Electromagnetic stimulation on the bone growth using backscattered electron imaging.

The events at the hydroxyapatite implant material/tissue interface following electromagnetic stimulation were studied in the rabbit. Two kinds of hydroxyapatite were used: natural (NA) and synthetic (HA) both with a grain size of <50 microm. Bone defects, artificially created in rabbit tibiae, were filled with the material examined. One group of animals was exposed immediately after surgery and every 12h thereafter to 30-min treatments with electromagnetic fields (PEMFs). A second group was used as a control (untreated). Two and 4 weeks after implantation, animals were sacrificed and bone samples processed for LM, TEM and SEM using a backscatter electron detector for the evaluation of bone growth. This study indicates that HA has more osteoconductivity than NA, and shows that PEMF-treatment results in a benefit in accelerating bone formation at early time periods.

Pirenzepine in the treatment of duodenal ulcer.

The effectiveness and safety of pirenzepine (PRZ) at 150 mg daily in the treatment of active duodenal ulcer was studied in an open pilot clinical trial followed by a double-blind trial against placebo. The open pilot study showed that pirenzepine was well tolerated and promoted ulcer healing in the 45% of patients within 2 weeks of treatment and in 90% within 4 weeks. The double-blind trial against placebo (PL) Confirmed these results: 60% of the patients in PRZ group and 10% of the patients in PL group showed endoscopic ulcer healing after 2 weeks. This difference was significant. Ninety per cent of the PRZ patients and 50% of the PL patients were completely healed after 4 weeks. In the PRZ group, gastric secretory tests showed a significant decrease in B.A.O. (69%), M.A.O. (33%) and P.A.O. (34%) after 2 weeks. In the PL group, the same parameters had only a small decrease, without statistical significance. There were no pathological changes in laboratory findings in either the open or the double-blind studies. Mild and transient side-effects were observed in 7 of 30 patinets receiving pirenzepine (diplopia and dryness of the mouth).

Pirenzepine versus cimetidine in the treatment of duodenal ulcer.

Fifty-five patients with active duodenal ulcer have completed, up to today, a one-month double-blind trial to compare the effects of pirenzepine (150 mg daily), cimetidine (1 g daily) and placebo on the healing of duodenal ulcer. Fifteen (71%) of the 21 patients treated with pirenzepine had healed ulcers compared with 14 (82%) receiving cimetidine (P less than 0.05. In the placebo group there were 7 (41%) healed ulcers. Symptomatic improvement in patients given pirenzepine was as substantial as in cimetidine-treated patients. No adverse effects were noted.

The reversible depolymerization of spinach chloroplast glyceraldehyde-phosphate dehydrogenase. Interaction with nucleotides and dithiothreitol.

The ligand-dependent dissociation of spinach chloroplast glyceraldehyde-phosphate dehydrogenase (Mr 600000) to protomers of Mr about 145000, previously shown by us in 1973, has been further characterized by the technique of velocity sedimentation in sucrose gradients. The process exhibits cooperativity and is accompanied by an increase of the apparent NADP+-dependent activity (reactivation) from a ratio of 0.1-0.2 to a ratio of 1 to 2 with respect to the NAD+-dependent activity. In addition to NADP+ and NADPH, most nucleotide triphosphates and, to some extent, Pi, act as dissociating agents. The enzyme is depolymerized and progressively inactivated in the presence of 2'-AMP. 2. Incubation with 20 mM dithiothreitol or 8-10 mM GTP increases the apparent NADP/H)-dependent activity, although addition of a small amount of a dissociating compound, such as 0.06 mM NADP+, is required for depolymerization. 3. NAD+, NADH and, to a lesser extent, glyceraldehyde 3-phosphate, NMN and cyclic AMP act as inhibitors of the dissociation and reactivation, however induced. They also favour the reassociation of protomers to tetramers. 4. The NADP(H)-linked activity is probably a property of the protomers only. The system described here resembles in many respects the light-dependent regulation of the NADP(H)-linked activity in vivo.
