[Methodological specifications on the Todd method and preliminary results].

In order to measure the potentiality and kinetics tissue fibrinolysis we have modified Todd's histochemical method preparing at least six sections taken from same sample tissue, by infraoperatory biopsy, esponing them to a fibrin film at a constant concentration; we have also chosen to incubate the section at 10, 20, 30, 45, 60 min. The measurement of the areas of fibrin, of tissue and fibrinolysis, at the above mentioned times, has been effected at standard magnification (15 X) by an image analyser (Videoplan) scale 1:8. For each sample we suggest to elaborate an Index of the Potentiality of Fibrinolysis (formula; see text) and an Index of the Kinetics of the Fibrinolysis (formula; see text). Applying this method to two different groups of thyroid disease (struma and adenoma) we have not pointed out any statistically significant difference.

Platelet membrane fatty acids in thrombocytosis due to myeloproliferative disorders.

The fatty acid composition of platelet membranes has been analysed in patients with thrombocytosis due to myeloproliferative disorders, who had not taken any drugs. A significant increase in palmitic and oleic acid, together with a decrease in stearic, linoleic and arachidonic acids was observed. The fatty acid pattern of platelet membranes was also analysed in patients during treatment with ASA (acetylsalicylic acid). ASA ingestion completely normalizes the platelet content of palmitic acid and partially that of stearic and arachidonic acid, whereas it has no effect on the level of linoleic acid and raises that of oleic acid. The altered pattern of fatty acids observed in patients may interfere with platelet function by decreasing membrane fluidity. Treatment of patients with ASA seems to act on platelet membranes by partially normalizing the fatty acid composition.

Platelet glyoxalases in thrombocytosis.

In platelets of patients suffering from thrombocytosis due to myeloproliferative disorders, glyoxalase I activity is significantly higher than in controls (P less than 0.01), while glyoxalase II levels are the same (P less than 0.3). The cellular concentration of glutathione is also increased in patients. Km values for methylglyoxal (glyoxalase I) and S-lactoylglutathione (glyoxalase II) are identical both in normal and pathological subjects, as are the thermostability of the two enzymes. The higher activity observed for glyoxalase I in patients could be related to a specific role of this enzyme in platelets.

Platelet lysosomal enzymes are normal in myeloproliferative disorders.

In platelets of subjects affected with myeloproliferative disorders the following lysosomal enzymes were studied: alpha-mannosidase, alpha-fucosidase, beta-galactosidase, beta-glucosidase, beta-glucuronidase, beta-N-acetylglucosaminidase and acid phosphatase. For each enzyme the specific activity, the optimum of pH and buffer, Km and saturating substrate concentrations, as well as thermostability were determined. Control and patient enzymes showed no difference.

Hemostatic disorders in 25 patients with limited and uncomplicated thyroid and breast cancer: prophylactic and therapeutic considerations.

In order to evaluate the efficacy of an anticoagulant treatment in neoplasia, we have looked for the existence and the possible role of hemostatic unbalance in patients affected by limited and uncomplicated thyroid and breast cancer by examining hemostasis in 25 patients. Our data allowed us to evidentiate an accelerated and increased fibrinoformation associated with the presence of plasminogen's activation inhibitor which overlaps the increased plasminogen's activators. We evidentiated also an increase of platelet functions in vitro and of their activation in vivo. These findings support the hypothesis of a possible role played by hemostasis alterations in the pathogenesis of thromboembolic complications, cancer growth and/or metastatization, and justify prophylactic and therapeutic anticoagulation.

Studies on the inactivation of glyceraldehyde-3-phosphate dehydrogenase by methylglyoxal.

Glyceraldehyde-3-P dehydrogenase (E.C. 1.2.1.12) from rabbit muscle is rapidly inactivated by methylglyoxal following pseudo first-order kinetics. Substrate, as well as inorganic phosphate, affords protection, whereas NAD+ is ineffective. The arginine residue implicated in the reaction is probably the anion binding site of the phosphate group of the substrate.

[Relationship between the platelet aggregation of the newborn and mother (author's transl)].

The platelets function of newborn aroused great interest in these last years. On this line, AA. made the stretching test according to Breddin K. and Burck K. H., and. Breddin's Aggregation test on blood coming from 30 couples mother-newborn, drawn immediately after the birth. 22 newborns (73.3%) showed a stretching less than 65%, as well as 21 mothers (70%). The average stretching of mothers resulted 59.5% (+/- 2.26), whereas the average of newborns resulted 55.03% (+/- 2.71). Student's T, calculated on these values, did not result statistically meaningfull. Data-difference of Aggregation test according to Breddin and Burck, pertinent the couples mother-newborn gave on contrary satisfactory statistical information (p less than 0.0001). The explanatory characteristics of stretching test did not allow further issues, whereas Aggregation test results allow to come to the conclusion that newborn's platelets show a less capacity, as to mother's platelets, to aggregata under a mechanical standardized stimulation.
