Human normal CD5+ B lymphocytes can be induced to differentiate to CD5- B lymphocytes with germinal center cell features.

A significant proportion of cord blood CD5+ B cells express the activation molecules CD23, CD25, and transferrin receptor; react with the cell-cycle-associated monoclonal antibody (MoAb) Ki67; can be induced to enter the S phase of cell cycle by interleukin-2 (IL-2), IL-4, or low-molecular-weight B-cell growth factor (Imw-BCGF) and, exposed to IL-1 and IL-2, acquire the features (sIgD-, CD5-, CD10+, CD38+) of B blasts proliferating in the germinal centers of secondary follicles. These findings indicate that CD5+ B cells are preactivated and, in the proper microenvironment, may give rise to CD5- B cells.

Comparative analysis of normal and malignant CD5+ B lymphocytes.

The T-cell related CD5 molecule is expressed by the major B cell population which forms primary follicles in fetal lymph nodes and spleen and circulates in cord blood but decreases to a numerically minor proportion (5-10% of all B cells) in adults [1, 3-5, 8, 9, 12, 14, 16]. The CD5 molecule is also expressed by the monoclonal B cells of B-chronic lymphocytic leukemia (B-CLL: reviewed in [5]). Even if there is no proof that CD5+ B cells are the target of the transforming events which lead to B-CLL, they are regarded as the normal counterpart of B-CLL. Therefore, the aim of the present work was the analysis of the phenotype, the cell cycle control and the cytoskeleton organization of normal CD5+ B lymphocytes in comparison with the data obtained on malignant CD5+ cells from B-CLL patients.

Dynamics of the binding capacity of plasma sex hormone binding globulin (SHBG) for testosterone and dihydrotestosterone during puberty.

Plasma sex hormone binding globulin binding capacity (SHBG-b.c.) has been evaluated in 203 normal subjects (114 males and 89 females) aged 3 to 51 years. The subjects were divided into groups: prepubertal, early pubertal (Tanner's Stages 1 and 2), late pubertal (Tanner's Stages 4 and 5) and adult. In both sexes, plasma mean values of SHBG binding capacity both for dihydrotestosterone (DHT) and testosterone (T) were significantly higher in prepubertal subjects, falling during puberty to adult levels. During pubertal development DHT-BG binding capacity and T-BG binding capacity showed different plasma values with respect to sex and phase of puberty. Our data do not support an absolute relationship between sex hormones and SHBG binding capacity, but suggest other mechanisms as well: (a) SHBG modifies its physicochemical properties during puberty, or (b) the binding capacity is the result of a pool of proteins which modifies its composition during pubertal evolution.

Increased plasma corticosteroid-binding globulin in insulin-dependent pubertal diabetics: relationships with other glycoproteins, growth hormone and prolactin.

A selected group of 13 juvenile-onset insulin-dependent diabetics in mid-late puberty (stages 3-4) and under comparable conditions of metabolic control was studied in order to evaluate the plasma levels of corticosteroid-binding globulin (CBG, transcortin: expressed as the binding capacity for cortisol) in relation to the levels of other glycoproteins and to growth hormone (GH) and prolactin (PRL) responsiveness to provocative tests (arginine infusion and TRH injection, respectively). Further evidence was provided that juvenile-onset diabetics show most frequently enhanced plasma CBG binding capacity; statistical significance with p-values less than 0.01 was attained in comparison to 25 age-matched controls. Among other variables examined, only hemoglobin A1c amd alpha 2-macroglobulin were significantly raised in the diabetic group (p less than 0.01 and less than 0.02, respectively). In our patients growth hormone response to arginine infusion was in the normal range, whereas PRL response to TRH was slightly but significantly supranormal in terms of maximum value and maximum increment above baseline value. No correlation was found between CBG binding capacity and other variables examined. We conclude that raised levels of CBG may occur as an additional alteration of the plasma glycoprotein pattern in juvenile-onset insulin-dependent diabetics. Specific regulatory factors conceivably subserve plasma concentrations of different glycoproteins.

Plasma bile pigment conjugation modalities in icterus syndromes of various origin.

Plasma azopigments derived from conjugated bilirubin were analyzed by thin-layer chromatography according to HEIRWEGH et al. in 14 cases of obstructive jaundice and in 11 of acute hepatitis. The chromatographic patterns were compared with those obtained from azopigments derived from 8 normal bile samples. The plasma pigment patterns did not differ from those of the bile in number and chromatographic mobility of the spots. However, the quantitative percentages of the plasma azopigments were significantly modified: the alpha 0 fraction (free azodipyrrolic pigment) increased in both icteric syndromes, while the delta fraction (mainly glucuronide azopigment) decreased. Moreover, the behavior of two closed components of the delta group showed significant differences in both icteric syndromes. It can be postulated that the synthesis of bilirubin diconjugates decreases both in hepatocellular and cholestatic jaundice, while monoglucuronidated as well as saccharide and glucoside conjugates increase. In cholestatic jaundice the conjugation with glucuronic acid mainly takes place in the normal way, whereas compounds with different features are formed in hepatitis.
