Monoclonal antibodies directed to the O antigen of Salmonella serogroup E cross-react with lipopolysaccharides of Salmonella serogroups C, F and S.

Four murine hybridoma clones secreting monoclonal antibodies (mAb) directed against lipopolysaccharide (LPS) antigen of S. newington, serogroup E1, were obtained after a fusion of spleen cells of mice immunized with formaldehyde-killed bacteria and mouse myeloma cells of the X63-Ag8.653 line. Antigen binding properties and specificity of the mAb were studied in bacterial agglutination tests, passive hemolysis and its inhibition, passive hemagglutination tests, passive hemolysis and its inhibition, passive hemagglutination and immunoenzyme tests (ELISA and immunoblotting). Three of the mAb (24E6, 29E1 and 45F6) were agglutinating and were active in all tests used, while mAb 31H12 did not agglutinate bacteria but revealed a high reactivity in the immunoenzyme reactions. It was found that the mAb reacted with LPS and Salmonella strains from serogroup E (E1, E2, E3 and E4) as well as from serogroups C (C1 and C4), F and S thus showing that the O3 antigen possesses more than one epitope, one of which is represented on the LPS antigens of the serovars from the cross-reacting groups mentioned. According to the known chemical the most probable recognized epitope consists of mannose with beta-linkage to the next monosaccharide residue in the LPS chain.

[Activation of lipid peroxidation and changes in vitamin E contents in the lungs under oxidative stress].

The lipid peroxidation (LPO) of the lung tissue and the bronchoalveolar lavage in rats under the influence of immobilization has been investigated. The effects accompanying the development of oxidative stress in animals--an increase in the content of conjugated dienes and fluorescent LPO products in biological objects and a strong decrease in the content of vitamin E in the lung tissue were registered.

Characterization of NAD(P)H diaphorase from boar spermatozoa using specific monoclonal antibodies.

1. After immunization of BALB/c mouse, four monoclonal antibodies against soluble NADH diaphorase from ejaculated boar spermatozoa were produced and characterized. The monoclonal antibodies were designated as follows Mab 1F2, Mab 4E2, Mab 5B8, Mab 5D8. 2. These monoclonal antibodies react with other enzyme forms-sedimentary NADH and NADPH and soluble NADPH and inhibit (although not completely) their activity. It is supposed that different forms of the enzyme share some common epitopes. 3. Treatment of ejaculated boar semen with 2O-methylcholanthrene causes an increase of the activity of the soluble diaphorase form only. 4. These results lead to the assumption that the sperm diaphorase is a dynamic enzyme system consisting of four immunologically similar isoenzymes although their functions are different.

[Premature sexual activity among girls and an attempt at sex education].

In connection with early sexual activity among juveniles the authors interview 292 girls at the age of 15-18 years in three towns (Sofia, Pleven, Cherven Bryag). Sexual activity is studied in girls as well as their initial information level in taking decision for sexual participation. 38 (13.01) of the girls were sexually experienced as 12 (13.57%) of them had sexual activity more than 2 years. Girls, aged 16 and 17 years, had the highest sexual activity (44.75%). 10 (26.31%) of the sexually active girls had more than 2 partners. 11 (28.95%) of the girls were informed very well, but 4 (10.53%) of them had almost no information. 4 (10.53%) of sexually experienced girls used contraceptives and those were mainly condoms and pills. The necessity of timely sexual education is stressed, taking into consideration that pregnancy is a consequence of early sexual activity at adolescent age.

[The effect of betamethasone (celeston) on fetal lung development in rats].

The influence of betamethasone (celestone) was studied on the fetal lung development in pregnant rats of Wistar strain. It was shown that the drug was without effect, administered singly. After treatment of the pregnant rats for a period of 3 days with a dose of 0.100 mg/kg the content of the lung surfactant in their amniotic fluids was increased, manifested by an elevation of the threshold dilution (TD), determined by the method of the black foam film. Administration of two-fold higher dose did not cause quicker synthesis of surfactant. The morphological examination of the fetal lung showed that celestone accelerated the synthesis, maturation and secretion of surfactant and intensified differentiation of prealvelocytes of I and II types after the 18th day of the fetal development, which occurred a day earlier than in the control rats.

[Experimental study of the action of corticosteroids on pulmonary surfactant].

The authors carried out studies on pregnant rats of Wistar strain, divided into three groups: a control group of rats treated with saline, a group of rats treated with celeston and a group of rats treated with oradexon. The amniotic fluid, obtained by amniocentesis from the amniotic sac, was examined. Threshold separation, showing the concentrations of phospholipids in the amniotic fluid, was determined by a new original physico-chemical method--the method of foam black film. The results from the 3 groups were compared. The difference in the values of threshold separation between the control and treated groups is obvious as well as between the groups treated with celeston and oradexon in favour of oradexon, which is a proof for the effect of corticosteroids on the surfactant system.

[Acute obstruction of the mesenteric vessels--a diagnostic and therapeutic problem].

Analysis was made of 175 patients treated for acute obstruction of the mesenteric vessels. The incidence was higher in males (male/female ratio = 2.5/l) and in winter--62 per cent of the cases. An the basis of thorough analysis of the most common diseases responsible for thrombus formation and thromboembolism and the basic symptoms in the clinical picture, ad ot table was elaborated for early diagnosis of acute obstruction of mesenterial vessels, enabling to take quick decision even without angiography. The level of obstruction in 57 per cent of the patients was in the stem of the mesenterial arteries; conclusion is hence made that, when possible, only reconstructive vascular operation associated or not with intestinal resection, will lead to permanent cure. Of all 175 patients in this series, 81 (46.5 per cent) were operated and 68 of them (83.9 per cent) died; only 13 survived (16 per cent). All of the unoperated 94 patients (53.5 per cent) died. Hence, a total of 162 patients died (92.6 per cent) and 13 survived (7.4 per cent).

Acute Disease

[Indomethacin and lung maturity].

The authors treated in advance two groups of pregnant rats of Wistar strain with indomethacin in doses of 3 mg/kg of body mass and 5 mg/kg of body mass for a period of 2 days. Then the animals were killed on 18th, 19th and 20th day of gestation and the change in the lung surfactant of amniotic fluid was determined by the method of the black foam film. There was threshold dilution in both groups of treated rats in comparison with the control group. They found changes in the gastrointestinal tract of the mother and fetus death of some fetuses after larger parenteral administration of larger dose of indomethacin. An inference is made for the negative effect of indomethacin on lung maturation.

Amniotic Fluid

[Porphobilinogen synthase activity and porphyrin biosynthesis in the erythrocytes after incubation with delta-aminolevulinic acid in pernicious anemia patients].

The activity of PBG-S was studied in erythrocytes according the standard method of Berlin and Schaller as well as the biosynthesis of porphyrins after incubation with delta-aminolevulinic acid according to Idel'son in a control group of 40 healthy males and females and 15 patients with pernicious anemia. The activity of porphobilinogen synthase and the level of porphobilinogen were elevated in the patients studied. The total quantity of synthesized porphyrins is increased as regards the control group of healthy on the account of the enhanced biosynthesis of uroporphyrins and coproporphyrins, whereas the quantity of the protoporphyrin synthesized was reduced. The changes established reflect the secondary disordered heme synthesis in the patients with pernicious anemia.
