[Sonographically guided fine-needle biopsy of peripheral lung tumors].

Sonographically guided transthoracic fine-needle biopsy of peripheral pulmonary masses was performed in seven patients. None of the masses was accessible to fiber bronchoscopy. Three tumors were apical, two paravertebral and two lateral. The puncture specimen allowed a histological diagnosis in all five cases of malignant disease. One of the other two patients was found to have tuberculosis and the last, radiation fibrosis. Pneumothorax did not occur in any of these cases.

[Atrophy of the corpus mucosa of the stomach simulating polyposis].

We report on five cases of mucosal atrophy of the gastric body, under the aspect of a polyposis: These polyps showed intact mucosa with abundant fundic glands and were situated in the diffuse atrophic mucosa of the gastric body. Four of these cases were manifestations of type A gastritis, in one case the lesion surrounded a peptic ulcer. Laboratory findings of serologically analysed cases with type A gastritis showed elevated parietal cell antibodies in all cases, two of three cases showed a high level of gastrin, the values for vitamin B12 showed different levels; no patients revealed antibodies against the intrinsic factor, and no patient had pernicious anemia. The family history revealed three cases with cancer of the stomach on one side of the parents. It is important to biopsy polyps and the flat mucosa separately in order to verify this form of atrophy of the gastric mucosa and to also exclude small polypous tumors of the mucosa. Observation of the number of polyps allows a control of the extent of the atrophy of the mucosa.

[Severe thrombocytopenia, hemolysis and liver function disorder in late pregnancy. HELLP syndrome].

A 27-year-old gravida 2 was hospitalized in the 37th week of pregnancy because of nausea, vomiting and upper abdominal pain. She had severe thrombocytopenia (600/microliter), haemolysis and markedly abnormal liver functions (bilirubin 7.4 mg/dl, GOT 512, GPT 650 and LDH 1772 U/l), indicating a probably immunologically induced syndrome (HELLP) of late pregnancy. After platelet infusion and antithrombin III substitution a slightly growth-retarded girl was delivered without complications by section because of threatened intrauterine asphyxia while the cervix was undilated. The maternal platelet count and the liver functions quickly returned to normal post-partum.

Release of core DNA from nucleosomal core particles following (ADP-ribose)n-modification in vitro.

Isolated nucleosomal core particles from rat liver chromatin were ADP-ribosylated in vitro and the consequences of this modification on intranucleosomal DNA-protein interactions were studied by retention gel analysis. A separating force of 11 fN caused the complete release of 146 bp DNA fragments from ADP-ribosylated core particles, while the DNA of unmodified core particles remained protein-associated. We conclude that posttranslational (ADP-ribose)n-modification of chromatin proteins may reduce DNA-protein interactions at the nucleosomal level of chromatin organization.

Tuberculous gastric ulcer.

This is a case report on tuberculosis of the stomach in a 70-year-old woman who, submitted to an endoscopic examination for uncharacteristic upper abdominal complaints, revealed multiple ulcers in the aboral part of the stomach, together with a thickening of the gastric wall. Histologically, caseating epithelioid cell granulomas were found, and only a single acid-fast tubercle (TB rod). As a further specific manifestation, tuberculous spondylitis developed, which was treated surgically. Under anti-tuberculous treatment, the gastric ulcers healed. The clinical-endoscopic picture, the pathogenesis, complications and differential diagnosis are discussed on the basis of current literature.

Periodic changes of chromatin organization associated with rearrangement of repair patches accompany DNA excision repair of mammalian cells.

We have used 8-methoxypsoralen to probe the chromatin structure of mammalian cells in situ while they repair pyrimidine dimers or bulky lesions in DNA. We observed that excision repair of these DNA lesions is accompanied by periodic alterations of chromatin organization. In parallel, fluctuations of the rates of repair patch synthesis accompanied these structural changes. Taking advantage of the accessibility of free DNA domains for psoralen intercalation, we have developed a technique to quantitatively isolate the micrococcal nuclease-sensitive, free DNA fraction of native bulk chromatin. We have determined the location of newly synthesized repair patches relative to free DNA domains as a function of repair time. Extensive rearrangements of repair patches from these domains into micrococcal nuclease-resistant DNA were observed. Our results indicate that periodic changes of chromatin organization associated with rearrangement of repair patches accompany the process of excision repair in mammalian cells.

[Poisoning with andromedotoxin-containing honey].

Two patients poisoned by andromedotoxin-containing honey were treated in Austrian hospitals in 1981 and 1982. The toxin-containing honey had in both cases been obtained in the coastal mediterranean part of Turkey. Both patients had severe arterial hypotension, bradycardic arrhythmias, syncope and CNS symptoms. Symptomatic treatment was fully successful in both patients within 24 hours.

Electropolymerization as a versatile route for immobilizing biological species onto surfaces. Application to DNA biochips.

Biosensors based on electronic conducting polymers appear particularly well suited to the requirements of modern biological analysis--multi-parametric assays, high information density, and miniaturization. We describe a new methodology for the preparation of addressed DNA matrices. The process includes an electrochemically directed copolymerization of pyrrole and oligonucleotides bearing on their 5' end a pyrrole moiety. The resulting polymer film deposited on the addressed electrode consists of pyrrole chains bearing covalently linked oligonucleotides (ODN). An oligonucleotide array was constructed on a silicon device bearing a matrix of 48 addressable 50 x 50 microns gold microelectrodes. This technology was successfully applied to the genotyping of hepatitis C virus in blood samples. Fluorescence detection results show good sensitivity and a high degree of spatial resolution. In addition, gravimetric studies carried out by the quartz crystal microbalance technique provide quantitative data on the amount of surface-immobilized species. In the case of ODN, it allows discrimination between hybridization and nonspecific adsorption. The need for versatile processes for the immobilization of biological species on surfaces led us to extend our methodology. A biotinylated surface was obtained by coelectropolymerization of pyrrole and biotin-pyrrole monomers. The efficiency for recognition (and consequently immobilization) of R-phycoerythrin-avidin was demonstrated by fluorescence detection. Copolymerization of decreasing ratios of pyrrole-biotin over pyrrole allowed us to obtain a decreasing scale of fluorescence.

Homogeneous immunoassays using rare earth cryptates and time resolved fluorescence: principles and specific advantages for tumor markers.

We have developed a new type of long lived fluorescent tracer, the rare earth cryptates formed by the inclusion of a Eu3+ ion into the intra molecular cavity of a macrobicyclic ligand containing bipyridine units. This tracer is used in a new homogeneous assay format based on spectral and temporal selectivity as well as on an amplification of the cryptate fluorescence. As a consequence of these features, the measurement of the specific signal is totally shielded from media interaction and corrected in real time for sample to sample optical variation. We show the example of a PSA assay where an analytical sensitivity of 0.03 ng/ml is obtained and an AFP assay where measurements in the first minutes of the incubation allow a rapid estimation of the AFP concentration and therefore an immediate dilution of the samples if required. These results illustrate the performance of this new homogeneous method and point out the specific advantages it allows in terms of sensitivity, precision and flexibility. It is therefore particularly well adapted for the assay of analytes like tumor markers where both sensitivity and wide dynamic range are needed.
