Evaluation of a new specific analysis of urinary delta-aminolevulinic acid in man.

A new specific method for urinary delta-aminolevulinic acid (ALA-U) measurement in man as a serial screening method for health control in occupational lead exposure was compared with a recommended, selective test (DAVIS method). The new test can be run fully automated, and the cost of material per sample is much lower than in the DAVIS method. ALA values obtained with this specific method were lower than those measured with the DAVIS method (= DAVIS values) in each case due to the elimination of interfering urinary compounds. In DAVIS values below the biological tolerance level (BAT-level) of 15 mg/l, the percentage of interfering compounds varied between 10 and 80%; above BAT level, these components represented about 20% of the total amount of ALA-U. A strong correlation can be demonstrated between specifically measured ALA values and the enzyme activity of ALA-dehydratase (ALA-D) in blood at lead levels (Pb-B) below 35 micrograms/dl (r = -0.768 for ALA-U/log ALA-D; n = 18).

Urinary excretion of mutagens in passive smokers.

Six healthy young volunteers with no history of active smoking were asked to keep on their Western diets avoiding the consumption of alcoholic beverages, excess coffee, any sort of medicament, and the known pro- and/or anti-mutagen-containing foods and drinks, 24 h before and during the experiments. They were exposed passively to cigarette smoke produced by 4 habitual smokers in an unventilated 48.6 m3 room for 8 h. The carbon monoxide concentration was 18.85 +/- 7.3 ppm during the 8-h exposure. Frameshift mutagens were isolated from 10-h urine samples using chloroform and were tested for mutagenicity in the Salmonella/mammalian microsome assay employing Salmonella typhimurium TA98. Although clearly enhanced, no significant mutagenic activity could be found with 25 ml equivalent urine/plate after passive exposure to cigarette smoke. The weak mutagenicities found were highly significant when 50 ml equivalent urine/plate was tested. No direct correlation was observed between urine mutagenicity and the urinary cotinine concentration. The results obtained are discussed with reference to inconsistent reports in the literature concerning the mutagenicity of urine after passive smoking.

Inhibitory action of transforming growth factor beta on endothelial cells.

In the present study, we show that transforming growth factor beta (TGF-beta) strongly inhibits fibroblast growth factor-induced proliferation and motility of bovine endothelial cells in tissue culture. TGF-beta also prevents the phorbol ester-induced invasion of capillary endothelial cells into collagen matrices--i.e., blocks angiogenesis in vitro. TGF-beta promotes the incorporation of fibronectin into the extracellular matrix of endothelial cells and stimulates the secretion of other proteins--mainly of 55- and 180-kDa components. We show furthermore that endothelial cells express TGF-beta receptors similar in size to those of other tissue culture cell lines: a 280-kDa complex is present in subconfluent cells, and 85- and 72-kDa protein bands are seen in confluent cells. The various effects of TGF-beta on endothelial cells suggest that these cells are an important target of TGF-beta during wound healing and angiogenesis.

[Oxalic acid resorption in patients with resection of the small intestine, jejunoileal bypass, Crohn disease and chronic pancreatitis].

The enteric absorption of oxalic acid with 14C-labelled oxalic acid was determined in patients with small bowel resection, jejunoileal bypass, Crohn's disease and chronic pancreatitis in comparison to the control group. Extreme hyperoxaluria were found in small bowel resections above 100 cm, after bypass operations and in ileocolitis Crohn with signs of clinical activity. Small bowel resections and relapses of Crohn's disease increase the absorption of oxalic acid. The significance of 14C-oxalic acid absorption test is the recognition of enteric hyperoxaluria.

[Pathogenesis of hyperoxalurias and calcium oxalate calculi in intestinal diseases].

Hyperoxaluria and calcium oxalate lithiasis have a multifactorial genesis in bowel diseases. The augmented synthesis of oxalic acid in the liver is of minor importance. A diminished bacterial degradation is to asses up to day only hardly. The increasing absorption is of highest importance by deficient production of calcium oxalate in the intestinal tract and by increasing permeability of colonic mucosa above all in steatorrhoea or in patients with augmented calcium absorption. The crystallization of calcium oxalate in the urine is promoted by shortage of vitamin A, citrate, zinc or magnesium.

[Treatment of mastopathies with cyclic mastodynia. Clinical results and hormonal profiles].

The therapeutic effect of a long-term Gestagen therapy (lynestrenol) and a phytotherapy (Mastodynon) in comparison to a placebo group is reported in 160 cases of severe mastopathy with cyclic mastalgia. In 82.1 percent of the patients in the gestagen-group and in 74.5 percent of the patients in the Mastodynon-group a good relief of the premenstrual symptoms was achieved. In the placebo group only 36.8 percent of the patients reported a relief of symptoms. The difference is significant. The evaluation and analysis of serum HPRL and serum-progesterone showed a significant rise of the HPRL-level and a significant decrease of the progesterone-level. Because of this severe influence on the endocrine balance the recommendation for an alternative phytotherapy prior to a long term gestagen therapy is justified.

[Resorption of oxalic acid in calcium oxalate nephrolithiasis].

In 43 healthy reference persons and 54 patients with relapsing calcium oxalate nephrolithiasis the absorption of oxalic acid was measured by means of 14C-oxalic acid. In patients with oxalate calculi the absorption of oxalic acid is totally increased and also in such ones with Whewellite-calculi. It is decreased in carriers of Weddelite calculi. The increase of the absorption of oxalic acid in patients with mixed calculi is not significant.
