[Hageman factor deficiency (factor XII)--hemorrhage or thrombosis?].

Four generations of a kin with congenital Factor XII deficiency were examined for coagulation and fibrinolysis, with the homozygous female carrier of features with a Factor XII below 1% also revealing certain indications of a disturbed fibrinolysis. The other members of the family had to be evaluated as heterozygous ones, showing values of Factor XII between 40 and 60%. The findings are discussed by referring to data from literature.

[Antithrombin V--a rare concomitant symptom in plasmacytoma and autoimmune diseases].

Antithrombin V as a pathological inhibitor of coagulation may occur in some rare cases as an accompanying symptom of various basic diseases. In the course of two years, antithrombin V could be identified in 8 patients, with plasmocytoma, lupus erythematodes visceralis, glomerulonephritis or colitis ulcerosa existing as basic diseases. Clinical findings in patients and those gained in analyzing coagulation, procedures of laboratory diagnostics for determining characteristic properties of antithrombin V as well as the therapeutic way of influencing the activity of antithrombin V are represented.

[Circadian behavior of the fibrinogen concentration in the plasma].

The concentration of the fibrinogen in the plasmas was determined in the course of a day (circadian rhythm) as well as in different position of the body. During the night hours a concentration minimum existed at 24.00 o'clock or at 4.00 o'clock, which however, could not be explained by the position of the body alone, which had changed in comparison to day-time. The time of the blood-taking and the position of the body must be taken into consideration in the interpretation of the laboratory values.

[Monitoring of heparin therapy with activated recalcification time].

In in-vivo- and in-vitro-examinations the activated recalcification time, the recalcification time of the plasma, the partial thromboplastin time and the thrombin time were compared concerning their suitability for the control of a heparin therapy. The laboratory methods tested considerably differed in their sensitivity compared with heparin and in the possibility of a quantitative statement. The use of a certain laboratory method depends on the indication for a heparin therapy and on the heparin concentration in the blood to be expected. While thrombin time, recalcification time of the plasma and the partial thromboplastin time are suited only for the proof of lower heparin concentrations, the activated recalcification time allows also the proof of higher heparin concentrations in the blood.

[Determination of coagulation factor VIII inhibitors].

Factor VIII inhibitors represent a severe complication of the haemophilia A. Different methodical approach in the diagnostics of the laboratory and various definitions of the inhibitor unit render difficult the comparability of the results of examination. Methods for the unitary performance of the qualitative proof of inhibitors and the quantitative estimation of inhibitors are described and their application in the examination of two patients with factor VIII inhibitors are shown.

[A suggestion for the uniform laboratory diagnosis of hemophilia].

The laboratory diagnosis of the haemophilia is performed in a different way in the individual laboratories. Differences in the selection and the performance of the methods used render differences in evidence, comparability and expenditure of time. The following proposition for a unitary laboratory diagnostics of haemophilia is made: 1. performance of a basic programme, 2. localisation of the coagulation defect with the help of the PTT using correction plasmas, 3. quantitative estimation of factors (possibly inhibitors) after one-step method, in which cases usual test sets of instruments with standardized directive for the performance are prerequisite for comparable results and a subdivision in degrees of severity.

[Prevention of thromboembolism in surgical gynecology].

It is reported on the prophylaxis of thromboembolic diseases with acetylsalicylic acid (Micristin) in gynaecological surgery. The number of thromboembolic complications clinically ascertained was diminished from 3,25% (without Micristin) to 0,82% (with Micristin). Side effects were rarely observed, laboratory control was not necessary, Low-dose-heparin-prophylaxis were executed on contraindications and incompatibilities of Micristin. The perioperative use of low doses of heparin combined with postoperative use of oral anticoagulants on patients with increased risk of thromboembolism are discussed.

[Phospholipid composition and fatty acid patterns of isolated phospholipids from rabbit reticulocyte mitochondria].

The phospholipid composition and fatty acid patterns of individual phospholipid classes were determined in mitochondria from rabbit reticulocytes. Compared to mitochondria from rat liver reticulocyte, mitochondria exhibit about twice the amount of phospholipids. The phospholipid pattern of reticulocyte mitochondria (phosphatidylcholine, phosphatidylethanolamine, phosphatidylinositol and cardiolipin) is comparable with other mitochondrial species. Mitochondrial fractions from reticulocytes are characterized, however, by an additional content of sphingomyelin. This sphingomyelin differs in its fatty acid composition from the sphingomyelin of the plasma membrane. The fatty acid patterns of all other phospholipids essentially correspond to those of mitochondria from other sources and to those of plasma membranes as well.

[Precision control of a single step determination of blood coagulation factor VIII in the plasma].

The findings of a precision control are reported for a one-stage determination of the factor VIII activity in the plasma for a period of 6 months. The storage property of the reagents was achieved by storing them in fluid nitrogen. Coefficients of variation under 55 were achieved at controlling the normal range as well as the pathological one. Thus the precision of this method may be compared with that of clinicochemical examinations.
