Duodenal somatostatinoma with psammoma bodies.

A duodenal somatostatinoma was found incidentally in a 60-year-old woman undergoing cholecystectomy. Microscopically, the tumor had a glandular architecture with abundant psammoma bodies. Electron microscopy revealed tumor cells resembling D-cells of the pancreatic islets. Immunohistochemically, there was staining for neuron-specific enolase, chromogranin, and diffuse cytoplasmic positivity for somatostatin only. By immunoelectron microscopy, somatostatin was identified predominantly in lucent membrane-bound secretory granules. X-ray-dispersive microanalysis showed the psammoma bodies contained calcium apatite crystals. This case is compared with other reported cases with a description of cellular localization of somatostatin and development of psammomatous calcification.

Adenohypophysial changes in conventional, germ free and food-restricted aging Lobund-Wistar rats. A histologic, immunocytochemical and electron microscopic study.

The pituitaries of aging male Lobund-Wistar rats were investigated by histology, immunocytochemistry and electron microscopy. In contrast to previous studies undertaken on aging rats of various strains, no lactotroph hyperplasia was found in the adenohypophysis of the present study. Lactotroph adenomas producing PRL were infrequent, whereas gonadotroph adenomas containing FSH and LH were common. The frequency of gonadotroph adenomas was decreased in germ free as well as food-restricted rats and in those in which the germ free state was associated with reduced food intake.

Uptake of horseradish peroxidase in spontaneous adenomas of the rat pituitary.

The uptake of horseradish peroxidase was investigated by electron microscopy in 9 non-tumorous adenohypophyses and 13 spontaneous pituitary adenomas of adult female Long-Evans rats. Pituitary adenoma cells retained their ability for endocytosis and exhibited more extensive deposition of horseradish peroxidase than non-tumorous adenohypophysial cells.

Effect of bilateral nephrectomy on growth hormone cells of the rat pituitary. A histologic, immunocytologic and electron microscopic study including ultrastructural morphometry.

Adult male Long Evans rats were bilaterally nephrectomized and their adenohypophyses studied at various time intervals by histology, immunocytology, electron microscopy and ultrastructural morphometry. In addition, radioimmunoassay was carried out to establish blood growth hormone concentrations. Growth hormone cells appeared to be more conspicuous and fine structural morphometry revealed an increase in the volume density of their secretory granules. Radioimmunoassay showed elevation of blood growth hormone levels. Morphologic alterations were also noted in some corticotroph cells and gonadotroph cells. Many capillaries showed congestion. The perivascular spaces had widened and edema fluid accumulated in the perivascular spaces. The pathogenesis of the changes remains to be elucidated.

Calcium deposition in human pituitary adenomas studied by histology, electron microscopy, electron diffraction and X-ray spectrometry.

The incidence and morphogenesis of calcification were investigated by histology, electron microscopy, electron diffraction and X-ray spectrometry in 312 surgically removed human pituitary adenomas. By light microscopy the incidence of calcification was 9%. The calcium deposits were evident in several adenoma types, but were most frequent and extensive in prolactin-producing adenomas. Histologically calcium deposits represented amorphous precipitates and psammoma bodies staining with hematoxylin, the von Kossa technique and alizarin red. By electron microscopy, calcified deposits were noted in the cytoplasm of adenoma cells and extracellular space. Electron diffraction and X-ray spectrometry of intracellular precipitates as well as psammoma bodies showed the presence of phosphate, calcium and organically bound sulphur with a ratio of 3.5:5:2, indicating that they contained calcium hydroxyapatite. Present findings provide evidence that mitochondria with microcrystal deposition of calcium represent the primary sites of calcification.

Pituitary prolactin cell hyperplasia.

In female Long-Evans rats, development of prolactin cell adenomas is preceded by hyperplasia of prolactin cells, suggesting that factors affecting the endocrine function of prolactin cells might have an important role in the genesis of prolactin-producing pituitary tumors.

Autolytic changes in the rat adenohypophysis. A histologic, immunocytologic and electron microscopic study.

Thirty-nine adult female Long Evans rats were decapitated and the heads stored at room temperature. The pituitaries were removed at intervals from 30 minutes to seven days, fixed, embedded and studied by histology, immunocytology and electron microscopy. Histologically, changes were noticeable after two hours postmortem. Immunoperoxidase staining showed postivity for growth hormone, prolactin, FSH, LH and TSH up to seven days after sacrifice, appearing even stronger in the advanced stages of autolysis. Fine structural alterations were evident at 30 minutes and more conspicuous later. Changes included dilation, partial degranulation and whorl formation of RER, swelling of Golgi complexes and mitochondria, chromatin clumping, lysis, rhexis and pyknosis of nuclei, cytosegresome formation and disruption of cell membranes. Secretory granules remained well preserved throughout, although some exhibited fusion or reduced electron density. Dilation of capillaries with accumulation of erythrocytes, platelets and fibrin fibers were prominent findings. The severity of changes varied considerably from cell to cell indicating that the rate of autolysis is not the same among different cell types and is possibly affected by the actual functional state of the cell. It appears that increased membrane permeability and disruption of plasmalemma represent important steps in the autolytic process.

Enigmatic bodies in human corticotroph cells.

Fine-structural study of large, vacuole-like structures of the human corticotroph cell ('enigmatic body'; EB) revealed that these bodies have a preferential juxtanuclear position in the vicinity of the Golgi complex. EBs have rugged outlines, frequently a narrow electrondense rim and numerous membrane-bound compartments in an almost homogeneous matrix of low electron density. Fusion between EBs and secretory granules and other cytoplasmic components was noted. The peripheral rim and the matrix exhibited moderate to strong acid phosphatase activity. The inner compartments were consistently negative. It is proposed that EBs represent special lysosomes and serve as centers for recycling secretory material, as well as for storage and release of degraded products.

Lysosomal disposal of secretory granules and their transformation into pigment particles in spontaneous prolactin cell adenomas of the rat pituitary.

Light and electron microscopic study of spontaneous prolactin cell adenomas, found at autopsy of 26 aging female Long-Evans rats and of 9 aging male Wistar rats, revealed pigment particles in the cytoplasm of some adenoma cells. It is postulated that most of the pigment particles derive from secretory granules. The presence of crinophagy and autophagy are interpreted as the morphologic expressions of lysosomal activation. The causative mechanisms accounting for lysosomal disposal of secretory granules and their transformation to pigment particles remain to be elucidated.
