[Change in the reaction of circulating colony forming units to ACTH in thymectomized mice].

It has been discovered that unlike animals with the intact thymus, thymectomized adult (CBA X C57B1)F1 mice did not demonstrate any reduction in the number of circulating colony-forming units (CFUs) induced by the elevated concentrations of endogenous glucocorticoids 2 weeks to 5.5 months following operation. It is assumed that the effect of glucocorticoids on CFUs is mediated by the T1 lymphocyte subpopulation.

[Change in the proliferative activity of hematopoietic stem cells following adrenalectomy].

An early decrease in the total cellularity and amount of bone marrow CFUs was recorded in (CBAxC57B)F1 adrenalectomized mice. This decrease was accompainied by a significant fall in the number of proliferating stem cells in the bone marrow and spleen, evaluated by the method of "thymidine suicide". After normalization of the total amount of CFUs followed by that of the bone marrow cellularity the level of cycling stem cells returned to initial.

[Migration of hematopoietic stem cells in thermal burns].

Experiments on mice (CBA X C57BL) F1 have shown that a high level of endogenous glucocorticoids in the blood 30 min to 6 hours after burn is accompanied by a 2--2 1/2-fold decrease of the circulating CFU. At this period CFU migration from the bone marrow area shielded during the irradiation (850 R) is inhibited. The stem cells migration is increased 3--4 days after the burn.

The factors controlling stem cell recirculation. i. Migration of hemopoietic stem cells in adrenalectomized mice.

The influence of bilateral adrenalectomy on hemopoietic stem cell (CFU) migration in mice has been studied. Formation of endogenous spleen colonies in lethally irradiated, leg-shielded mice was sharply increased by prior adrenalectomy, and this increase was not dependent on the volume of shielded bone marrow. Adrenalectomy was shown to increase endogenous spleen colony formation in sublethally irradiated mice as well. However, it had no affect on formation of spleen colonies in lethally irradiated mice injected with syngeneic bone marrow. The CFU content of murine bone marrow decreased acutely after removal of the adrenals, and this decrease was accompanied by a concomitant increase in the peripheral blood and splenic CFU. Thus, adrenalectomy appeared to have no affect on the splenic plating efficiency or proliferative rate of hemopoietic stem cells, but it did result in increased migration of stem cells from the bone marrow to the blood, and thence to the spleen. It is concluded that the adrenal steroids may be of physiologic importance in the regulation of ehmopoietic stem cell migration.

The factors controlling stem cell recirculation. II. ACTH-induced inhibition of migration of hemopoietic stem cells.

Injections of ACTH of prolonged action (twice 1.5 units per mouse) given to lethally (800 R) irradiated mice with a hind limb shielded to the middle of the tibia brought about a twofold decrease in the number of spleen colonies. ACTH injections after sublethal whole-body irradiation (600 R) did not affect the number of endogenous spleen colonies. ACTH injection of normal mice brought about a substantial decrease in the CFU content in the circulating blood at the time of increased 11-hydroxycorticosteroid concentrations in the plasma. The results obtained are interpreted as inhibition of CFU migration from a shielded area of bone marrow induced by a high 11-hydroxycorticosteroid content in the plasma that followed ACTH injections which had no mitostatic effect on CFU proliferation.

[Factors controlling stem cell recirculation. 5. Modification of the effects of endogenous glucocorticoids on CFUs migration in T-deficient mice].

It was shown that at a continuously increased level of endogenous glucocorticoids (injection of ACTH) in thymectomized and B mice the degree of inhibition of CFUs migration, that was observed in T-deficient mice without ACTH injection, did not increase. With T-deficiency the stimulatory effect of the hypocorticoid state (adrenalectomy) on the CFUs migration persisted but was less pronounced than in animals with intact thymus.
