Effect of glycine on microcirculation in pial vessels of rat brain.

Single application of glycine in a final dose of 40 mg/kg to the surface of the parietal area of rat brain produced a potent vasodilatory effect. The diameter of arterioles increased to 250% from the baseline level 1-3 min after treatment. These changes persisted for 5-10 min. In the follow-up period the diameter of vessels progressively decreased to the baseline level. Repeated application of glycine in the same dose also induced dilation of arterioles. Application of physiological saline under similar conditions did not produce these changes.

Monoassociation with Lactobacillus acidophilus UFV-H2b20 stimulates the immune defense mechanisms of germfree mice.

Probiotics are formulations containing live microorganisms or microbial stimulants that have some beneficial influence on the maintenance of a balanced intestinal microbiota and on the resistance to infections. The search for probiotics to be used in prevention or treatment of enteric infections, as an alternative to antibiotic therapy, has gained significant impulse in the last few years. Several studies have demonstrated the beneficial effects of lactic acid bacteria in controlling infection by intestinal pathogens and in boosting the host's nonspecific immune response. Here, we studied the use of Lactobacillus acidophilus UFV-H2b20, a lactic acid bacterium isolated from a human newborn from Viçosa, Minas Gerais, Brazil, as a probiotic. A suspension containing 10(8) cells of Lactobacillus acidophilus UFV-H2b20 was inoculated into groups of at least five conventional and germfree Swiss mice to determine its capacity to stimulate the host mononuclear phagocytic activity. We demonstrate that this strain can survive the stressing conditions of the intestinal tract in vivo. Moreover, the monoassociation of germfree mice with this strain for seven days improved the host's macrophage phagocytic capacity, as demonstrated by the clearance of a Gram-negative bacterium inoculated intravenously. Monoassociated mice showed an undetectable number of circulating E. coli, while 0.1% of the original inoculum was still present in germfree animals. Mice treated with viable or heat-killed Lactobacillus acidophilus UFV-H2b20 presented similarly improved clearance capacity when compared with germfree controls. In addition, monoassociated mice had twice the amount of Kupffer cells, which are responsible for the clearance of circulating bacteria, compared to germfree controls. These results suggest that the L. acidophilus strain used here stimulates a nonspecific immune response and is a strong candidate to be used as a probiotic.

[Use of gnotobiotic animals for the study of the pathogenesis of cholera intoxication].

Germ-free suckling rabbits and minipigs can be recommended as models suitable for the study of different aspects of the pathogenesis of cholera intoxication. In minipigs, individual representatives of intestinal autochthonous microflora produce different effect on the sensitivity of the animals to the toxigenic and choleragenic action of Vibrio cholerae antigen introduced by oral administration, that should also be taken into consideration in the determination of residual toxicity during the trial of new vaccine preparations against cholera.

[Morphological study of the effect of cholera toxin on germ-free animals].

The effect of purified cholera toxin and the O-antigen of Vibrio cholerae inoculated separately into the intestines of germ-free suckling rabbits on the structure of the viscera and enzymatic changes in them was studied. The purified cholera toxin exerted mostly a local effect on the intestinal tract but caused lesions in the lymphoid tissue. The O-antigen was toxic causing primarily lesions in the liver and kidneys. Crude cholera toxin exerted a complex effect on gnotobiotic rabbits typical of the cholerogen and O-antigen.

[Age and peripheral blood and phagocytosis indices in gnotobiotic and ordinary Wistar rats].

Age-related features of the peripheral blood (red cell and leukocyte counts, hemoglobin level) and phagocytic activity of peritoneal macrophages were studied in experiments on gnotobiotic and conventional Wistar male rats aged 2, 5, 9 and 35 months. With aging, the conventional Wistar rats showed a significantly lower phagocytic activity in the presence of a progressively increasing leukocyte count as compared with the gnotobiotic animals. The red cell count and hemoglobin level dropped in the conventional species with age, while the gnotobiotic ones manifested no appreciable changes in the indices reviewed.

[Cyclic 3',5'-adenosine monophosphate in macrophages, intestinal mucosa, and blood plasma of germ-free and ordinary animals].

The level of cAMP in macrophages, intestinal mucosa and blood plasma as well as its formation in intestinal mucosa of germfree animals under the effect of lipopolysaccharide E. coli 055 (LPS) were studied in experiments on germfree and ordinary mice and guinea-pigs. The concentration of cAMP in intestinal mucosa of ordinary guinea-pigs was 5-fold higher than in germfree animals. LPS induced an increase in cAMP level in intestinal mucosa, but this level did not reach that in ordinary animals. The levels of cAMP in blood plasma of germfree guinea-pigs in macrophages of germfree mice increased 2-fold and 4-fold, respectively, 30 minutes following the treatment with LPS. The increased level of cAMP was accompanied by its intensive secretion into the exocellular medium. Macrophages of ordinary animals had a moderate output of cAMP. A conclusion is made about the relationship between cAMP formation and microbial contamination of the microorganism as well as about an important role of the cyclic nucleotide in the mechanism of nonspecific resistance and homeostatic reactions of the body to microbial exposure.

[Abacterial principle in surgery].

The authors dispose an experience with 250 surgical interventions with employment of abacterial insulators. The operations were carried out upon the abdominal organs, thorax, bones, vessels, urogenital organs etc. in neonates and elder children. A number of advantages of the surgical method with employment of abacterial insulators are ascertained.

[Age and changes in the splenic lymphoid tissue of germ-free rats during the postnatal period of ontogenesis].

In germfree rats the splenic lymphopoiesis is not demonstrated by the investigation to be suppressed, and lymphoid follicules undergo age changes. Up to 15 days, periarterial zone of T-lymphocytes, responsible for cell immunity, is determined, and by the 30th day, as in control animals, peripheral zone of B-lymphocytes, responsible for hormonal immunity, is distinctly observed. In 4-month-old germfree animals, a large amount of free iron crystals is detected in the spleen, while in the organ of control animals its amount is still small. In 10-month-old germfree rats, with the appearance of reactive centers, the zone of B-lymphocytes widens in lymphoid follicules and iron crystals integrate in the red pulp. Lymphopoiesis in the spleen of the germfree rats seems to be maintained by certain local conditions which are connected with increased haemolytic function of the organ. This produces a discharge of a large amount of the products of erythrocytosis which, like autoantigens, stimulate lymphopoiesis in the spleen even when microflora is absent in the organism, while lymphopoiesis in lymph nodes in germfree animals is sharply inhibited.

[Routes of E. coli 055 bacterial penetration through the intestinal wall in gnotobiotic and ordinary animals].

Peculiarities of permeability of intestinal barrier in germfree animals for enteropathogenic E. coli 055 were studied. Both germfree and conventional guinea pigs and rats were used. An increase of bacteriemia was revealed in gnotobiotes during the first day of oral E. coli 055 monocontamination; bacteriemia was transient in conventional animals. Under electron microscopy alterations of intercellular contacts and formation of spaces between enterocytes containing numerous microorganisms were found in the intestinal mucous membrane in gnotobiotes. Also more pronounced changes of microvessels of the intestinal mucous membrane were discovered in gnotobiotes. The processes of ingestionan and digestion of Escherichia coli by enterocytes and leukocytes were noted in conventional animals. The revealed derangements of the intestinal barrier in gnotobiotes explain the cause of higher bacteriemia in germfree animals. An important role of the microbial factor in the formation of intestinal barrier is indicated by the data obtained.

[Nonspecific resistance in germ-free and E. coli monocontaminated miniature piglets].

Phagocytic activity of leukocytes, as well as the complement, properdin, and lysozyme levels in the blood serum of miniature piglets, germfree and monocontaminated with E. coli 055 and E. coli 083, were studied. E. coli 055 phagocytosis was decreased in the presence of autologous serum and complement and increased under the effect of specific opsonins (antibodies to E. coli 055). Complement, properdin, and lysozyme levels were decreased in the germfree, in comparison with conventional animals. In the E. coli contaminated piglets properdin and complement production was stimulated most, and lysozyme formation--less. No antibodies to E. coli 055 were revealed in monocontaminated piglets. The highest lysozyme levels were found in the ex-germfree animals, this indicating the participation of factors other than E. coli contamination in lysozyme stimulation. It is concluded that microbial contamination played an important role in the development of cellular and humoral factors of the organism resistance.

[Obtaining and rearing germ-free miniature piglets for biomedical research].

The method of obtaining and rearing germfree miniature piglets for medical-biological research is described. The gnotobiotes were reared in rigid organic glass isolation cells up to 3 months. Milk diets supplemented with minerals and vitamins were used for nutrition. The gnotobiotes gained weight well during the whole observation period.

[Body temperature and the reaction to pyrogenal in germ-free and ordinary animals].

Body temperature, as well as pyrexia in response to pyrogenal in germfree and conventional mice and miniature piglets were studied. A decrease of the mean body temperature in the intact germ-free mice and miniature piglets in comparison with conventional animals of the corresponding species was revealed. The absence of marked pyretic response to pyrogenal after intraperitoneal injections of 10 minimal pyrogenic doses to mice and after intramuscular injections of 500 minimal pyrogen doses of pyrogenal to miniature piglets was observed in germfree animals. The data obtained indicated an important role of autoflora in the development of the organism capacity to temperature reaction and pyrexia.
