[Therapeutic effect of vegetable oils and ubiquinone-9 on radiation injuries].

The comparison was made of the protective (the administration 3 h before irradiation with a dose of 7.3 Gy) and therapeutic (the administration immediately and later after exposure) effects of soya oil (150 mg/kg) and oil solution of ubiquinone-9 (100-200 mg/kg) on survival of exposed rats. It was shown that soya oil and ubiquinone-9 increased the survival rate of rats when administered before and, to a lesser extent, immediately after irradiation. Corn oil administered immediately after exposure increased the survival rate as well. DMF for the therapeutic effect of soya oil solution of ubiquinone-9 was 1.08.

[Ubiquinone-9 of the flagellates Crithidia oncopelti and Astasia longa].

The content of ubiquinones (Co Q) of the Astasia longa and Crithidia oncopelti protozoa was studied. The protozoa were grown on an artificial nutrient broth. The cells were separated, washed, freeze-dried, and refluxed with KOH and pyrogallol in ethanol media. The hydrolyzate was concentrated. The residue was stored at -20 degrees and filtered. An ubiquinone fraction was isolated from the filtrate by TLC on silica gel. Identification of the ubiquinone homologues was carried out by reverse phase TLC and mass spectrometry. Ubiquinones were quantitated with respect to the difference in the density between the oxidized and reduced forms of Co Q at 275 nm. The A. longa and C. oncopelti flagellates were shown to contain ubiquinone-9 (Co Q9) at a concentration of 0.48 and 1.14 mumole/g dry cells, respectively. The higher Co Q level in zooflagellates as compared to that in phytoflagellates is discussed.

[Effect of native vitamin A metabolites and their 4-oxo derivatives on formation of specific cytolytic T lymphocytes].

Immunomodulating effect of some structural derivatives of retinoic acid (RA) on formation of specific cytolytic T lymphocytes (T killers) was studied in mixed cell culture of spleen lymphocytes from allogenic mice. Native vitamin A metabolites (RA, 13-cis-RA) and the structure-like substances (C15-C20-retinoids and others) at 10(-6) = 10(-7) M concentrations increased 2-2.5-fold the formation of T killers in vitro after stimulation of the lymphocytes by suboptimal doses of allogenic X-ray irradiated cells, the lymphocyte proliferation was inhibited in the mixed culture at 10(-5) M and higher concentrations of the substances. Optimal concentrations of retinoids did not increase the 3H-thymidine incorporation into DNA of lymphocytes stimulated by alloantigens or mutagens. The data obtained suggest that the previously found antitumoral efficiency of the compounds studied may be due to stimulation of specific cellular immunity.

[Toxicological and pharmacological properties of ubiquinone-9 and hexahydroubiquinone-4].

Experiments were made on random-bred and linear mice, Wistar rats, rabbits and dogs to examine pharmacological properties of ubiquinone-9 and hexahydroubiquinone-4. Both compounds (oily solutions) administered intraperitoneally, intragastrically and subcutaneously in single and multiple doses produced no changes in the behavior or status of the animals, which might have attested to their toxicity. They were found to exert a regulatory action on the central nervous system, decreasing its sensitivity to numerous narcotic and convulsant substances without appreciable effects on the blood, arterial pressure, ECG and diuresis. The main obstacle for comprehensive experimental study and application of these substances in medical practice lies in the absence of a perfect medicinal form.
