Accounting for outliers and heteroskedasticity in multibreed genetic evaluations of postweaning gain of Nelore-Hereford cattle.

The objectives of this study were to demonstrate the utility of hierarchical Bayesian models combining residual heteroskedasticity with robustness for outlier detection and muting and to evaluate the effects of such joint modeling in multibreed genetic evaluations. A 3 x 2 factorial specification of 6 residual variance models based on several distributional (Gaussian, Student's t, or Slash) and variability (homoskedastic or heteroskedastic) assumptions was used to analyze 22,717 postweaning gain records from a Nelore-Hereford population (40,082 animals in the pedigree). To illustrate the utility of the 2 robust distributional specifications (Student's t and Slash) for outlier detection and muting, 3 records from the same contemporary group (an extreme residual outlier, a mild residual outlier, and a near-zero residual) were chosen for further study. The posterior densities of the corresponding weighting variables of these records were used to assess their degree of Gaussian outlyingness and the ability of the robust models to mute the effects of deviant records. The Student's t heteroskedastic provided the best-fit model among the 6 specifications and was preferred for genetic merit inference. Kendall rank correlations of the posterior means of the additive genetic effects of the animals, used to compare the selection order of the Student's t and Gaussian models, were reasonably high across all animals within the most frequent genotypes, ranging from 0.83 to 0.91 and from 0.89 to 0.95 for the homoskedastic and the heteroskedastic versions, respectively. However, when considering only animals ranked in the top 10% by the customary Gaussian homoskedastic model, these rank correlations were reduced considerably, ranging from 0.29 to 0.57 and from 0.72 to 0.85 between the 2 residual densities within the homoskedastic and heteroskedastic versions, respectively. Rank correlations between the homoskedastic and heteroskedastic versions within each of the Gaussian and Student's t error models tended to be smaller, with a range from 0.68 to 0.90 across all animals and from 0.28 to 0.67 for animals ranked in the top 10%. These results support the implementation of robust models accounting for sources of heteroskedasticity to increase the precision and stability of multibreed genetic evaluations with proper statistical treatment of deviant records.

Analysis of the bovine neutrophil transcriptome during glucocorticoid treatment.

The objective of this study was to characterize a large portion of the bovine neutrophil transcriptome following treatment with the anti-inflammatory glucocorticoid dexamethasone (Dex). Total RNA was isolated from blood neutrophils of healthy cattle (5 castrated male Holsteins) immediately following cell purification (0 h) or after ex vivo aging for 4 h with or without added Dex. Additional neutrophils were cotreated with a glucocorticoid receptor (GR) antagonist (RU486) and Dex for 4 h. RNA was amplified, dye labeled (Cy3 or Cy5), and hybridized to a series of National Bovine Functional Genomics Consortium (NBFGC) microarrays. LOWESS data normalization followed by mixture model analyses showed that 11.15% of the spotted NBFGC cDNAs (2,036/18,263) were expressed in 4-h (untreated) neutrophils. Subsequent two-step mixed-model analysis detected (P < or = 0.05) 1,109 differentially expressed genes, of which contrast analysis indicated those that were independently responsive to aging (1,064), Dex (502), RU486 + Dex (141), or RU486 (357). In silico analysis revealed that 416 of the differentially expressed genes are unknown, 59 did not cluster well based on known function, and 634 clustered into 20 ontological categories. Independent validation of differential expression was done for 14 of the putatively Dex-responsive genes across these categories. Results showed that Dex induced rapid translocation of GR into the neutrophil nucleus and signaled dramatic alterations in expression of genes that delay apoptosis, enhance bactericidal activity, and promote tissue remodeling without inflammation or fibrosis. Thus these findings revealed hitherto unappreciated plasticity of blood neutrophils and potentially novel anti-inflammatory/wound-healing actions of glucocorticoids.

Glucosamine and chondroitin sulfate regulate gene expression and synthesis of nitric oxide and prostaglandin E(2) in articular cartilage explants.

OBJECTIVE: Glucosamine (GLN) and chondroitin sulfate (CS) are widely used to alleviate symptoms of osteoarthritis (OA). However, the mechanism(s) of action of these nutraceuticals remains unresolved. In the present study, we determined the effect of physiologically relevant concentrations of GLN and CS on gene expression and synthesis of nitric oxide (NO) and prostaglandin E(2) (PGE(2)) in cytokine-stimulated articular cartilage explants. METHODS: Using bovine articular cartilage explants in culture stimulated with IL-1, the effects of physiologically relevant concentrations of GLN and CS on gene expression of inducible nitric oxide synthase (iNOS), endothelial nitric oxide synthase (eNOS), cyclooxygenase-2 (COX-2) and microsomal prostaglandin E synthase-1 (mPGEs1) were assessed with quantitative real-time polymerase chain reaction (Q-RT-PCR). The production of NO and PGE(2) was also quantified. RESULTS: CS and the GLN and CS combination at concentrations attainable in the blood down-regulated IL-1 induced mRNA expression of iNOS at 24 and 48 h post-culture. Up-regulated iNOS expression at 24h by IL-1 was also suppressed by GLN. GLN and CS transiently repressed the cytokine-stimulated mPGEs1 transcript. Synthesis of NO was reduced with CS alone and the combination after 24h of culture. Repression of COX-2 transcripts by GLN and CS was accompanied by concomitant reduction in PGE(2). CONCLUSION: Our results indicate that physiologically relevant concentrations of GLN and CS can regulate gene expression and synthesis of NO and PGE(2), providing a plausible explanation for their purported anti-inflammatory properties.

Gene expression profile of mechanically impacted bovine articular cartilage explants.

Traumatic injury to a joint can initiate cartilage degradation. Blunt trauma increases matrix damage and decreases proteoglycan synthesis in in vitro models. Few studies have investigated gene expression of articular cartilage (AC) following mechanical loading. Recent advances in microarray technology allow analysis of a number of genes, and may elucidate pathways of AC degradation. In the present study, we used a bovine cDNA microarray to determine how acute trauma of cartilage explants in the absence of underlying bone alters gene expression. Results indicate that at least 19 genes were differentially expressed at 3 h after trauma. Fourteen genes were up-regulated and five genes were down-regulated relative to control explants. The up-regulated genes included cytokine and chemokine receptors, enzymes, and molecules involved in signal transduction. Genes of adhesion molecules and apoptosis were down-regulated. The results of this study highlight the potential benefits of using a bovine cDNA microarray to study cartilage metabolism.

Multiple-breed genetic inference using heavy-tailed structural models for heterogeneous residual variances.

Multiple-breed genetic models recently have been demonstrated to account for the heterogenous genetic variances that exist between different beef cattle breed groups. We extend these models to allow for residual heteroskedasticity (heterogeneous residual variances), specified as a function of fixed effects (e.g., sex, breed proportion, breed group heterozygosity) and random effects such as contemporary groups (CG). We additionally specify the residual distributions to be either Gaussian or based on heavier-tailed alternatives such as the Student's t or Slash densities. For each of these three residual densities using either homoskedastic (homogeneous variance) or heteroskedastic error specifications, we analyzed 22,717 postweaning gain records from a Nelore-Hereford population based on a Markov chain Monte Carlo animal model implementation. The heteroskedastic Student's t error model (with estimated df = 7.33 +/- 0.48) was clearly the best-fitting model based on a pseudo-Bayes factor criterion. Breed group heterozygosity and, to a lesser extent, calf sex seemed to be marginally important sources of residual heteroskedasticity. Specifically, the residual variance in F1 animals was estimated to be 0.70 +/- 0.16 times that for purebreds, whereas the male residual variance was estimated to be 1.13 +/- 0.09 times that for females. The CG effects were important random sources of residual heteroskedasticity (i.e., the coefficient of variation of CG-specific residual variances was estimated to be 0.72 +/- 0.06). Purebred Nelores were estimated to have a larger genetic variance (124.84 +/- 21.75 kg2) compared with Herefords (40.89 +/- 6.70 kg2) under the heteroskedastic Student's t error model; however, the converse was observed from results based on a homoskedastic Student's t error model (46.24 +/- 10.90 kg2 and 60.11 +/- 8.54 kg2, respectively). These results indicate that allowing for robustness to outliers and accounting for heteroskedasticity of residual variances has potentially important implications for variance component and genetic parameter estimates from data on multiple-breed populations.

Comparison of ovarian function and circulating steroids in estrous cycles of Holstein heifers and lactating cows.

Ovarian function was compared between nulliparous heifers (n = 29; 10 to 16 mo old) and lactating Holstein cows (n = 31; 55.9 +/- 3.5 d postpartum). Follicular dynamics, corpus luteum growth, and regression, and serum steroid concentrations were evaluated through ultrasonography and daily blood sampling. Most heifers (27 of 29) but only 14 of 31 cows had typical spontaneous estrous cycles after cycles were initiated. Twelve cows had atypical cycles, and 5 became anovulatory during the study. The 12 cows with atypical estrous cycles had low serum estradiol after luteolysis and failed to ovulate the dominant follicle present at luteolysis. Heifers and cows with typical cycles were compared directly. Interovulatory intervals were similar between heifers (22.0 +/- 0.4 d) and cows (22.9 +/- 0.7 d). Those animals had estrous cycles with either 2 (15 heifers; 11 cows), 3 (9 heifers; 2 cows), or 4 follicular waves (3 heifers; 1 cow). Cows ovulated later after luteolysis than heifers (5.2 +/- 0.2 vs. 4.6 +/- 0.1 d, respectively), and had more multiple ovulations (17.9 vs. 1.9%). Maximal serum estradiol concentration preceding ovulation was lower in cows than in heifers (7.9 +/- 0.8 vs. 11.3 +/- 0.6 pg/mL) even though ovulatory follicles were larger in cows (16.8 +/- 0.5 vs. 14.9 +/- 0.2 mm). Similarly, maximal serum progesterone concentration was lower for cows (5.6 +/- 0.5 vs. 7.3 +/- 0.4 ng/mL), whereas maximal volume of luteal tissue was larger for cows than heifers (11,120 +/- 678 vs. 7303 +/- 308 mm3). Thus, higher incidence of reproductive anomalies in lactating cows, such as low conception rate, ovulation failure, delayed ovulation, and multiple ovulations, may be due to lower circulating steroid concentrations in spite of larger ovulatory follicles and luteal structures.

Improvement in recovery of embryos/ova using a shallow uterine horn flushing technique in superovulated Holstein heifers.

The aim of this study was two-fold: (1). to compare recovery of embryos/ova from superovulated Holstein heifers by flushing the uterine horns through insertion of the catheter very close to the tip of the horn (deep) or just after the uterine bifurcation (shallow) and (2). to evaluate the hormonal and superovulatory response to estradiol benzoate (EB) treatment prior to superovulation. Ten Holstein heifers (12-16 months) underwent two superovulatory treatments in a cross-over design. Heifers were treated with decreasing doses of FSH from Days 8 to 12.5 of a synchronized estrous cycle. At 4 days prior to superovulation, half of the heifers received EB (5mg, i.m.) or served as Controls, followed by the alternative treatment in the subsequent superovulation. At embryo recovery, one uterine horn was flushed with deep ( approximately 7 cm caudal to the tip of the horn) and the other with shallow ( approximately 5 cm cranial to the beginning of the uterine bifurcation) flushing techniques. Embryos/ova were recovered, counted, and scored. Number of ovulations was estimated by ultrasound. Pretreatment with EB reduced circulating FSH and regressed the first wave dominant follicle with no change in number of large follicles, number of ovulations, number of embryos/ova recovered, or number of transferable embryos. The shallow flushing technique was superior to the deep technique for number of embryos/ova recovered per horn (5.4+/-1.1 versus 3.9+/-0.8) or percentage of embryos/ova recovered per CL (63.9+/-8.6% versus 37.4+/-6.5%). Thus, flushing the entire uterine horn increased recovery of embryos/ova.

Ovarian structures and circulating steroids in heifers and lactating cows in summer and lactating and dry cows in winter.

Two experiments compared follicular and luteal development and circulating steroid concentrations from induced luteolysis to ovulation in lactating Holstein cows (n = 27; 40.0 +/- 1.5 kg milk/day) vs. nulliparous heifers (n = 28; 11 to 17 mo-old) during summer (Experiment 1), and in lactating (n = 27; 45.9 +/- 1.4 kg milk/d) vs. dry cows (n = 26) during winter (experiment 2). All females received PGF2,, 6 d after ovulation and were monitored until next ovulation by daily ultrasound and assay of serum progesterone (P4) and estradiol (E2). Every female was used two or three times. In Experiment 1, lactating cows had high incidence of multiple ovulation (63.5%) compared with heifers (1.3%). Among single ovulators, there was no difference in maximal size of ovulatory follicles between lactating cows and heifers (15.8 vs. 16.5 mm, respectively). However, lactating cows had lower peak serum E2 (8.6 vs. 12.1 pg/ml), took longer to ovulate after luteolysis (4.6 vs. 3.8 d), developed more luteal tissue volume (7,293.6 vs. 5,515.2 mm3), and had lower serum P4 on d 6 after ovulation (2.0 vs. 3.0 ng/ml) than heifers (data included multiple ovulators). In experiment 2, multiple ovulations were similar between lactating and dry cows (17.9 vs. 17.2%, respectively). Peak serum E2 was also similar between lactating and dry cows (7.6 vs. 8.5 pg/ml) although lactating cows had larger ovulatory follicles (18.6 vs. 16.2 +/- 0.4 mm). Lactating cows took longer to ovulate (4.8 vs. 4.2 d), developed more luteal tissue (7,599 vs. 5,139 +/- 468 mm3), but had similar serum P4 (2.2 vs. 1.9 ng/ ml) compared with dry cows. Therefore, lactating cows had similar or lower circulating steroid concentrations than dry cows or heifers, respectively, despite having larger ovarian structures.

Electromyographic validation of the trapezius and serratus anterior muscles in the rowing and frontal-lateral cross, dumbbells exercises.

The trapezius and serratus anterior muscles were studied in four modalities of rowing exercises executed with two grips, middle and closed, in comparison with the four different modalities of frontal-lateral cross, dumbbells exercise. It was used 24 male volunteers, 18 to 25 years old using a 2-channel TECA TE 4 electromyograph and Hewlett Packard surface electrodes. The results showed that TS acted in a higher significant way in all the modalities of rowing than in the supine lateral raise, inclined supine lateral raise and reverse supine lateral raise, dumbbells exercices, and demonstrated no standing frontal-lateral cross, dumbbells. The SI acted more significantly difference among all the execution modalities of rowing and the inclined supine lateral raise, dumbbells exercises than in all the rowing exercises; even though the activity levels do not make us suggest them as an excellent group of exercises for the development of this muscle.

Electromyographic validation of the trapezius and serratus anterior muscles in rowing exercises with closed grip.

Because the lack of specialized textbooks, the select of basic exercises for physical conditioning programmes is based on empirical knowledge. This fact led the authors to propose the study on electromyographic activity of the trapezius (upper portion) (TS) and the serratus anterior (lower portion) (SI) muscles in rowing exercises with closed grip in three different modalities. The tests were carried out with 24 male volunteers, 18 to 25 years old, by using a two-channel TECA TE 4 electromyograph and Hewlett Packard surface electrodes. For exercises execution, a long bar made of light wood was used. TS acted significantly in the three different modalities, that is, upright, sitting and bent over, while SI acted preferentially in upright and sitting rowing exercises, justifying their inclusion as basic exercises in physical conditioning programmes.

Electromyographic validation of the trapezius and serratus anterior muscles in frontal-lateral cross, dumbbells exercises.

Based on the lack of electromyographic researches on sport and programmes of physical conditioning, we can say that it is necessary to reexamine some exercises routinely used in the programmes of physical conditioning. Thus, the trapezius and serratus anterior muscles were studied electromyographically so that we could evaluate the validity in some ways of execution of the frontal-lateral cross, dumbbells exercises for the development of these muscles. We analyzed 24 male volunteers, 18 to 25 years old, using a 2-channel TECA TE 4 electromyograph and Hewlett Packard surface electrodes. For the execution of the exercise it was used a supine bench, a straight board and two bars of 40 cm made of light wood. The results showed that TS acted preferentially in standing modality and in the inclined supine modality, however with activity levels that do not justify its inclusion in physical fitness programmes.

Electromyographic validation of the trapezius and serratus anterior muscles in rowing exercises with middle grip.

Because the lack of textbooks based on electromyographic studies in the area of exercises for physical conditioning, the authors proposed to study the trapezius (upper portion) (TS) and the serratus anterior (lower portion) (SI) muscles during the execution of four different modalities of rowing exercises with middle grip in 24 male volunteers, 18 to 25 years old. For the recordings, it was used a two-channel TECA TE 4 electromyograph and Hewlett Packard surface electrodes. For the movements, a supine bench and a 120 cm-long bar made of low weight wood were used. The results showed that TS acted significantly in upright, sitting and inclined rowing, justifying its inclusion in physical conditioning programmes, while SI, in spite of acting preferentially in upright and sitting rowing, presented activity levels which do not justify its inclusion.
