Molecular cloning of Renibacterium salmoninarum DNA fragments.

A Renibacterium salmoninarum enriched recombinant DNA library was constructed to isolate DNA fragments which could be used as probes to detect gene sequences specific for the causative agent of bacterial kidney disease in salmonid fish. One fragment of 149 base pairs was isolated and its specificity and sequence determined. This probe may prove useful in the design of diagnostic tests for the disease in asymptomatic fish and ova.

[Gastric neurofibroma. Report of 2 cases].

Two patients consulting for upper gastrointestinal bleeding, associated to impaired gastric emptying in one, were studied with X rays and endoscopy. A gastric neurofibroma was confirmed by histologic study in both patients. One patient was submitted to subtotal gastrectomy. An endoscopic resection of the tumor was performed in the other.

[Post-transfusional hepatitis. Preliminary study].

In Venezuela post-transfusional hepatitis (PTH) data is unknown. We studied, prospectively, 147 patients who received blood transfusions (1 to 48 different blood components). They were screened for HBsAg (Ortho HBsAg Elisa Test System), anti HBc (Ortho HBc Elisa Test System and CORZYME, Abbott Laboratories) and amino-transferases (Doles), before and after transfusions. Only the last 36 patients were tested for anti HCV (Ortho HCV antibody Elisa Test)R from the beginning. The test were performed at 2, 7, 12 and 24 weeks subsequently in the negative recipients. PTH for type B virus was 3.8% (5/131), and for NANBH 2.7% (3/110). In both groups those recipients who had more transfusions had the greatest incidence of PTH. We conclude that anti HBc and ALT must be done in all blood donors to prevent the occurrence of PTH either B virus or NANBH. We suggest also, to include anti HCV screening in blood donors as a mean to make blood transfusion safer.

Antigens in electron-dense granules from Entamoeba histolytica as possible markers for pathogenicity.

In vitro interaction of Entamoeba histolytica with collagen induces intracellular formation and release of electron-dense granules (EDG) and stimulation of collagenolytic activity. Purified EDG contain 1.66 U of collagenase per mg of protein. Thus, EDG may participate in tissue destruction during invasive amebiasis. Monoclonal antibodies (MAbs) L1.1 and L7.1 reacted specifically with EDG in enzyme-linked immunosorbent assay (ELISA) and immunofluorescence and immunoelectron microscopy. MAb L7.1 immunoprecipitated three polypeptides with molecular weights of 95,000, 68,000, and 28,000 from lysates of biosynthetically labeled E. histolytica. Both MAbs recognized the pathogenic E. histolytica axenic strains HM1:IMSS, HM38:IMSS, and HK-9 but failed to react in ELISA with Entamoeba moshkovskii, Entamoeba invadens, and E. histolytica-like Laredo. In addition, MAb L7.1 reacted with one E. histolytica isolate from a symptomatic patient but did not react with four of five isolates from asymptomatic patients. EDG antigens were detected by a MAb L7.1-based ELISA in E. histolytica-containing fecal samples from symptomatic, but not asymptomatic, individuals. These results suggest that the EDG antigen detected with MAb L7.1 may be differentially expressed in pathogenic and nonpathogenic E. histolytica.

[Use of a specific DNA probe to confirm sickle cell anemia in a caucasian woman].

A specific DNA probe containing part of the structural B-globulin gene was used to confirm the diagnosis of sickle cell anemia in a caucasian woman. The patient's genomic DNA was digested with the restriction endonuclease Dde I, fractioned by agarose electrophoresis and Southern blotting. Molecular hybridization was performed with the DNA probe prepared by chemical labelling with photobiotin. The beta 8/beta 8 genotype rendered only 1 fragment of length 376 bp. Upon digestion with Dde I, the DNA of an individual with the normal genotype containing the enzyme recognition sequence at the site of sickle cell mutation, resulted in 2 fragments of 201 and 175 bp. The pedigree of the patient's caucasian family was studied by Hb electrophoresis. Four out of 7 brothers carried the sickle cell trait.

[Preservation of blood platelets concentrates at different temperatures].

The temperature storage of the platelet concentrates (PC) it's of utmost importance in order to preserve the viability and the hemostatic function of the platelets. In the following study we made a comparative evaluation of the PC stored at three different temperatures: Volume, platelet count, pH, pCO2, pO2, in vitro aggregation, bacterial growth, ultrastructure changes and survival, to test its quality and usefulness. The results provide evidence that the PC kept at 27 degrees C showed marked differences compared with the PC stored at 22 degrees C. For example, the platelets showed the strongest morphological changes, less survival and probably a higher metabolic activity than those kept at 22 degrees C. Although the in vitro function was altered, it was recovered after 90 minutes of incubation with fresh frozen plasma. According with these results we can conclude that the in vivo hemostatic activity of these PC is as efficient as those stored at 4 and 22 degrees C, but due to the decreased of the in vivo platelet survival, we support that the 22 degrees C remain as the best temperature for the PC storage.

Biological activity of nitroguaiacol ether derivatives of streptomycin.

The correlation between the "in vivo" and "in vitro" inhibitory properties of six nitroguaiacol ether derivatives of streptomycin is studied. The differential activity of the derivatives is more closely related to differences in efficiency in the transport of the drugs than to their capacity to inhibit poly(U) directed polyphenylalanine synthesis. The results obtained are discussed in relation to the characteristics of the different derivatives.

[Typical dishes consumed in Sonora: regionalization and nutrient contribution].

This study deals with the development of a method to identify typical food dishes, successfully applied in the State of Sonora, Mexico. The State was divided into six regions considering the size of the territory and existence of different physical, economic and sociocultural conditions, and how they influence the consumption patterns. A list of 66 regional food dishes was prepared by interviewing housewives from different parts of Sonora who now lived in the city of Hermosillo. From this list, 47 dishes were identified as regionally consumed, and a State-wide survey was performed to determine their frequency of consumption. Based on the categories of "very frequently", "frequently" and "less frequently", a group of 15 dishes (2 per region, plus 3 dishes throughout the State) was selected. A questionnaire was used to obtain the different ways they had of preparing them. The "typical recipe" for each dish was also determined, using the mode as statistical parameter. Samples of these food dishes were prepared at the laboratory following the selected home recipes for proximal analysis and determination of their vitamin A, thiamine, riboflavin, niacin, calcium, phosphorus and iron content. The results showed that a portion of "gallina pinta" contributed more than 25% of the daily recommended intake established by the National Institute of Nutrition "Salvador Zubirán" (INNSZ) for an adult man for energy, protein, iron, niacin and 24.3% thiamine and 21.0% calcium. The "chivichangas de queso", for example, contributed 45.2% of the calcium requirement, and the "tamales de carne", 36% iron.(ABSTRACT TRUNCATED AT 250 WORDS)
