[Effectiveness of oxygen delivery to tissues during complete replacement of blood by bovine polyhemoglobin].

The study has been performed to test the hypothesis on feasibility of omitting pyridoxylation in production of bovine polyhemoglobin as compared to polyhemoglobin from human red cells. The model of a complete replacement of rabbit blood for polyglucin (Het approximately 2%) was used to compare efficacy characteristics for O2 delivery to tissues upon introduction of bovine polyhemoglobin against rabbit red cells. Immediately after the replacement, O2 release efficacy and that of circulation were identical, whereas 30 and 60 min after it, developing decompensation of hemodynamics indicated inability of bovine heterohemoglobin, unlike own red cells, to increase A-B difference by oxygen, probably due to metform rise.

[Treatment of experimental metabolic alkalosis by sorbamine].

A new infusion solution called alkamine has been developed for the treatment of metabolic alkalosis. It contains amino acids in the form of hydrochlorides (arginine, lysine and histidine hydrochlorides), potassium chloride and sorbit. The solution has been tested in anesthetized cats with experimental metabolic alkalosis induced by the injection of sodium hydrocarbonate solution. In control experiments without any therapy, a stable alkalytic shift of acid-base balance in the blood, a fall in the blood pressure, hypopotassemia and death of the animals within the first days were observed after experiments. The treatment of metabolic alkalosis by alkamine (25 experiments) entailed a complete correction of acid-base balance and hypopotassemia, stabilization of blood pressure and the survival of 23 out of the 25 animals. Alkamine solution has been approved for clinical trials by the Pharmacological Committee of the Ministry of Health of the USSR.

[Oxygen-dissociation properties and the regulation of the oxygen affinity of a hemoglobin polymer in the bull].

Oxygen-dissociation properties and some aspects of allosteric regulation of bovine hemoglobin (Hb) and its polymerized form (HbP) have been examined. It has been shown that bovine HbP as well as bovine Hb does not require the presence of organic phosphates for normal functioning and has gas transport characteristics which are similar to those of the whole human blood. Pyridoxal-5'-phosphate (PP) works as an allosteric regulator of bovine HbP only in the absence of Cl- ions and vice versa. However, in covalent binding of PP to HbP both the effectors (PP and Cl- ions) can work simultaneously and independently. Thus, by varying the Hb/PP ratio it is possible to prepare HbP solutions with preset P50 values within the range 30-55 mm Hg. The results can be used in simulation of gas transport characteristics of artificial oxygen carriers.
