 4 the C 1 fixation rose slightly and significantly respectively. Work is in progress to find a possible relation of the results with the stage of the disease.

Serum beta1A/C-globulin level in early myocardial infarction.

Serial titration of the third component of complement (beta1A/C-globulin) was performed in 170 serum samples of 39 patients in the early period of acute myocardial infarction. During the first 24 hours there was a rise in mean concentration values. During the next 24 hours they remained unchanged, in severe cases the mean values even decreased. Subsequently the levels continued to increase. In 27 cases a sharp fall of the level exceeding the limit of normal changes was observed. It is suggested that during the early period of acute myocardial infarction the serum concentration of beta1A/C-globulin is under the influence of two opposite processes, one inducing a decrease, the other an increase of the level.
