[CT and angiography in hepatic metastases (author's transl)].

In a series of patients affected by hepatic metastases, the authors have found a diagnostic accuracy of 84% by means of CT, poorly inferior to that of angiography: 163 patients were studied by means of angiography, 88 by CT, 12 with both techniques besides scintigraphy, echotomography and other investigations. In many patients when scintigraphy and echotomography do not lead to a diagnosis, CT can avoid an angiographic investigation. Nevertheless angiography must be performed in all those patients in which the investigations suggest a possible surgical intervention. Despite employing all possible investigations, there are some cases in which the hystological diagnosis of the lesion can be reached only by optic by means.

Synthesis and antitumor activity of 4-demethoxyadriamycin and 4-demethoxy-4' -epiadriamycin.

Two new analogs of adriamycin have been obtained by chemical synthesis, 4-demethoxyadriamycin and 4-demethoxy-4' -epiadriamycin. Both compounds were highly effective against experimental mouse tumors at doses about ten times lower than those effective for adriamycin. At the optimal dose, 4-demethoxyadriamycin displayed antitumor activity similar to that of adriamycin in solid Sarcoma 180 (S180), L1210, P388, and Gross leukemias, and mammary carcinoma, while it did not markedly inhibit the growth of Moloney sarcoma virus-induced sarcoma in mice treated before the virus infection. At the optimal dose, 4-demethoxy-4' -epiadriamycin was as active as adriamycin against L1210, P388,and Gross leukemias, and less active against solid S180. The results show that anthracycline derivatives characterized by the absence of the methoxyl group at the C-4 position are markedly more potent than the parent compound, and may exhibit a differential antitumor effect on a number of mouse tumors.

Synthesis of derivatives of 3-amino-2,3-dideoxy-L-hexoses related to daunosamine (3-amino-2,3,6-trideoxy-L-lyxo-hexose).

The synthesis is described of 3-amino-2,3-dideoxy-L-arabino-hexose (10), methyl 2,3-dideoxy-alpha-L-lyxo-hexopyranoside(17), methyl 3-amino-2,3-dideoxy-alpha-L-ribo-hexopyranoside (21), methyl 2,3-dideoxy-3-trifluoroacetamido-alpha-L-xylo-hexopyranoside (26), and certain derivatives from methyl 4,6-O-benzylidene-2-deoxy-alpha-L-arabino-hexopyranoside (3). Conversion of 2-deoxy-L-arabino-hexose into 3 by modified, standard procedures, and on a large scale, gave a 75% yield.

[Progress of clinical pharmacology in essential headaches].

By testing venomotor receptors (venous-constriction test), it was possible to show a potentiation of the venous constriction effect of serotonin by methysergide and ergotamine. This 5-HT potentiation was observed only when these drugs were sampled in concentrations similar to those in therapeutic use. When ergotamine and methysergide are sampled in concentrations higher than those used in clinical practice, the expected 5-HT antagonist effect is clearly evident. We could therefore conclude that methysergide and ergotamine can be considered pro-serotonin and not anti-serotonin drugs. The pro-serotonin effect of methysergide and ergotamine might take place peripherally on vasomotor or sensitive receptors and/or at a CNS level, making up for the lack of serotonin which, according to the central theory of essential headache, provokes the painful cephalic syndrome.

Synthesis and anticancer activity of 5-diethylaminomethyl derivatives and nitrogen mustards of uracil and 2-thiouracils.

Several 5-diethylaminomethyl derivatives and nitrogen mustards of uracil and 2-thiouracil have been synthesized and tested for their potential anticancer activity in vitro on KB cells and in vivo on Ehrlich carcinoma. Among the alkylating derivatives tested several showed cytotoxic activity in vitro and compound V [5-[bis(2-chloroethyl) amino] methyl-6-propyluracil hydrochloride] showed both in vitro and in vivo anticancer activity.

Serotonin vascular reactivity and other clinical pharmacological investigations in two intestinal carcinoid patients.

In two patients with carcinoid syndrome, a reduced venoreactivity to serotonin (5-HT) was found by means of the venoconstriction test. Parachlorophenylalanine (PCPA) treatment increased 5-HT venoreactivity. A normal noradrenaline venoreactivity was detected before and during PCPA administration. The altered platelet monoamine oxidase (MAO) activity corrected by PCPA treatment. The abnormal 5-HT turnover may alter the synthesis of MAO and provoke the production of isoenzymes. Low plasma amine oxidase and high plasma histaminase activity were found in 1 of these 2 patients. A low level of plasma prekallikrein in the two patients remained unchanged during PCPA administration.
