[Endo beta-N-acetylglucosaminidase in human liver].

Endo beta-N-acetylglucosaminidase activity was found in a alpha-L-fucosidase preparation purified from human liver homogenates, using glycopeptides from human parotid glycoprotein as a substrate. An oligosaccharide was purified and after reduction N-acetylglucosaminitol was shown by paper chromatography. Thus, the specificity and the significance of this endoglycosidase in glycoprotein catabolism are discussed.

Heterogeneity of the carbohydrate chains of sulfated bronchial glycoproteins isolated from a patient suffering from cystic fibrosis.

Sulfated glycoproteins having blood group H activity were isolated from the sputum of a child suffering from cystic fibrosis, by reduction of the fibrillar mucus, chromatography on ECTEOLA-cellulose, and gel filtration on Sepharose 4B. The sulfated glycoproteins were degraded with alkaline borohydride, and the degradation products were fractionated by chromatography on ion exchange resins and by gel filtration. The carbohydrate chains thus obtained have a wide heterogeneity with regard to acidity and molecular size. The neutral chains contain blood group H active oligosaccharides and incomplete chains as short as 1 residue of 2-acetamido-2-deoxy-D-galactose. The minimal size of the neuraminic acid-containing chains is less than that of the sulfated chains, which increases with the degree of sulfation. The sulfate groups are linked at C-6 at the D-galactose residues.

[Study of individual variations of the composition of human parotid saliva collected under stimulation].

Human parotid saliva was studied for its qualitative and quantitative composition; alpha-amylase activity, immunoglobulin A and composition were investigated and showed significant variations from one individual sample to another. The same phenomenon was observed with the inorganic components: CA, P, K, Mg, Na, Cl. The influence of the frequency and length of collection of parotid saliva is discussed.

Effect of radiotherapy on pelvic lymph node metastasis in cervical cancer stages IB2-IVA: a retrospective analysis of two comparative series.

Efficiency of radiotherapy in controlling lymph node metastasis is a controversial issue. A continuous series of 87 patients affected by cervical cancer stages IB2-IVA and treated using pelvic radiotherapy is presented. A retrospective comparison is made between two populations. In the two populations, a staging lymphadenectomy was carried out before the onset of the therapeutic program. In the first population (53 patients), the pelvic nodes only were dissected and in the second one (34 patients), the pelvic lymph nodes were left in place and the paraaortic nodes only were dissected. In both series, a completion surgery was performed after finalization of the radiotherapy. It was carried out at open abdomen in both series. It included a systematic pelvic dissection for the patients whose pelvic nodes had been intentionally left in place at the time of the initial staging lymphadenectomy. Both series were identical as far as classic risk factors were concerned (FIGO stage, maximal tumor diameter, lymphovascular space involvement). The radiotherapy administered to the pelvis was the same in both populations. The number of patients with pelvic lymph node metastasis was 21 (39.6%) in the first population versus 6 (17.6%) in the second one (P = 0.03). The percentage of positive lymph nodes among the retrieved lymph nodes was 18.94 in the first population versus 2.8 in the second one (P = 0.0001). Pelvic radiotherapy is likely to control most of the pelvic lymph node metastasis, but not all of them. Practical deductions and further developments are discussed.

[Polymorphism and glycoprotein character of Ricinus communis lectins purified by affinity chromatography].

Two lectin fractions (S20W = 6,8 and 4,9 S) were purified from Ricinus communis seeds. The purification was carried out in four steps : ammonium sulfate fractionation, affinity chromatography on Sepharose 4 B, gel filtration on Sephadex G 150 and chromatography on CM celluloes. The purified lectins were glycoproteins whose chemical composition was determined. Amino terminal analysis of the two fractions revealed glycine and serine. Polyacrylamide gel electrophoresis of the higher molecular weight fraction allowed the separation of several components with different affinity for PAS staining.
