Viscous gel containing 2.5 mg of PG F2 alpha in 10 ml was locally applied for cervix maturation to 106 primigravidae, prior to termination of pregnancy. Spontaneous abortion occurred to 33 women. Cervix maturation was up to expectation in 66 cases, and it was not in seven cases. Side-effects were of minor importance. The method is considered to be highly suitable for nulliparous women.

We investigated in vitro accumulation of adenosine 3',5'-monophosphate (induced by 5-hydroxytryptamine and dopamine) and of guanosine 3',5'-monophosphate (induced by acetylcholine) in the anterior byssus retractor muscle of Mytilus. The response to 5-hydroxytryptamine exceeded that induced by equimolar concentrations of dopamine. 1-methyl lysergic acid, a 5-hydroxytryptamine-blocking agent, diminished the 5-hydroxytryptamine-induced increase of cyclic AMP level. This parallels the effect of this amine on the contracted muscle. Acetylcholine, which causes a tonic contraction of the muscle, increased intracellular levels of cyclic GMP in a dose-dependent (max. 45-fold at 10(-4) M ACh) manner. The time course of the rise in cyclic GMP level was rapid and transient (peak concentration of cyclic GMP at 2 min). Mytolon was the most effective of all cholinergic blockers tested. It was concluded that cyclic nucleotides may play a role in the modulatory process of the transmitters. A direct relation to the relaxation-contraction process could not be established.
Injection in normal mice of IgM antibodies against sheep erythrocytes or dextran, in the form of an immune serum depleted og IgG, induces direct plaque-forming cells of the same specificity as the injected antibodies. The response is 10--70 times higher than the background plaque-forming cell titer of untreated control mice. Nanogram amounts of IgM induce a detectable response, and a ceiling is reached with a few hundred nanograms of monoclonal IgM. The inducing agent is not residual antigen: (i) treatment of the injected material and the recipients with dextranase abolishes the immunogenicity of dextran, but not the response to anti-dextran IgM; (ii) monoclonal IgM specific for sheep erythrocytes or trinitrophenyl likewise induces plaque-forming cells of the respective specificity, but variant hybridoma products (in which the light chain is that of the myeloma parent) are inactive. In normal mice, IgM-induced antibody responses were observed with antibodies to both thymus-dependent and thymus-independent antigens, but such could not be induced in athymic (nude) mice. Because the mechanism underlying this phenomenon would operate also in a normal immune response and, presumably, in the normal dynamic state of the immune system of unstimulated animals, a network regulation among the elements of the immune system itself is implied.
Hybrid cells secreting one, two, or three different immunoglobulins were constructed. The loss of immunoglobulin heavy or light chain expression was monitored. Chain loss was random only in lines with an excess of active light chain genes over heavy chain genes. In all other combinations preferential heavy chain loss was observed. Variant cells altered in heavy or light chain synthesis exhibited an altered chain loss pattern. It is therefore proposed that free immunoglobulin heavy chain is toxic for the cells. The interdependence of the two gene products gives a possible molecular explanation of apparent directed chromosome loss in hybrid cells.
Immunoglobulin was obtained from a hybridoma cell line, which was a reclone of a hybrid between a rabbit cell and mouse myeloma cell (X63-Ag8). The immunoglobulin, isolated from the cell culture medium was found to be homogeneous by isoelectrofocusing and immunoelectrophoresis and consisted of mouse heavy and rabbit light chains, linked by disulphide bonds. All immunoglobulin molecules carried both mouse and rabbit determinants; mouse determinants were associated only with the heavy chains while rabbit determinants were only associated with the light chains. The rabbit light chains were of Ab4 allotypical specificity, but possess only some of the Ab4 determinants normally present in Ab4/Ab4 animals. It was suggested that the restriction in allotypical specificity may be a general property of light chain Ab allotypes; the normal serum immunoglobulins may be heterogeneous with respect to the allotypic determinants and any one molecule may possess only a proportion of determinants detected by a conventional anti-allotype antiserum.
In a clinical trial on 12 healthy volunteers total gastrointestinal blood loss was measured with 51Cr-labelled erythrocytes. This double-blind cross-over study showed a highly significant difference (p < 0.001) between indometacin and [1-(p-chlorobenzoyl)-5-methoxy-2-methylindol-3-acetoxy[acetic acid (acemetacin, TV 1322, Rantudil¿) after oral application of 200 mg acemetacin, 200 mg indometacin per day and placebo for a week. Average total blood loss within seven days was 12.4 ml after indometacin, 4.5 ml after acemetacin and 5.0 ml after placebo. In a clinical trial on 16 rheumatic patients taking commercial non-steroidal antiinflammatory drugs over a long time total gastrointestinal blood loss was determined likewise. After a preceding wash-out period the blood loss due to acemetacin (240 mg daily) and indometacin (200 mg daily) for seven days was compared. There was a highly significant difference (p < 0.001) between both drugs. Total weekly blood loss was measured as 13.8 ml due to the treatment with indometacin and 6.5 ml due to acemetacin. During the wash-out period blood loss was detected as 6.0 ml weekly. There was no difference between the results of both trials. These clinical trials on 35 patients and volunteers do not give any indication that the new antiinflammatory drug acemetacin may cause mucosal lesions in the gastrointestinal tract evedenced by blood loss.
Hormonal contraception is at present the safest method to prevent pregnancy. In this survey the most important aspects of side-effects, drug interactions contraindications and indications for discontinuance are dealt with.
Metabolism and pharmacokinetics of [1-(p-chlorobenzoyl)-5-methoxy-2-methylindole-3-acetoxy]-acetic acid (acemetacin, TV 1322, Rantudil) in comparison to equimolar doses of indometacin was investigated in human volunteers after a single oral application and in rheumatic patients after multiple application. After multiple application (t.i.d. for 10 days) of equimolar doses of acemetacin and indometacin, mean blood level curves. The bioavailability of acemetacin derived from these data, corresponds to that of indometacin, i.e., approx. 100%, whereas after a single dose bioavailability of acemetcin is found to be smaller (66% from blood level, 64% from urine). This difference between single and multiple application is explained by slow filling-up of compartments. Degradation of acemetacin occurs by esterolytic cleavage to indometacin, by O-demethylation and N-desacylation and by partial conjugation of all these compounds to glucuronic acid. Blood level ratios of acemetacin and indometacin are equal after single and after multiple application of acemetacin. Therefore, degradation of acemetacin is not induced after multiple application. After a steady-state has been reached half-life of elimination is 4.5 +/- 2.8 h, which is longer than for indometacin (2.2 +/- 0.5 h). Interindividual differences are in the same range as described for indometacin.

Under the picture of acute inflammatory events of adnexa there may be sometimes be hidden diseases of the intestines. Out of them acute state of ileus is of utmost importance. State post appendectomy, atypical anamnesis and antiinflammatory treatment without effect is suspect of an ileus. An early laparotomy presents a low risk in such cases.
It is reported on experiences with 138 Cooper-T-200 IUDs, resulting in 1868 months of use. Both the Pearl-Index with 1,3 and the expulsion rate with 1,44% are no significant lower, than in a compared DANA-Super-IUD collective. In both groups the rate of all events like pains, spottings and hypermenorrhea is almost identically. Hypermeorrhea is of significant frequenter occur in the Copper-T-IUDs.

We have investigated immunoglobulin production by cloned hybrid cells. gamma2b heavy chains, but not gamma1 or gamma21, exist in two forms, differing in molecular weight, galactose labeling and charge. Hybrid molecules containing gamma2b and gamma1 heavy chains were secreted by hybridoma cells synthesizing Ig molecules of both subclasses. However, cells synthesizing both IgM and IgG chains did not secrete mu-gamma hybrid molecules. We have surveyed many new hybridomas derived from different mouse strains: 10% of those secreting IgM produce a mu chain which is about 5000 daltons larger than normal mu. Two IgM-secreting lines were studied in detail and were found to bear mu but not delta in their membrane. Intra- and extracellular mu differ in molecular size; pentameric IgM-containing molecules of both sizes were observed in the membrane fraction.
