The process of activation of T lymphocytes by ionophoric substances was analysed according to current concepts of T-lymphocyte activation. Proliferation induced by the ionophore A23-187 was found to be strictly dependent on the exogenous availability of growth factors. Lack of production of growth factors in spleen cell cultures containing the ionophore A23-187 explained the failure to directly stimulate mouse T-lymphocyte proliferation. It was concluded that the main action of A23-187 is the induction of expression of growth receptors, and that the increase in Ca++ uptake is, in itself, not sufficient to induce mitotic events.
Autoantiidiotypic antibodies to anti-dextran antibodies did not influence the kinetics of the immune response, but profoundly affected the secondary response and caused a more or less complete inability to produce anti-dextran antibodies after a primary immune response.
The HSD17B4 gene codes for a 80 kDa multifunctional enzyme containing three distinct functional domains and is localized in peroxisomes. The N-terminal part exhibits 3-hydroxyacyl-CoA dehydrogenase and 17beta-hydroxysteroid dehydrogenase activity whereas the central part shows enoyl-CoA hydratase activity. The carboxy-terminal part of the protein has sterol-carrier-protein activity. The protein is widely expressed, however in several tissues like brain, uterus and lung its expression is limited to specific cells like Purkinje cells or luminal epithelium. The HSD17B4 gene consist of 24 exons and 23 introns with classical intron-exon junctions spanning more than 100 kbp. The importance of the HSD17B4 protein is stressed by the identification of patients with severe clinical abnormalities due to mutations in the HSD17B4 gene. We have now checked the consequences of one frequent mutation, G16 S, which results in inactivation of the enzyme due to loss of interaction with NAD+.
Previous studies have shown that genetic factors control the susceptibility to mercury-induced immunoglobulin (Ig)G1 antibody formation, IgE synthesis, renal IgG deposits and antinucleolar autoantibodies (ANolA) production in the susceptible mice. In this study, we examined the genetic control of resistance to these characteristics after HgCl2 injection in F1 hybrid crosses between the highly mercury resistant DBA/2 and mercury susceptible NZB (H-2d), SJL (H-2 s), A.CA (H-2f) and DBA/1 (H-2q) mice and also in backcross hybrids between (DBA/2 x SJL)F1 and SJL mice. We observed that mercury-induced immune/autoimmune manifestations were profoundly downregulated in most (if not all) of the F1 hybrids, indicating that the resistance to mercury was a dominant trait. Analysis of mercury-induced immune/autoimmune responses in the (DBA/2 x SJL) x SJL backcross hybrids suggested that only one gene or a cluster of genes determined the resistance to the ANolA production, whereas the resistance to other characteristics was controlled by two and/or three gene loci. By H-2 genotyping the backcross mice, it was found that H-2d haplotype per se could confer resistance to ANolA production. However, we did not find any significant association between the H-2d haplotype and the resistance to increase of IgG1 and IgE synthesis and the development of renal IgG1 deposits. Thus, while in DBA/2 mice, gene(s) in the H-2 loci strictly contribute to the inheritance of resistance to ANolA production; non-H-2 genes mainly govern the inheritance of unresponsiveness regarding other characteristics.
INTRODUCTION: Diagnosis of Guillian-Barré Syndrome usually is not difficult, but diagnostic failure occurs for the variable initial presentation. Diagnosis is based on physical examination showing loss of motor strength in more than one limb and loss of deep tendon reflexes. Ventilatory assistance, pharmacologic maintenance of cardiovascular homeostasis, corticosteroids, IgG and plasma exchange are the dominant therapeutic measures. CASE: This article reports on a case of a 59-year old surgeon suffering from degenerative disc disease in the lumbar spine. The patient developed a severe course of the Guillian-Barré Syndrome with persisting motor weakness of the legs. CONCLUSION: If the primary problem at presentation is limb and back pain the pathology appears to be in the musculoskeletal rather than in neurological system. The awareness of this presentation of Guillian-Barré-Syndrome will eliminate delay in diagnosis.
This study explored the performance of experienced laboratories in the analysis for total selenium in water by a variety of analytical methods. The goal of the study was to examine intra- and interlaboratory variability. Replicates (n = 7) of 7 sample types that included a reference material of known Se concentration, natural waters, and treated wastewaters were submitted to 7 laboratories with prequalified Se analytical experience. Results of the study indicated wide ranges in minimum and maximum results, distinct differences in laboratory precision, and routine reporting of numerical results below statistical limits of quantitation. Hydride generation as a sample introduction technique demonstrated superior performance. In general, the study supports a caution advisory about using low-level Se data, especially results lower than about 10 micrograms Se/L, without quantitating the statistical uncertainty of the data. Because this study used data from samples that were submitted in bulk to participating laboratories prequalified for Se analytical expertise and experience, it can be considered a best-case demonstration of performance.
A multiresidue screen for quantitative determination of 43 organophosphorus insecticides in 5 g of plant and animal tissues is described. The organophosphorus insecticides are extracted with methanol-dichloromethane (10 + 90, v/v) and cleaned up using automated gel permeation chromatography with hexane-ethyl acetate (60 + 40) eluant and in-line silica gel minicolumns. Concentrated extracts are analyzed by gas chromatography with flame photometric detection. The method recovers 43 organophosphorus insecticides in the range of 72 to 115%. Analysis of fortified bovine liver (n = 5) shows an average 95.9 +/- 4.8% recovery at the 0.05 micrograms/g level and 93 +/- 3.8% at the 0.5 micrograms/g level. Analysis of fortified bovine rumen content (n = 5) shows an average 98 +/- 4.2% recovery at the 0.1 micrograms/g level and 98.7 +/- 2.8% at the 1 micrograms/g level. Method detection limits ranged from 0.01 to 0.05 micrograms/g for the compounds studied using a nominal 5 gram sample.
Total selenium is determined by inductively coupled plasma (ICP) atomic emission using hydride vapor generation. A 1 g sample is wet ashed in a 16 x 150 mm 10 mL volumetric test tube on a programmed heating block with nitric, sulfuric, and perchloric acids at up to 310 degrees C. After treatment with hydrochloric acid, the selenium is reduced by sodium borohydride to hydrogen selenide is a simplified continuous flow manifold. A standard pneumatic nebulizer effects the gas-liquid separation of H2Se, which is quantified by ICP atomic emission at 196.090 nm. The instrument detection limit for the method has been determined to be 0.4 microgram/L. For a 10:1 dilution of a nominal 1 g sample, the detection limit is 4 micrograms/kg and the linear range is up to 4 mg/kg. The method has demonstrated statistical control for samples of biological and environmental interest and is especially well suited to analysis of small samples.
A rapid, ion-exchange liquid chromatographic method for the determination of nitrate and nitrite in biological fluids is presented. Samples are deproteinated by ultrafiltration followed by removal of chloride using a silver form cation-exchange resin. Nitrate and nitrite are measured by ion-exchange liquid chromatography with conductivity detection. Recoveries from serum, ocular fluid, and water were determined for fortifications from 10 to 150 mg/L. Average recoveries ranged from 96 to 104% for nitrate and from 89 to 105% for nitrite. Pooled RSD values ranged between 1.5 and 1.9% for these analytes in all matrixes examined. The method of joint confidence hexagons was applied to the data to determine constant and relative bias of the method for each of the 3 matrixes in the study.
Con A can activate cytotoxic effector cells, helper cells and suppressor cells. In all cases Con A activates polyclonally and thus the entire T cell repertoir can be revealed by Con A activation. Nylon purified T cells cannot be activated by Con A. however, in the presence of helper cells, such as adherent cells, or in the presence of serum from several species, Con A activates purified T cells. The serum factors that are responsible for Con A activation of purified T cells are present in the albumin fraction. It is suggested that Ia molecules may be responsible for the ability of Con A to activate purified T cells and a general hypothesis for T cell activation is outlined.

The evidence for the one non-specific signal hypothesis stating that the B lymphocytes are activated by non-clonally distributed receptors which are not the Ig receptors, have been summarized. Even though protein A is a polyclonal B cell activator, it does not exert its effect by interacting with the Fc part of Ig receptors. One consequence of the one non-specific signal concept is that thymus-dependent antigens cannot activate or tolerize B cells. It was shown that B cells from animals tolerant to a thymus-dependent protein antigen could be activated by polyclonal B cell activators to produce antibodies against the tolerogen. Experimentally induced tolerance did not differ from tolerance to self antigens, since LPS and PPD induced autoantibodies capable of lysing autologous red cells and isotope labelled autologous and syngeneic spleen cells. Thus, B cells cannot discriminate self from nonself whereas T cells have been shown to possess this ability.
