The oncostatic and immunosuppressive action of new nitrosourea derivatives containing sugar radicals.

Four new nitrosourea derivatives represent an appreciable progress in the treatment and cure of L1210 leukemia. Their therapeutic index is higher than that of CCNU and MeCCNU. Of these compounds, RFCNU may prove the most promising, as its therapeutic index is the highest of those for all the four compounds studied; moreover, unlike the other products, it is not immunosuppressive, whether administered before or after the antigen.

Further purification of the lymphocyte inhibiting extract from the thymus.

A crude thymic extract, demonstrated to possess both immunosuppressive activity on T lymphocytes in vivo, and inhibitory action on DNA synthesis in T lymphocytes in vitro, has been purified by ultra-filtration and ion exchange chromatography procedures. The immunosuppressive activity was found to be associated with a high molecular weight fraction (greater than 50,000 daltons), devoid of immunosuppressive properties, a basic peptide has been isolated which retains the in vitro inhibitory activity on DNA synthesis.

Meningeal localisation of acute leukaemias.

A preventive chemoradiotherapeutic treatment of the central nervous system has to be started as soon as a complete remission is obtained in acute lymphoid leukemia. Any meningeal localisation is barely sensitive to treatment and reduces the long term chances of survival. The best treatment, according to our study, consists in the intrathecal administration of methotrexate and cytosine arabinoside followed by an irradiation of the central nervous system at a dose of 1,500 rads. The present trials have attempted to reduce the number of intrathecal injections, to increase the irradiation dose to 2,400 rads, and to restrict it to the skull.
